Staphylococcus aureus escapes endosomes and autophagosomes via α-hemolysin.

The interplay between endosomal and autophagic pathways is crucial for intracellular transport and modulation of pathogen replication. However, the mechanisms underlying the interaction between endosomes and autophagosomes (APs) in the context of Staphylococcus aureus (S. aureus) infection are not well understood. Specifically, the fusion of multivesicular bodies (MVBs) or recycled endosomes (REs) with APs remains unexplored. Here, we reveal that S. aureus induces crosstalk between endosomes and APs, a process modulated by Ca2+ influx. Depletion of Rab11A from REs impairs APs formation and tra

[Treatment prospects of lysosomal storage disorders].

Lysosomal storage disorders are caused either by deficiencies or decreased activity of enzymes localised in lysosomal vesicles or transport failure of these enzymes or their substrates. Accumulation of macromolecules destroy cell function presenting in clinical symptoms. Up to date, there are about 40 different lysosomal storage disorders according to the accumulated macromolecules. Till the last decades supportive therapy was the only option by these disorders. Enhanced researches in the last decades have presented some breakthrough results in the field of storage disease therapy. The review

Endosomes as central hubs of interorganellar communication and cellular homeostasis.

Endocytosis mediates the internalization of extracellular cargo via vesicular trafficking, enabling targeted delivery to specific organelles and maintaining cellular homeostasis. Endosomes function as dynamic hubs that orchestrate intracellular communication. While they primarily internalize material from the cell surface, their role extends far beyond passive transport. Through continuous cycles of sorting, fusion, and fission, endosomes engage in extensive crosstalk with other organelles via three fundamental modes: vesicle-mediated cargo transport, membrane contact site-mediated non-vesicul

Early glucagon-positive cells restrict pancreatic branching through VMAT2-noradrenaline-mediated ROS signaling.

Vesicular monoamine transporter 2 (VMAT2) transports monoamines into storage vesicles. We have previously reported that VMAT2 negatively regulates pancreatic progenitor differentiation into endocrine beta cells; however, the underlying mechanism remains unknown. Using a pancreatic bud explant culture, we show here that inhibiting VMAT2-mediated catecholamine uptake promotes pancreatic epithelial branching and increases the number of insulin-expressing cells. The increase in pancreatic branching was mimicked by inhibition of tyrosine hydroxylase, monoamine oxidase A, dopamine beta-hydroxylase o

How Important Is the Use of Cocaine and Amphetamines in the Development of Parkinson Disease? A Computational Study.

We studied dopamine levels in three compartments of the dopaminergic synapse, including the presynaptic neuron cytosol, dopamine storage vesicles, and the synaptic gap. By considering three transport pathways (dopamine transporter (DAT), vesicular transporter (VT), and exocytosis), four simulated scenarios were investigated: homeostasis, application of cocaine, methamphetamine, and reserpine. Recent experiments show that upon cocaine administration, the Drosophila melanogaster DAT permeation rate constant is decreased by 55% and we adopted this value for the human DAT. Two membrane proteins from Bifidobacterium breve UCC2003 constitute an ABC-type multidrug transporter.

Intrinsic resistance to drugs is one of the main determining factors in bacterial survival in the intestinal ecosystem. This is mediated by, among others, multidrug resistance (MDR) transporters, membrane proteins which extrude noxious compounds with very different chemical structures and cellular targets. Two genes from Bifidobacterium breve encoding hypothetical membrane proteins with a high homology with members of the ATP-binding cassette (ABC) family of multidrug efflux transporters, were expressed separately and jointly in Lactococcus lactis. Cells co-expressing both proteins exhibited e

Actomyosin Fluctuations Couple Temperature to Intracellular Transport and ELPV Phase Separation.

How living cells transduce thermal perturbations into altered intracellular organization remains a fundamental biophysical question. Here, using single-particle tracking of quantum dot (QD) diffusion and endocytic vesicle transport, we show that temperature-enhanced transport is not a simple passive solvent effect; instead, heating enhances active cytoplasmic dynamics mediated by myosin-II-dependent actomyosin activity. While active fluctuations enhance random QD diffusion, they also accelerate active vesicular transport by reducing its mechanical resistance. Using thermoresponsive elastin-lik

Extracellular vesicles: Critical bilateral communicators in periphery-brain crosstalk in central nervous system disorders.

Growing evidence shows that there is a comorbid mechanism between the central nervous system (CNS) and the peripheral organs. The bilateral transmission of signal molecules in periphery-brain crosstalk plays an important role in the underlying mechanism, which result from complex networks of neurohumoral circuits. Secreted by almost all cells and considered innovative information transport systems, extracellular vesicles (EVs) encapsulate and deliver nucleic acids, proteins, lipids, and various other bioactive regulators. Moreover, EVs can cross the blood-brain barrier (BBB), they are also ide

Disease-associated mutations in human BICD2 hyperactivate motility of dynein-dynactin.

Bicaudal D2 (BICD2) joins dynein with dynactin into a ternary complex (termed DDB) capable of processive movement. Point mutations in the BICD2 gene have been identified in patients with a dominant form of spinal muscular atrophy, but how these mutations cause disease is unknown. To investigate this question, we have developed in vitro motility assays with purified DDB and BICD2's membrane vesicle partner, the GTPase Rab6a. Rab6a-GTP, either in solution or bound to artificial liposomes, released BICD2 from an autoinhibited state and promoted robust dynein-dynactin transport. In these assays, B

[Essential trace metals and brain function].

