Case Report: FAS spontaneous mutation in a familial hemophagocytic lymphohistiocytosis patient with a complex heterozygous mutation in PRF1.

Familial hemophagocytic lymphohistiocytosis type 2 (FHL2), caused by perforin 1 (PRF 1), is a rare and fatal autosomal recessive disorder characterized by a hyperinflammatory syndrome and the accumulation of activated T lymphocytes and histiocytes in the reticuloendothelial system. Autoimmune lymphoproliferative syndrome (ALPS) is an autoimmune disease that typically presents in children with lymphadenopathy, splenomegaly, and cytopenias or lymphomas. We report a case of a 9-year-old boy who was newly diagnosed with FHL, carrying a new type of compound heterozygous mutations (c.305G>T and c.13

Differential Expression of Proteins in an Atypical Presentation of Autoimmune Lymphoproliferative Syndrome.

Autoimmune lymphoproliferative syndrome (ALPS) is a rare disease defined as a defect in the lymphocyte apoptotic pathway. Currently, the diagnosis of ALPS is based on clinical aspects, defective lymphocyte apoptosis and mutations in Fas, FasL and Casp 10 genes. Despite this, ALPS has been misdiagnosed. The aim of this work was to go one step further in the knowledge of the disease, through a molecular and proteomic analysis of peripheral blood mononuclear cells (PBMCs) from two children, a 13-year-old girl and a 6-year-old boy, called patient 1 and patient 2, respectively, with clinical data s

Expression of TRAIL and Fas in Primary Hyperparathyroidism.

AIM: Differentiating between parathyroid lesions is still difficult and ambiguous. In cases of primary hyperparathyroidism, appropriate and prompt diagnosis is of great importance for effective treatment and follow-up. A great amount of mechanisms contribute to the pathogenesis of primary hyperparathyroidism, such as disturbance in balance between pro- and anti-apoptotic factors. Therefore, we examined whether immunohistochemical expression of apoptotic factors, TNF-related apoptosis-inducing ligand (TRAIL) and Fas, could have clinical utility as a marker of proliferative lesions of parathyroi

[Cell death of salivary gland epithelial cells and involvement of HTLV-I in Sjögren's syndrome].

Chronic sialadenitis in Sjögren's syndrome (SS) is associated with cell death induced by Fas or cytotoxic granules. Furthermore, tumor necrosis factor-related apoptosis inducing ligand or toll-like receptor3 are known to induce apoptosis in the salivary gland epithelial cells (SGECs) derived from patients with SS. Anti-apoptotic molecules that are closely related to epidermal growth factor are known to inhibit apoptosis. Epidemiologically, high prevalence of HTLV-I in primary Sjögren's syndrome (SS) patients has been found in an endemic area. However, by comparison of radiographic imaging with

Serum soluble Fas/APO-1 is increased in patients with primary Sjögren's syndrome.

The aim of the study was to elucidate the involvement of Fas antigen in human autoimmune disease, by analysing serum levels of soluble Fas/APO-1 protein in patients with various autoimmune diseases, including system lupus erythematosus (SLE), rheumatoid arthritis (RA), systemic sclerosis (SSc), polymyositis/dermatomyositis (PM/DM), Behçet's syndrome and Sjögren's syndrome (SjS). The levels of soluble Fas/APO-1 in sera were quantitated by a sandwich enzyme-linked immunosorbent assay. Soluble Fas/APO-1 levels were significantly increased in serum from patients with primary Sjögren's syndrome (pr

Precursor B cells for autoantibody production in genomically Fas-intact autoimmune disease are not subject to Fas-mediated immune elimination.

