Tryptophan hydroxylase 2 deficiency alters autism-related behavioural phenotypes in rats.

Serotonin is a crucial neurotransmitter implicated in various physiological processes. Dysregulation of the serotonergic system, particularly in early life, may be associated with neuropsychiatric disorders, including autism spectrum disorder. This study utilises a rat model lacking the central serotonin-synthesising enzyme tryptophan hydroxylase 2 (TPH2) to investigate the effects of brain serotonin depletion on core autism-related behaviours. The animals' social behaviour was assessed using social interaction and resident-intruder tests, along with ultrasonic vocalisation analysis to evaluat

Abnormal behaviours relevant to neurodevelopmental disorders in Kirrel3-knockout mice.

In the nervous system, Kirrel3 is involved in neuronal migration, axonal fasciculation, and synapse formation. Recently, genetic links have been reported between mutations in the KIRREL3 gene and increased risk of neurodevelopmental disorders, including autism spectrum disorder (ASD) and intellectual disability. To elucidate the causal relationship between KIRREL3 deficiency and behavioural abnormalities relevant to neurodevelopmental disorders, we generated global Kirrel3-knockout (Kirrel3-/-) mice and investigated the detailed behavioural phenotypes. In the three-chambered social approach te

Issues in the use of visual supports to promote communication in individuals with autism spectrum disorder.

PURPOSE: Visual supports are widely used and generally regarded as an effective resource for intervention with individuals who function on the autism spectrum. More cross-contextual research into their efficacy is required. METHOD AND OUTCOMES: In this article, we selectively review the research literature around visual supports based on an original conceptual model that highlights their contribution in the interpersonal social and communicative milieu of classrooms, homes and other daily living contexts. Attention is drawn to a range of practical and research issues and challenges in the use

Exploring the Efficacy of the Occupational Therapy-Fundamental Motor Skills Training (OT-FMST) Protocol on Functional Performance in Children With Autism Spectrum Disorder.

Background Autism spectrum disorder (ASD) is primarily characterized by deficits in social communication and restricted behaviors, but motor impairments are also highly prevalent and debilitating. Children with ASD often experience challenges in fundamental motor skills (FMS), particularly in coordination, postural control, and locomotor abilities. These impairments not only limit physical activity but also restrict social interaction, play, and communication, thereby compounding developmental challenges. Although sensory integration and FMS-based interventions have each shown positive outcome

Approaching Social Determinants of Health in Otolaryngology - Head Neck Surgery: Linking Needs to Resources to Promote Health Equity.

Social determinants of health influence patient outcomes across all otolaryngology specialties, necessitating collaborative efforts among healthcare teams to promote health equity. Health disparities permeate healthcare, creating inequities related to race, ethnicity, socioeconomic status, environment, access to healthcare, education, gender, and sexual orientation. These disparities impact the diagnosis, management, and outcomes of conditions such as chronic otitis media, obstructive sleep apnea, sinusitis and allergies, hearing and balance disorders, head and neck cancer, and laryngeal or ai

Behavioral manifestations and neural mechanisms of empathic pain.

Empathy is an important trait that allows individuals to comprehend and share the emotions and sentiments of others. It not only facilitates effective interpersonal communication, but also helps in establishing meaningful connections and fostering trust and understanding. Impaired empathy development can manifest as excessive self-centeredness, extreme egoism, and antisocial behaviors. Many psychiatric disorders, such as autism, narcissistic personality disorder, and schizophrenia, are often accompanied by empathy disorders. Pain empathy, which is a common behavioral paradigm of empathic behav

Neurexin dysfunction in adult neurons results in autistic-like behavior in mice.

Autism spectrum disorders (ASDs) comprise a group of clinical phenotypes characterized by repetitive behavior and social and communication deficits. Autism is generally viewed as a neurodevelopmental disorder where insults during embryonic or early postnatal periods result in aberrant wiring and function of neuronal circuits. Neurexins are synaptic proteins associated with autism. Here, we generated transgenic βNrx1ΔC mice in which neurexin function is selectively impaired during late postnatal stages. Whole-cell recordings in cortical neurons show an impairment of glutamatergic synaptic trans

Challenged and changed: Quiet ego and posttraumatic growth in mothers raising children with autism spectrum disorder.

Posttraumatic growth theory posits that when life circumstances are perceived as stressful, secondary appraisal processes can be recruited in ways to facilitate both coping efforts and personal growth. Using a mixed-methods approach, we found mothers' most challenging experiences involved child behavior (e.g. aggression, communication, and social issues) and psychosocial impacts (e.g. lack of social support, perceived judgment of others, perceived loss, and personal distress). Descriptions of most rewarding experiences reflect posttraumatic growth frameworks including constructive perceptions

Hmong mental health needs assessment: a community-based partnership in a small mid-Western community.

The overall goal of this assessment was to verify the mental health needs of Hmong living in a mid-west community in order clarify the format, content, and feasibility of providing mental health services for Hmong in the future. Using a Community-Based Participatory Research (CBPR) model, we held four focus groups with 36 men, women, adolescents and professionals, all of Hmong descent, as well as interviewed 28 individual medical, mental health, education, and social service providers in the Eau Claire community. Our Hmong sample was frequently unclear about what "mental" health meant, indicat

Cannabidiol for the treatment of autism spectrum disorder: hope or hype?

