Discovery and characterization of YSA64, a RBM39 degrader with in vivo efficacy and potent cellular activity in pediatric Ewing sarcoma A673.

Depletion of the splicing factor RBM39 disrupts spliceosome function and induces widespread RNA splicing defects, leading to antiproliferative effects in susceptible cancer cells. Here, we report the discovery and characterization of a new series of biphenyl-containing RBM39 degraders. The lead compound 42 promotes RBM39 degradation through formation of a ternary complex with RBM39 and DCAF15/DDB1 in a Cullin-RING E3 ligase- and proteasome-dependent manner, consistent with a molecular glue mechanism. Transcriptomic analyses in HCT-116 and K562 cells revealed extensive alternative splicing alte

Limb Salvage Using Knee Arthrodesis Endoprosthesis for Ewing Sarcoma With Extensor Mechanism Loss: A Case Report and Surgical Decision-Making Review.

INTRODUCTION: Malignant tumors around the knee pose a complex reconstructive challenge, particularly in the setting of an unreconstructable extensor mechanism. Although limb-salvage surgery with megaprosthetic reconstruction is the standard approach, its success is contingent upon extensor mechanism preservation. When this is not achievable, knee arthrodesis with modular endoprosthetic systems remains a robust salvage strategy. CASE PRESENTATION: A 17-year-old male presented with a one-year history of progressive left knee swelling, pain, and functional impairment. Imaging demonstrated extens

Safety and Tolerability of Interval Compressed Chemotherapy Schedule in Children Receiving Treatment for Ewing Sarcoma: A Real-world Experience From India.

BACKGROUND AND AIMS: Chemotherapy with alternating cycles of vincristine-doxorubicin-cyclophosphamide and ifosfamide-etoposide, along with primary tumor treatment with surgery or radiotherapy or both, constitute the usual treatment of Ewing sarcoma. The AEWS0031 study demonstrated survival benefits after interval-compressed chemotherapy without significant toxicity. The aim of this study was to assess the tolerability of dose-intensified chemotherapy in developing countries like India. METHODS: This was a retrospective analysis of children younger than 18 years of age with newly diagnosed Ewi

Giant calvarial metastasis from secondary Ewing sarcoma with chronic subdural hygroma in a childhood hematological cancer survivor - An unusual case report.

We report an unusual case of a 9-year-old boy with Hodgkin lymphoma post chemoradiotherapy, who later developed Ewing sarcoma involving the eighth rib, which had metastasised to the cranial vault with meningeal and subdural involvement. Presenting with seizures, headache and hemiparesis, he underwent craniotomy and gross total tumour resection, with histopathology confirming metastatic Ewing sarcoma. Despite initial improvement, the patient presented with further cranial metastasis on follow-up. This case contributes to the limited literature on Ewing sarcoma metastasis to the CNS in patients

Peripheral Nerve Stimulation for Arm Pain in an Adolescent Girl with Ewing Sarcoma as Bridge Therapy to Chemotherapy: A Case Report.

BACKGROUND: Cancer commonly presents discomfort from abnormal cell growth in healthy tissue and is often inadequately managed. Peripheral nerve stimulation (PNS), mainly for non-cancer chronic pain, has emerged as a minimally invasive option for neuropathic cancer-related pain when conventional methods fail. Limited research, primarily in adults, has focused on PNS in the non-oncological population. CASE REPORT: A 14-year-old adolescent girl with a history of obesity, diabetes, and Ewing sarcoma in her left proximal humerus experienced severe pain, initially rated at 10/10. After undergoing u

Ewing sarcoma of the temporal bone with aneurysmal bone cyst-like changes: A rare case report with an unusual radiological presentation.