Trace metals such as zinc, manganese, and iron are necessary for the growth and function of the brain. The transport of trace metals into the brain is strictly regulated by the brain barrier system, i.e., the blood-brain and blood-cerebrospinal fluid barriers. Trace metals usually serve the function of metalloproteins in neurons and glial cells, while a portion of trace metals exists in the presynaptic vesicles and may be released with neurotransmitters into the synaptic cleft. Zinc and manganese influence the concentration of neurotransmitters in the synaptic cleft, probably via the action ag

Multiple human isoforms of drug transporters contribute to the hepatic and renal transport of olmesartan, a selective antagonist of the angiotensin II AT1-receptor.

Olmesartan, a novel angiotensin II AT1-receptor antagonist, is excreted into both bile and urine, with minimal metabolism. Because olmesartan is a hydrophilic anionic compound, some transporters could be involved in its hepatic and renal clearance. In this study, we characterized the role of human drug transporters in the pharmacokinetics of olmesartan and determined the contribution of each transporter to the overall clearance of olmesartan. Olmesartan was significantly taken up into human embryonic kidney 293 cells expressing organic anion-transporting polypeptide (OATP) 1B1, OATP1B3, organi

P-glycoprotein (P-gp)-driven cancer drug resistance: biological profile, non-coding RNAs, drugs and nanomodulators.

Drug resistance has compromised the efficacy of chemotherapy. The dysregulation of drug transporters including P-glycoprotein (P-gp) can mediate drug resistance through drug efflux. In this review, we highlight the role of P-gp in cancer drug resistance and the related molecular pathways, including phosphoinositide 3-kinase (PI3K)-Akt, phosphatase and tensin homolog (PTEN) and nuclear factor-κB (NF-κB), along with non-coding RNAs (ncRNAs). Extracellular vesicles secreted by the cells can transport ncRNAs and other proteins to change P-gp activity in cancer drug resistance. P-gp requires ATP to

Sec14 family of lipid transfer proteins in yeasts.

The hydrophobicity of lipids prevents their free movement across the cytoplasm. To achieve highly heterogeneous and precisely regulated lipid distribution in different cellular membranes, lipids are transported by lipid transfer proteins (LTPs) in addition to their transport by vesicles. Sec14 family is one of the most extensively studied groups of LTPs. Here we provide an overview of Sec14 family of LTPs in the most studied yeast Saccharomyces cerevisiae as well as in other selected non-Saccharomyces yeasts-Schizosaccharomyces pombe, Kluyveromyces lactis, Candida albicans, Candida glabrata, C

eNOS knockout mouse as a model of fetal growth restriction with an impaired uterine artery function and placental transport phenotype.

Fetal growth restriction (FGR) is the inability of a fetus to reach its genetically predetermined growth potential. In the absence of a genetic anomaly or maternal undernutrition, FGR is attributable to "placental insufficiency": inappropriate maternal/fetal blood flow, reduced nutrient transport or morphological abnormalities of the placenta (e.g., altered barrier thickness). It is not known whether these diverse factors act singly, or in combination, having additive effects that may lead to greater FGR severity. We suggest that multiplicity of such dysfunction might underlie the diverse FGR

Mechanisms involved in the blood-brain barrier increased permeability induced by Phoneutria nigriventer spider venom in rats.

We have recently demonstrated by electron microscopy, using lanthanum nitrate as an extracellular tracer, that the intravenous injection of Phoneutria nigriventer spider venom (PNV) induces blood-brain barrier (BBB) breakdown in rat hippocampus. One and nine days after PNV injection, tracer was found in pinocytic vesicles crossing the endothelium and in the interendothelial cleft, suggesting that BBB breakdown had occurred through enhanced transendothelial transport and/or tight-junction opening. In the present work, we investigated the mechanisms by which PNV (850 microg/kg, i.v.) increased t

Water channels and urea transporters.

1. It now appears that when water crosses an endothelium which is not fenestrated, or an epithelium with tight junctions, it does so rapidly, and with low energy cost, only if the cell membrane contains an adequate number of specific water channels, encoded by one of at least six different genes. 2. The water channel genes so far cloned encode a series of integral membrane proteins called aquaporins, all of approximately 30 kDa (265-282 amino acids), in the unglycosylated state. All but one (AQP3) are specific water channels and all but one (AQP4) are inactivated by mercurial compounds. 3. Aqu

Differential cystine and dibasic amino acid handling after loss of function of the amino acid transporter b0,+AT (Slc7a9) in mice.

Cystinuria is an autosomal recessive disease caused by mutations in SLC3A1 (rBAT) and SLC7A9 (b(0,+)AT). Gene targeting of the catalytic subunit (Slc7a9) in mice leads to excessive excretion of cystine, lysine, arginine, and ornithine. Here, we studied this non-type I cystinuria mouse model using gene expression analysis, Western blotting, clearance, and brush-border membrane vesicle (BBMV) uptake experiments to further characterize the renal and intestinal consequences of losing Slc7a9 function. The electrogenic and BBMV flux studies in the intestine suggested that arginine and ornithine are
