Intracellular antigens (microtubule-associated protein copurified with glycosomal enzymes)--possible vaccines against trypanosomiasis.

African trypanosomes are motile unicellular eukaryotes that can cause diseases such as sleeping sickness in humans and nagana in animals, debilitating millions of people and livestock. All members of the Trypanosomatidae family contain subpellicular microtubules cross-linked to each other and to the plasma membrane by unique trypanosomal microtubule-associated proteins (MAPs). These MAPs may serve as specific intracellular target sites for therapeutic attack against trypanosomiasis. A trypanosomal MAP (p52) copurifies with two glycosomal enzymes (aldolase and GAPDH) on phosphocellulose columns…

Correlation of autoantibody titres with central nervous system pathology in experimental African trypanosomiasis.

CD-1 mice infected with the protozoan parasite Trypanosoma brucei brucei developed few signs of central nervous system pathology associated … to that seen in the fatal post-treatment reactive encephalopathies that can occur in human African trypanosomiasis. Enzyme-linked immunosorbent assays were used to measure autoantibody titres to double-stranded DNA, myelin basic protein and to the myelin-specific galactocerebrosides and gangliosides in groups of infected mice, with or without the post-treatment …

[Cardiac complications of American trypanosomiasis (Chagas disease). Various case reports and general observations].

Three young female patients with heart problems and positive serologic tests or characteristic histologic lesions for Chagas' disease (American trypanosomiasis) are reported, one of them having presented with acute anterior myocardial infarction with only minor coronary lesions. The most prominent features of the disease are discussed with emphasis on late cardiac complications. Chagas' cardiomyopathy is the leading cause of sudden death in Latin America, where an estimated 10 to 12 million people are infected in endemic areas from southern Mexico to southern Argentina. It has also been termed…

Human African trypanosomiasis: use of double centrifugation of cerebrospinal fluid to detect trypanosomes.

A double-centrifugation technique for the detection of trypanosomes in samples of cerebrospinal fluid is described and evaluated. The results of the analysis of samples of cerebrospinal fluid from 128 patients with Trypanosoma brucei gambiense sleeping sickness from the Daloa area of Côte d'Ivoire, obtained using single centrifugation, are compared with those obtained using the new method. Double centrifugation is at least twice as sensitive as single centrifugation and results in an increase of 37% in the early detection of late-stage case of the disease. The technique is easily implemented u

Cerebral trypanosomiasis in cattle with mixed Trypanosoma congolense and T. brucei brucei infections.

Six Boran steers were infected simultaneously with Trypanosoma congolense and T. brucei brucei while another group of 3 was inoculated with T. b. brucei one year after infection with T. congolense. Three further steers were infected with T. b. brucei alone. Whereas, the six animals which received simultaneous infections developed clinical signs of cerebral trypanosomiasis as evidenced by depression, ataxia and occasional circling, those infected with T. b. brucei alone did not. At necropsy, 4 out of the 6 simultaneously infected animals had a mild to severe disseminated non-suppurative meningo…

Lymphocyte function in experimental African trypanosomiasis. III. Loss of lymph node cell responsiveness.

The relationship between immunosuppression and suppressor cell activity in the lymphoid organs of animals with experimental African trypanosomiasis has been examined further. In the present study we measure the primary in vitro PFC response to SRBC by spleen and lymph node cells from Trypanosoma rhodesiense infected or drug-cured C57BL/6 mice. Passive transfer experiments with this culture system tested for the presence or absence of suppressor cells. We demonstrate that infected mice exhibit immunosuppression in the spleen cell population several weeks before becoming suppressed at the level …

Geospatial Analysis of Tsetse Infestation and Animal African Trypanosomiasis in Busia County, Kenya.

African trypanosomiasis (AT) is a vector-borne disease caused by protozoan parasites of the genus Trypanosoma, primarily transmitted through the bite of infected tsetse flies in the genus Glossina. The disease is a major constraint to livestock production, causing reduced productivity, anemia, and mortality in affected animals. A cross-sectional study was conducted from November to December 2024 to assess the spatial prevalence of AAT in cattle as well as tsetse fly abundance in Busia County, Kenya. A total of 762 cattle randomly selected from seven villages in Busia County were screened for A…

Dermal trypanosomes in seropositive suspects of gambiense human African trypanosomiasis in Côte d'Ivoire.