The Fas/Fas ligand (FasL) system participates in regulation of the immune system through the apoptotic process. However, the extent to which abnormalities in this system are involved in the loss of self-tolerance and development of autoimmune disease not associated with Fas/FasL mutations remains unknown. The present study addresses this issue in Fas/FasL-intact, systemic lupus erythematosus (SLE)-prone (NZB x NZW) (NZB/W) F1 mice. While splenic B cells from 2-month-old mice before overt SLE expressed Fas poorly, in vitro stimulation with an agonistic anti-CD40 mAb up-regulated their Fas expre

Exploring a mucoacting spray-dried nanoparticle-in-microparticle inhalable formulation for targeted pulmonary delivery of sorafenib in lung cancer therapy.

In this study, we developed an inhalable Nano-in-Micro (NiM) system for localized delivery of Sorafenib (Sor) in non-small cell lung cancer (NSCLC) therapy. Sor was encapsulated into mucus-penetrating and folate-targeted polymeric nanoparticles (FA-PPP-NPs@Sor), produced starting from the α,β-poly(N-2-hydroxyethyl)-D,L-aspartamide (PHEA), functionalised with poly(lactic-co-glycolic acid) (PLGA), and folate-conjugated polyethylene glycol (PEG-FA) FA (DDPLGA = 3.3  mol %, DDPEG-FA ≈ 8 chains/molecule). Nanoprecipitation produced 109  nm particles, easy to redisperse, with a DL of 10 wt%, which s

A Dual-Functional Magnetic Nanoparticle for Rapid Extraction and Targeted Tumor Delivery of Tiancimycin.

Anthraquinone-fused enediynes (AFEs) are a class of microbial natural products with remarkable cytotoxicity and exhibit promising application prospects in tumor therapy. Taking tiancimycins (TNMs) as an example, this class of compounds holds great potential against triple-negative breast and liver cancers. However, the low production titers of AFEs in actinobacterial hosts make their enrichment and separation processes both cumbersome and costly. Therefore, the main objective of this study was to develop a rapid and reusable magnetic platform for TNM enrichment and recovery. Inspired by the pi

Androgen excess uncouples circulating and hepatic lipid homeostasis in females on a high-calorie diet.

Androgen excess is a common feature of polycystic ovary syndrome, congenital adrenal hyperplasia, gender affirming hormone therapy and is a known driver of disrupted glucose homeostasis. However, its impact on hepatic lipid regulation remains poorly understood with conflicting results. In age and weight matched female mice, dihydrotestosterone (DHT) treatment under a regular chow diet did not alter hepatic or circulating lipid levels. In contrast, under a western diet, DHT protected against metabolic dysfunction-associated steatotic liver disease by reducing hepatic triglyceride (TG) accumulat

Folate-targeted PLGA nanoparticles enhance paclitaxel-induced cytotoxicity and apoptosis in B16 melanoma cells.

Melanoma is among the most aggressive forms of skin cancer, characterized by high metastatic potential and limited responsiveness to conventional chemotherapy. Paclitaxel (PTX) is an efficacious anticancer agent; however, its clinical utility in melanoma is restricted by poor aqueous solubility, non-specific biodistribution, and dose-limiting systemic toxicity. To overcome these limitations, this study aimed to develop and evaluate folate-targeted poly(lactic-co-glycolic acid) nanoparticles (FA-PTX-PLGA-NPs) as a targeted nanotherapeutic approach for melanoma treatment. Paclitaxel-loaded PLGA

Intestinal Acyl-CoA synthetase 5 (ACSL5) deficiency potentiates postprandial GLP-1 & PYY secretion, reduces food intake, and protects against diet-induced obesity.

OBJECTIVE: In the small intestine, the products of digestion of dietary triacylglycerol (TAG), fatty acids (FA) and monoacylglycerol, are taken up by absorptive cells, enterocytes, for systemic energy delivery. These digestion products can also bind receptors on endocrine cells to stimulate the release of hormones capable of influencing systemic energy metabolism. The initial phase of intestinal FA absorption involves the acylation of FAs to acyl-CoA by the acyl-CoA long chain synthetase (ACSL) enzymes. ACSL5 is abundantly expressed in the small intestinal epithelium where it is the major ACSL

Autoimmune lymphoproliferative syndrome presenting with glomerulonephritis.

