Mortality among individuals with cannabis, cocaine, amphetamine, MDMA, and opioid use disorders: a nationwide follow-up study of Danish substance users in treatment.

This is a register-based cohort study of 20,581 individuals in treatment for illicit substance use disorders in Denmark between 1996 and 2006. All in all, 1441 deaths were recorded during 111,445 person-years of follow-up. Standardized mortality ratios (SMRs) associated with different primary substance types were calculated and Cox-regression analyses were performed in order to establish hazard ratios (HR) associated with injection drug use and psychiatric comorbidity. SMRs for primary users of specific substances were: cannabis: 4.9 (95% confidence interval (CI): 4.2-5.8), cocaine: 6.4 (CI: 3

[Hallucinogen induced psychic disorders. Subjective experiences, psychopathology and differential diagnosis].

The concepts of intoxication, intoxication psychosis, and drug-induced psychosis are frequently used to refer to psychopathological states caused by the intake of hallucinogenic substances. The considerable increase of intoxications with substances of the chemical class of amphetamines (in particular, "Ecstasy") in western countries should cause major concern. Moreover, the number of schizophrenic patients who also use hallucinogenic drugs is increasing. High incidence and comorbidity cause clinical problems and call for detailed knowledge of the drug related acute and chronic effects of the v

Chronic methamphetamine psychosis after long-term abstinence in Japanese incarcerated patients.

Eighty female incarcerated subjects who maintained a long-term methamphetamine (Meth) abstinence, but manifested psychosis, were examined for correlations among symptom severity premorbid diathesis, recurrence, and adverse effects of pharmacotherapy. Baseline symptoms were assessed with a 24-item version of the Brief Psychiatric Rating Scale (BPRS) prior to, and 2 weeks after, the commencement of pharmacotherapy. Fifty-four subjects who had abused volatile solvents in their teenage years commenced Meth abuse at a significantly lower age than the rest of the subjects (18.4 vs. 22.5 years old, p

Epigenetic landscape of amphetamine and methamphetamine addiction in rodents.

Amphetamine and methamphetamine addiction is described by specific behavioral alterations, suggesting long-lasting changes in gene and protein expression within specific brain subregions involved in the reward circuitry. Given the persistence of the addiction phenotype at both behavioral and transcriptional levels, several studies have been conducted to elucidate the epigenetic landscape associated with persistent effects of drug use on the mammalian brain. This review discusses recent advances in our comprehension of epigenetic mechanisms underlying amphetamine- or methamphetamine-induced beh

Repeated administration of amphetamine induces a shift of the prefrontal cortex and basolateral amygdala motor function.

The role of the medial prefrontal cortex (mPFC) and basolateral amygdala (BLA) in the expression of behavioural locomotor sensitization to amphetamine (Amph) has been poorly studied. In the present study, we investigated how lidocaine infused in the mPFC or BLA modulated motor responses to acute and repeated (sensitization) Amph administration. We showed that reversible blockade of mPFC or BLA by lidocaine increased both locomotor and rearing responses to acute Amph, but blocked the expression of behavioural sensitization to Amph. These findings indicate that under free-lidocaine conditions re

Prevalence of gamma-hydroxybutyrate (GHB) in serum samples of amphetamine, metamphetamine and ecstasy impaired drivers.

Two hundred and forty-seven serum samples which have been collected by police during roadside testing and have been found positive for amphetamine, methamphetamine, 3,4-methylenedioxyamphetamine (MDA), 3,4-methylenedioxymethamphetamine (MDMA) and/or 3,4-methylenedioxyethamphetamine (MDE) were analyzed for gamma-hydroxybutyrate (GHB). Serum samples were spiked with deuterated GHB as internal standard and acetonitrile was added to achieve dilution and protein precipitation. Samples were analyzed with a LC-MS/MS system operated in the multiple reaction monitoring mode (MRM) using a TurboIonSpray

Effects of dopamine and glutamate on synaptic plasticity: a computational modeling approach for drug abuse as comorbidity in mood disorders.

Major depressive disorder (MDD) affects about 16% of the general population and is a leading cause of death in the United States and around the world. Aggravating the situation is the fact that "drug use disorders" are highly comorbid in MDD patients, and VICE VERSA. Drug use and MDD share a common component, the dopamine system, which is critical in many motivation and reward processes, as well as in the regulation of stress responses in MDD. A potentiating mechanism in drug use disorders appears to be synaptic plasticity, which is regulated by dopamine transmission. In this article, we descr

Impulsivity and positive psychotic symptoms influence hostility in methamphetamine users.

Methamphetamine (MA) use is associated with hostility, aggression, and positive psychotic symptoms. However, little is known of the processes or mechanisms that underlie this relationship. The present research was designed to investigate putative mediating and moderating variables between MA dependence and hostility in a sample of injecting MA users (N=237). Both positive symptoms of psychosis and higher levels of impulsivity functioned as mediators and moderators of this relationship. This pattern of findings suggests that MA use leads to greater hostility by increasing positive psychotic sym

Quitting ecstasy: an investigation of why people stop taking the drug and their subsequent mental health.