In the population at risk of gambiense human African trypanosomiasis (gHAT), the prevalence of extravascular parasite carriage remains unclear. Here, we conducted an observational clinical study in the hypo-endemic gHAT foci of Sinfra and Bonon in Côte d'Ivoire from 2019 to 2022. A total of 74 individuals were enrolled, including 45 suspects previously found positive at least once in a serological test for gHAT and followed by the national elimination programme of Côte d'Ivoire, as well as 29 seronegative controls. No significant differences between groups were observed for any epidemiological…

A CRISPR-based diagnostic tool to survey drug resistance in human African trypanosomiasis.

The World Health Organization aims to eliminate human African trypanosomiasis caused by Trypanosoma brucei gambiense (gHAT) by 2030. With the decline of reported cases, maintaining active surveillance is essential, including for the potential emergence of drug-resistant parasites. We have developed new highly specific diagnostic tools, using the Cas13a-based Specific High-Sensitivity Reporter Enzymatic UnLOCKing (SHERLOCK) technology, for the detection of drug-resistant genotypes that (i) are already circulating, such as the AQP2/3(814) chimera providing resistance to pentamidine and melarsopr…

Correlation of PET/CT and Brain MRI Findings in Human African Trypanosomiasis Encephalitis.

A 49-year-old woman from Cameroon presented with history of several months of worsening headache, lethargy, left arm weakness, and generalized lymphadenopathy. Brain MRI demonstrated multifocal signal abnormality and enhancement involving the bilateral basal ganglia and cerebral white matter. FDG PET/CT performed as part of lymphadenopathy evaluation demonstrated patchy areas of increased metabolic activity of the brain parenchyma. Human African trypanosomiasis or African sleeping sickness is a protozoan infection caused by Trypanosoma brucei gambiense transmitted by the tsetse fly in sub-Saha…

Advances in Drug Discovery against Neglected Tropical Diseases: Human African and American Trypanosomiasis.

Human African and American trypanosomiasis are the vector-borne parasitic diseases that have killed millions of people early, and many people are yet suffering from these neglected diseases. The causative agents of these infections are parasitic protozoans of the genus Trypanosoma. Current treatment regimens against these endemic diseases have several limitations in terms of safety, efficacy, route of administration, and some of them have lost efficacy due to the emergence of resistance in their respective parasites. In this review, the most promising compounds identified by different strategi…

Pharma to farmer: field challenges of optimizing trypanocide use in African animal trypanosomiasis.

Trypanocides are a key control component of African animal trypanosomiasis (AAT) in tsetse-infested areas of sub-Saharan Africa. While farmers are dependent upon trypanocides, recent research highlights their inappropriate and ineffective use, problems with drug quality, and treatment failure. There are currently gaps in knowledge and investment in inexpensive AAT diagnostics, understanding of drug resistance, and the effective use of trypanocides in the field. Without this important knowledge it is difficult to develop best practice and policy for existing drugs or to inform development and u…

Cardiac involvement in trypanosomiasis in sheep experimentally infected by Trypanosoma vivax (Ziemman, 1905).

The objective of the present study was to evaluate the clinical signs, electrocardiographic signs and evolution of histopathological lesions in the heart of sheep experimentally infected by Trypanosoma vivax during the acute and chronic phases of infection as well as to investigate the presence of parasitic DNA in the heart using polymerase chain reaction (PCR). Twenty-two male sheep were divided into the following four groups: G1, which consisted of six sheep infected by T. vivax that were evaluated until 20 days post-infection (dpi; acute phase); G2, which consisted of six sheep infected by

Trypanosomiasis challenge estimation using the diminazene aceturate (Berenil) index in Zebu in Gabon.

A longitudinal study was conducted within a cattle ranch in Gabon to determine the diminazene aceturate (Berenil) index (DAI) in a group of Zebu, raised under low tsetse density; this measure providing an assessment of trypanosomiasis risk. The objective was to evaluate the trypanosomiasis pressure thus informing trypanosomiasis control methods and cattle management. Twenty female adult Zebu were monitored for 24 weeks during the dry season. Blood samples were collected on a weekly basis and subjected to parasitological and haematological analysis (n = 480), using the buffy-coat method and the…
