Expediting chemoenzymatic access to bacterial 'sialic acid-like' sugars via mapping of wild type sialic acid aldolase enzyme substrate tolerance and promiscuity.

Bacterial 'sialic-acid-like' sugars, including legionaminic acid and pseudaminic acid, are critical for the virulence and immune evasion of numerous human pathogens, including pan-drug resistant Acinetobacter baumannii and global gut pathogens Campylobacter jejuni and Helicobacter pylori. Despite this importance, a detailed understanding of the role these sugars play in pathogenesis is hampered by the challenging synthesis of their 9-deoxy nonulosonic acid backbones. Herein, we expedite access to these complex glyco-architectures through exploration of the biocatalytic potential of sialic acid…

The structural language of fungal cell wall sugars in WHO priority fungi: the determinant of pathogenicity and the gateway to new therapies.

Carbohydrates, far beyond their nutritional role, are fundamental biomolecules that encode biological information and mediate critical cellular interactions. In pathogenic fungi, cell wall (CW) glycans serve as both a protective armor and a dynamic interface with the host, directly influencing virulence and immune evasion. This review discusses the intricate architecture and composition of the CW in the four WHO-critical priority fungal pathogens: Aspergillus fumigatus, Candidozyma auris, Cryptococcus neoformans, and Candida albicans. We detail how distinct carbohydrate polymers, including α-

Ethnicity modifies the relationship between added sugars and fructose exposure in the first 1000 days and offspring body composition at 24 months.

Added sugars (AS) and fructose intake are linked to obesity. Hispanic populations experience high AS intake and obesity rates. It is underexplored if early sugars exposure influences offspring growth, especially across ethnic groups. This secondary analysis examined if AS and fructose intake during pregnancy, infancy, and toddlerhood influenced offspring adiposity at 24 months, and if ethnicity modified outcomes. We hypothesized that higher sugars exposure would predict increased adiposity. Mother-child pairs from a prenatal docosahexaenoic acid supplementation RCT and the offspring follow-up …

Effect of Important Food Sources of Fructose-Containing Sugars on Inflammatory Biomarkers: A Systematic Review and Meta-Analysis of Controlled Feeding Trials.

BACKGROUND: Fructose-containing sugars as sugar-sweetened beverages (SSBs) may increase inflammatory biomarkers. Whether this effect is mediated by the food matrix at different levels of energy is unknown. To investigate the role of food source and energy, we conducted a systematic review and meta-analysis of controlled trials on the effect of different food sources of fructose-containing sugars on inflammatory markers at different levels of energy control. METHODS: MEDLINE, Embase, and the Cochrane Library were searched through March 2022 for controlled feeding trials ≥ 7 days. Four trial de…

Catabolism of 1,5-anhydro-D-fructose in Sinorhizobium morelense S-30.7.5: discovery, characterization, and overexpression of a new 1,5-anhydro-D-fructose reductase and its application in sugar analysis and rare sugar synthesis.

The bacterium Sinorhizobium morelense S-30.7.5 was isolated by a microbial screening using the sugar 1,5-anhydro-D-fructose (AF) as the sole carbon source. This strain metabolized AF by a novel pathway involving its reduction to 1,5-anhydro-D-mannitol (AM) and the further conversion of AM to D-mannose by C-1 oxygenation. Growth studies showed that the AF metabolizing capability is not confined to S. morelense S-30.7.5 but is a more common feature among the Rhizobiaceae. The AF reducing enzyme was purified and characterized as a new NADPH-dependent monomeric reductase (AFR, EC 1.1.1.-) of 35.1

In-syringe solid-phase extraction coupled with UPLC-MS/MS for simultaneous determination of reducing sugars, animo acids and Amadori compounds in tobacco.

Considering the critical role of Maillard reaction-related compounds in determining the sensory and chemical stability of tobacco, this study develops an in-syringe SPE coupled with UPLC-MS/MS method for simultaneous determination of 43 Maillard reaction-related compounds in tobacco, covering reducing sugars (3), amino acids (17), Amadori compounds (17), and structurally related intermediates (6). The sample pretreatment utilizes HLB-packed self-assembled columns for SPE, followed by methanol elution. The extracted solution is then analyzed using a Titank C18 column (150×2.1 mm, 1.8 μm), with …

Mechanisms of caries induced by sugars: a narratives review from microbial metabolism to oral ecological imbalance and public health strategies for caries prevention.

Dental caries is defined as a chronic, multifactorial disease characterized by the demineralization of dental hard tissues resulting from the acid production by oral microbial communities metabolizing dietary sugars. The ingestion of sugars is a pivotal ecological factor in the progression of caries, with mechanisms that extend beyond merely providing substrates for cariogenic bacteria. This review explores the influence of sugars on the metabolism, adhesion, biofilm formation, and interspecies interactions of oral microorganisms, with a particular focus on species such as Streptococcus mutans…