The regular use of ecstasy (3,4-methylenedioxymethamphetamine, MDMA) has been associated with depressed mood, anxiety and hostility, but it is not known whether such effects persist after people stop using the drug. Furthermore, little is known about what factors might influence the decision to quit using MDMA. The aim of the present study was to examine the reasons why ex-users had stopped using this drug and to assess their current levels of depression, anxiety, anger and aggression. Telephone interviews were conducted with people who used to take MDMA on a regular basis but who no longer us

Sexual abuse, physical abuse, and posttraumatic stress disorder among women participating in outpatient drug abuse treatment.

Findings from a prospective, longitudinal study of 182 women and 148 men in outpatient drug abuse treatment programs indicate that women are significantly more likely than men to experience sexual and physical abuse. Sexual and physical abuse are associated with higher levels of posttraumatic stress disorder (PTSD) symptomatology. Moreover, women are more likely than men to possess an array of psychological factors that predict relapse to drug use after treatment, including low self-esteem, depression, anxiety, and suicidal behavior, among others. But contrary to expectation, PTSD is not assoc

Outpatient treatment and outcome of prescription drug abuse.

Forty-six consecutive patients who voluntarily sought outpatient treatment for abuse of one or more prescription drugs were studied. Barbiturates, amphetamines, and diazepam were the most common drugs abused. Desired treatments by patients included counseling, medical withdrawal, or medical maintenance with the drug of abuse or a chemically related drug. Twenty-two (47.8%) patients left treatment and relapsed within one month; another eight (17.4%) patients relapsed between one and three months after entering treatment. Only 13 (28.3%) reported abstinence 90 days after entering treatment. This

Nanoparticle-Mediated Brain-Targeted Drug Delivery for Methamphetamine Use Disorder: Advances, Challenges and Translational Perspectives.

Methamphetamine use disorder (MUD) poses a significant global health challenge, lacking FDA-approved pharmacotherapies. Despite extensive exploration of therapeutic targets and interventions, effective treatment remains scarce due to challenges in delivering therapeutic agents to relevant brain regions at effective concentrations. Nanoparticle-mediated delivery offers a promising approach for transporting various therapeutic cargos to the central nervous system, including small molecules, nucleic acids, peptides, proteins, and gene-editing systems. However, the application of nanomedicine stra

Gender differences in the impact of psychological distress on methamphetamine use disorder outcomes and treatment effect.

BACKGROUND AND AIMS: Methamphetamine use disorder (MUD) is a major public health concern, often complicated by co-occurring psychological distress (PD). Evidence suggests gender differences in both the prevalence of PD and its impact on treatment outcomes. This study examined impacts of PD on MUD treatment outcomes, focusing on gender differences. DESIGN: Secondary analysis of pooled data from five randomized controlled trials of pharmacotherapy for MUD available on the NIDA DataShare site (accessed 19 October 2024). Individual participant data meta-analysis methods were used, adjusting for s

Childhood Maltreatment and Drug Craving in Patients With Methamphetamine Use Disorder: The Independent and Serial Mediating Roles of Emotional Dysregulation and Impulsivity.

Growing evidence suggests that childhood maltreatment (CM) is associated with adverse emotional and behavioural outcomes in individuals with methamphetamine use disorder (MUD). However, the psychological mechanisms underlying the association between CM and drug craving remain insufficiently understood. This study examined whether emotional dysregulation (ED) and impulsivity independently and serially mediated the association between CM and drug craving in patients with MUD. A total of 407 male inpatients with MUD were recruited from a drug rehabilitation centre in China. CM, ED, impulsivity an

A History of Methamphetamine Use Disorder in People with HIV Is Associated with Altered Functional Response to Risky Choice.

Methamphetamine (METH) use is highly prevalent among people with HIV (PWH) and those at risk and may contribute to overall worse health outcomes. Poorer health-related problems may be mediated by METH enhancing risky decision-making among PWH. While both METH and HIV are known to have overlapping and deleterious effects on the frontostriatal neural circuitry essential for decision-making, few studies have examined their combined effects. Eighty-eight participants stratified by HIV and a history of METH use disorder completed a risky decision-making paradigm, which involved choosing among safe

Evaluating The Efficacy Of Cognitive Behavioural Therapy For Amphetamine Use Disorder: Impact Of Socio-Demographic And Behavioural Factors In Pakistan.

BACKGROUND: Amphetamine use disorder (AUD) is a public health concern on a global scale and is a rising epidemic in Pakistan. Cognitive Behavioural Therapy (CBT) is a well-established treatment for substance use disorders, but its efficacy may be dependent on what is known as socio demographic and behavioural variables. The aims of this study are to provide evaluation of efficacy of CBT for AUD in Pakistani population and to find whether the presence of factors like education, job, duration of addiction, residence, age, family support, smoking and alcohol intake may affect treatment outcomes.