Autoimmune lymphoproliferative syndrome (ALPS) is characterized clinically by chronic non-malignant lymphoproliferation and autoimmunity and is caused by a genetic defect in programmed cell death (apoptosis). Most patients with ALPS have heterozygous mutations in the Fas gene. We describe an 11-year-old Brazilian boy with hepatosplenomegaly, lymphadenopathy, hemolytic anemia, and hypergammaglobulinemia since early infancy. T cell lines from the patient were defective in Fas-mediated apoptosis. He was diagnosed as having ALPS and found to have a novel Fas gene mutation (IVS4+1G>A). In addition,

Resistance of lymphocytes to Fas-mediated apoptosis in Behçet's disease and Vogt-Koyangi-Harada syndrome.

BACKGROUND: Behçet's disease and Vogt-Koyanagi-Harada (VKH) syndrome are characterized by a severe intraocular inflammation with a long course and recurrent episodes. Activated autoreactive lymphocytes are considered to be responsible for the development of this inflammation. These lymphocytes may also be involved in the perpetuation and recurrence of this inflammation if they do not properly go into apoptosis. To investigate this hypothesis, a study was carried out to evaluate the resistance of lymphocytes to apoptosis in these two uveitis entities. METHODS: Blood samples were obtained from

[Effect of abnormal apoptosis in human decidual tissue during early gestation on pregnancy].

OBJECTIVE: To investigate the relationship between abnormal cellular apoptosis of the decidual tissue during the early stages of gestation and failure of pregnancy. METHODS: Samples of the decidual tissues of early gestation were obtained from 60 healthy women with pregnancy, and cellular apoptosis was assessed by using terminal transferase-mediated nick-end labeling (TUNEL). The expressions of Fas and FasL in the decidual tissues at different stages of gestation were examined by in situ hybridization and immunohistochemistry. RESULTS: On day 41 of normal pregnancy, evident apoptosis in the

Soluble Fas (APO-1/CD95) isoform in adult T-cell leukemia.

Fas (APO-1/CD95) consists mainly of 2 isoforms, membrane-anchored (mFas) and soluble (sFas), both of which can mediate apoptosis through the Fas-signalling process, not only in normal but also in leukemia T-cells. This suggests that aberrant expression of either mFas or sFas may affect the natural history of T-cell neoplasms, such as adult T-cell leukemia (ATL). For studying the tumor biology related to Fas-mediated apoptosis, ATL cells with up-regulated Fas proteins and its mRNAs are convenient and useful for understanding apoptotic oncology as it occurs in nature. Most attention, so far, has

Expression of Fas antigen on hepatocytes in viral hepatitis patients.

To evaluate the relationship between viral hepatitis and apoptosis, expression of Fas antigen in liver tissue sections of patients with viral hepatitis was investigated by immunohistochemical study using antibody to Fas antigen. A total of 46 liver tissues were obtained from 12 patients with acute viral hepatitis (AH) and 34 patients with chronic hepatitis (CH). Fas antigen was more markedly expressed in hepatocytes of AH patients than of CH patients. The Fas-positive hepatocytes of AH patients was localized rather diffusely in the inter lobular area and they were associated with closely to th

Mixed Lanthanide Oxide Nanoparticles Coated with Alginate-Polydopamine as Multifunctional Nanovehicles for Dual Modality: Targeted Imaging and Chemotherapy.

Integrating anticancer drugs and diagnostic agents in a polymer nanosystem is an emerging and promising strategy for improving cancer treatment. However, the development of multifunctional nanoparticles (NPs) for an "all-in-one" platform characterized by specific targeting, therapeutic efficiency, and imaging feedback remains an unmet clinical need. In this study, pH-responsive mixed-lanthanide-based multifunctional NPs were fabricated based on simple metal-ligand interactions for simultaneous cancer cell imaging and drug delivery. We investigated two new systems of alginate-polydopamine compl
