Ion-activated gellan gum in situ gel for prolonged ocular administration of tofacitinib citrate: An innovative treatment approach for dry eye disease.

This study developed a novel tofacitinib citrate-loaded in situ gel (TFC-ISG) using low-acyl gellan gum to enhance ocular retention and bioavailability for improved dry eye disease (DED) treatment. This formulation remained stable after storage at 40 °C/25 % RH for 6 months. It demonstrated rapid gel formation when mixed with artificial tears and exhibited shear-thinning properties. The ion-triggered sol-gel transition of gellan gum did not impede drug diffusion, as evidenced by unaltered release profiles and corneal/scleral permeability. Compared to the TFC solution formulation, this preparat

Pathogenic Mechanism of Dry Eye-Induced Chronic Ocular Pain and a Mechanism-Based Therapeutic Approach.

PURPOSE: Dry eye-induced chronic ocular pain is also called ocular neuropathic pain. However, details of the pathogenic mechanism remain unknown. The purpose of this study was to elucidate the pathogenic mechanism of dry eye-induced chronic pain in the anterior eye area and develop a pathophysiology-based therapeutic strategy. METHODS: We used a rat dry eye model with lacrimal gland excision (LGE) to elucidate the pathogenic mechanism of ocular neuropathic pain. Corneal epithelial damage, hypersensitivity, and hyperalgesia were evaluated on the LGE side and compared with the sham surgery side

Meibomian gland dysfunction is suppressed via selective inhibition of immune responses by topical LFA-1/ICAM antagonism with lifitegrast in the allergic eye disease (AED) model.

PURPOSE: The etiology of meibomian gland dysfunction (MGD) is incompletely understood, despite being a common ophthalmic condition and an area of unmet medical need. It is characterized by an insufficiency in glandular provision of specialized lipids (meibum) to the tear film and is a major cause of dry eye. Work in the allergic eye disease (AED) mouse model has revealed an immunopathogenic role in MGD causation, now raising interest in the applicability of immunomodulatory therapies. As such, we herein ask whether inhibition of lymphocyte function associated antigen (LFA)-1/intracellular adhe

Increased sensitivity of the non-human primate eye to microwave radiation following ophthalmic drug pretreatment.

Previous studies in our laboratory have established that pulsed microwaves at 2.45 GHz and 10 mW/cm2 are associated with production of corneal endothelial lesions and with disruption of the blood-aqueous barrier in the non-human primate eye. In the study reported here we examined ocular damage in monkeys (M. mulatta and M. fascicularis) following topical treatment with one of two ophthalmic drugs (timolol maleate and pilocarpine) that preceded exposure to pulsed microwaves. Anesthetized monkeys were sham exposed or exposed to pulsed, 2.45 GHz microwaves (10 microseconds, 100 pps) at average po

Permanent Vision Loss Due to Vascular Variation After Ear Hyaluronic Acid Injection: Two Case Reports and Literature Reviews.

BACKGROUND: In line with modern aesthetic principles, a prominent and voluminous auricle is considered to enhance facial structure by making it appear smaller and more refined. Recently, there has been a growing trend toward using injectable treatments as an alternative to traditional surgical procedures for correcting lying ears. This shift has led to an increase in reported complications associated with these interventions. OBJECTS: This report aims to investigate the mechanisms leading to vision loss following hyaluronic acid (HA) injections in the ear and to propose preventive measures an

Heating-driven self-assembled glycyrrhizin nanomicelles loading bisdemethoxycurcumin: Preparation, characterization, and efficacy evaluation on experimental dry eye.

In this study, a simple but novel preparation method was developed by heating a mixture of dipotassium glycyrrhizinate (DG) and bisdemethoxycurcumin (BDMC) in aqueous solution, and a DG self-assembled nanomicelles-loading BDMC (named B@DNM) ophthalmic solution was successfully fabricated with this heating-driven process. AutoDock simulation analysis revealed that Pi-Alkyl hydrophobic interactions between BDMC and DG played important role in this self-assembled B@DNM. The optimized B@DNM, with a DG:BDMC mass ratio of 40:1 and heating time of 6 h, had a high encapsulation efficacy of 96.70 ± 0.1