RATIONALE: Autism spectrum disorder (ASD) is defined as a group of neurodevelopmental disorders whose symptoms include impaired communication and social interaction, restricted and repetitive patterns of behavior, and varying levels of intellectual disability. ASD is observed in early childhood and is one of the most severe chronic childhood disorders in prevalence, morbidity, and impact on society. It is usually accompanied by attention deficit hyperactivity disorder, anxiety, depression, sleep disorders, and epilepsy. The treatment of ASD has low efficacy, possibly because it has a heterogen

Engrailed 2 deficiency and chronic stress alter avoidance and motivation behaviors.

Autism spectrum disorder (ASD) is a pervasive neurodevelopmental disorder characterized by impairments in social interaction, cognition, and communication, as well as the presence of repetitive or stereotyped behaviors and interests. ASD is most often studied as a neurodevelopmental disease, but it is a lifelong disorder. Adults with ASD experience more stressful life events and greater perceived stress, and frequently have comorbid mood disorders such as anxiety and depression. It remains unclear whether adult exposure to chronic stress can exacerbate the behavioral and neurodevelopmental phe

The effects of context processing on social cognition impairments in adults with Asperger's syndrome.

Social cognition-the basis of all communicative and otherwise interpersonal relationships-is embedded in specific contextual circumstances which shape intrinsic meanings. This domain is compromised in the autism spectrum disorders (ASDs), including Asperger's syndrome (AS) (DSM-V). However, the few available reports of social cognition skills in adults with AS have largely neglected the effects of contextual factors. Moreover, previous studies on this population have also failed to simultaneously (a) assess multiple social cognition domains, (b) examine executive functions, (c) follow strict s

Redefining Peer Tutoring: Academic and Social Gains Through Twice-Exceptional Student Leadership.

This study explored the dual effects-both academic and social-of a peer tutoring framework where a twice-exceptional student with autism spectrum disorder (2e-ASD) served as a tutor for three students with learning disabilities (LDs). The goal was to assess whether peer-mediated instruction could improve tutees' reading accuracy while fostering tutors' social development. Using a multiple baseline design across participants, with a numerous probe structure, ensured rigorous methodology and intervention fidelity. Reading accuracy was systematically monitored across phases, and social validity f

Motion Processing in ASD: From Low-Level Information to Higher-Level Social Information.

From birth, our visual system is sensitive to movement. Motion, as defined by any change in spatial position over time, is part of our daily lives and can refer to various visual information from elements of nature (like a tree swaying in the wind), objects (like a moving car), animals (like a running dog) or people (like two people dancing). Atypical motion processing, in particular for social and biological movement cues, could lead to difficulties in social interaction and communication, like those observed in Autism Spectrum Disorder (ASD). Extensive research has focused on coherent and bi

Experts' perspectives on the impact of visual impairment and comorbid mental disorders on functioning in essential life domains.

BACKGROUND: Visual impairment (VI) with comorbid mental disorders (MDs) are expected to have a major impact on people's daily functioning, for which tailored support is needed. However, this has been barely investigated. Therefore, this study aimed to (1) determine the impact of VI and comorbid MDs on functioning in essential life domains, (2) gain insight into best-practices that are currently used to support this target group, and (3) determine strategies to optimize care in the future. METHODS: A four-step qualitative Delphi method was used to obtain input from 31 Dutch professionals who w

The search for universal transdiagnostic and trans-therapy change processes: Evidence for intolerance of uncertainty.

The search for universal processes associated with symptom change across emotional disorders and different forms of psychotherapy offers hope of increased theoretical parsimony and treatment efficiencies. This study investigated whether intolerance of uncertainty (IU) is a universal process by examining whether changes in IU were associated with changes in symptoms across three different cognitive behavior therapy protocols for depression (n=106), social anxiety disorder (n=88), or generalized anxiety disorder (n=62) in a community mental health clinic. IU was associated with reductions in rep

Motivational and cognitive correlates of community integration in homeless veterans entering a permanent supported housing program.

Homelessness is a major public health problem, and serious mental illness (SMI) is highly prevalent in the homeless population. Although supported housing services-which provide permanent housing in the community along with case management-improve housing outcomes, community integration typically remains poor, and little is known about the underlying determinants of poor community integration postresidential placement. The general SMI literature has indicated that motivational and cognitive ability factors are key determinants of successful community integration, which provides a foundation fo

Social Interaction Skill Intervention for Autistic Adults with Intellectual Disability and Limited Language: A Pilot of the SKILL Program.

There is a dearth of research that focuses on social intervention efforts for adults on the autism spectrum with intellectual disability and limited conversational language. Using a multiple baseline experimental design, this pilot investigation of the Socialization Knowledge for Individuals with Limited Language (SKILL) program evaluated a novel peer-facilitated group program specifically designed to target social interaction skills for this population. Findings from five pilot participants yielded evidence of social improvements across specific verbal skills (on-topic conversational contribu