Ewing sarcoma (ES) is a malignant small round cell tumor, accounting for 10-15% of all primary bone tumors and approximately 3% of all pediatric cancers. Primary ES of the cranial bone is unusual with reported incidence from 1% to 6% of all ES cases. This report shows a rare case of primary ES of the squamous temporal bone in a 12-year-old boy with a history of swelling of the right temporal region and symptoms of increased intracranial pressure. We illustrate the extremely unusual radiological presentation of this primary ES of temporal bone associated with large aneurysmal bone cyst-like (AB

No difference in survival after HLA mismatched versus HLA matched allogeneic stem cell transplantation in Ewing sarcoma patients with advanced disease.

Patients with advanced Ewing sarcoma (AES) carry a poor prognosis. Retrospectively, we analyzed 66 AES patients treated with allogeneic stem cell transplantation (allo-SCT) receiving HLA-mismatched (group A, n = 39) versus HLA-matched grafts (group B, n = 27). Median age at diagnosis was 13 years, and 15 years (range 3-49 years) at allo-SCT. The two groups did not differ statistically in distribution of gender, age, remission status/number of relapses at allo-SCT, or risk stratum. 9/39 (23%) group A versus 2/27 (7%) group B patients developed severe acute graft versus host disease (GvHD). Abemaciclib Is Active in Preclinical Models of Ewing Sarcoma via Multipronged Regulation of Cell Cycle, DNA Methylation, and Interferon Pathway Signaling.

PURPOSE: Ewing sarcoma (ES) is a rare and highly malignant cancer that occurs in the bone and surrounding tissue of children and adolescents. The EWS/ETS fusion transcription factor that drives ES pathobiology was previously demonstrated to modulate cyclin D1 expression. In this study, we evaluated abemaciclib, a small-molecule CDK4 and CDK6 (CDK4 and 6) inhibitor currently under clinical investigation in pediatric solid tumors, in preclinical models of ES. EXPERIMENTAL DESIGN: Using Western blot, high-content imaging, flow cytometry, ELISA, RNA sequencing, and CpG methylation assays, we char

Acquisition of secondary structural chromosomal changes in pediatric ewing sarcoma is a probable prognostic factor for tumor response and clinical outcome.

BACKGROUND: The Ewing sarcoma (ES) group of tumors commonly have the t(11;22)(q24;q12) or other rearrangements involving 22q12. In addition to these consistent aberrations, both numeric and structural aberrations have been reported: namely gains of chromosomes 8 and 12, the unbalanced translocation t(1;16), and deletions at the short arm of chromosome 1. METHODS: To evaluate the frequency and to study the prognostic implications of some of these aberrations in children, the authors performed a pilot study of 26 ES pediatric patients by classic cytogenetics and/or interphase fluorescence in si

Role of fluoroethyl tyrosine positron emission tomography-computed tomography scan in differentiating ewing's sarcoma from osteomyelitis.

Ewing's sarcoma is a kind of undifferentiated reticulocytic sarcoma, which was first reported in 1921 by James Ewing. It is difficult to differentiate Ewing's sarcoma from osteomyelitis on computed tomography (CT) and X-ray and hence cytological confirmation is needed. Fluorodeoxy glucose being a nonspecific tracer cannot differentiate between malignant and inflammatory lesions. However, it is found that Ewing's sarcoma has increased LAT1 transporter expression at the cell surface. This property has been utilized to specifically target the tumor cells and differentiate them from inflammatory l

Primary Ewing's sarcoma of the sinonasal tract, eroding the ethmoid and sphenoid sinus with intracranial extension: A rare case report.

Ewing's sarcoma (ES) is an aggressive tumour that may present with skeletal and extraskeletal forms. The extraskeletal form is rarely encountered in the head and neck region and is extremely rare in the sinonasal tract. This is the case report of a ES of the ethmoid sinus with intracranial and orbital extension in a 33-year-old male patient who presented with anosmia, epistaxis, reduction of visual acuity in the left eye and headache. On otorhinolaryngological clinical examination and biopsy via flexible endoscope, the lesion was misdiagnosed as ethmoid sinus carcinoma. The subsequent magnetic
