Association of TREM-1, IL-1β, IL-33/ST2, and TLR Expressions With the Pathogenesis of Ocular Toxoplasmosis in Mouse Models on Different Genetic Backgrounds.

Ocular toxoplasmosis (OT) is one of the most common causes of posterior uveitis. The signaling of triggering receptor expressed on myeloid cells (TREM)-1 amplifies inflammation, whereas TREM-2 signaling is anti-inflammatory. IL-1β is a major driver of inflammation during infection. Toll-like receptors (TLRs) play important roles in protective immune response during Toxoplasma gondii infection, and interleukin (IL)-33 receptor (T1/ST2) signaling prevents toxoplasmic encephalitis in mice. However, the pathogenic mechanisms of OT are not yet well elucidated. To investigate the role of TREM-1, TRE

Mitochondrial DNA nucleotide changes in primary congenital glaucoma patients.

PURPOSE: Primary congenital glaucoma (PCG) is the second most common cause of blindness, accounting for 0.01%-0.04% of total blindness worldwide. Most congenital glaucoma cases are mapped to the GLC3A locus, and many aspects of PCG are still unknown. Recent studies have reported an increased frequency of mitochondrial DNA (mtDNA) sequence changes in primary open-angle glaucoma, primary angle-closure glaucoma, and pseudoexfoliation glaucoma compared to controls. Thus, this study was planned with the aim of detecting mitochondrial DNA variations in PCG cases. METHODS: Twenty primary congenital

Presynaptic metabotropic glutamate receptors in adult and developing neurons: autoexcitation in the olfactory bulb.

The mitral cell of the olfactory bulb is the primary relay neuron that transmits information from the olfactory receptors to the rest of the brain. This excitatory neuron releases glutamate from presynaptic dendrites and axon terminals. All rat mitral cells studied showed strong, selective, and widespread metabotropic glutamate receptor mGluR1 alpha immunoreactivity on the presynaptic membrane of dendrites, often at the synaptic vesicle release site, when examined with light and electron microscopy. The finding of glutamate receptors on mitral cell secondary dendrites supports the conclusion t

Acute Environmental Triggers and Intermediate-Term Modulators of Emergency Migraine-Related Health Care Encounters.

BACKGROUND AND OBJECTIVES: Migraine activity is shaped not only by the biological vulnerability but also by environmental factors that may influence susceptibility and contribute to fluctuations in migraine occurrence, although the evidence remains inconsistent. We aimed to evaluate the intermediate-term and short-term effects of environmental exposures on migraine activity and to examine a conceptual layered model comprising biological vulnerability, intermediate-term environmental modulators, and acute triggers. METHODS: We conducted a population-based case-crossover study among patients wi

Ononin-modified Chitosan/β-glycerophosphate hydrogel-loaded adipose-derived stem cells promote the repair of diabetic wounds in rats.

OBJECTIVE: To evaluate the mechanistic synergy of an ononin-modified chitosan/β-glycerophosphate (CS/β-GP) hydrogel loaded with adipose-derived stem cells (ADSCs) in repairing diabetic wounds (DWs). We specifically aimed to determine if ononin could biochemically remodel the hostile diabetic microenvironment to enhance ADSCs survival and potency. METHODS: Primary rat ADSCs were characterized by flow cytometry. A thermosensitive CS/β-GP hydrogel was fabricated, and its microstructure was characterized by scanning electron microscopy (SEM). he biological response of ADSCs to ononin was assessed

Arimoclomol-loaded Pluronic F127 nanomicelles in experimental endometriosis: drug incorporation, in vitro release, and early morphological and histological responses.

Endometriosis is a chronic, estrogen-dependent inflammatory disease characterized by ectopic endometrial-like glands and stroma, persistent pelvic inflammation, pain, infertility, and frequent recurrence. The limited availability of nonhormonal and lesion-directed therapies supports the investigation of alternative pharmacological mechanisms and regional drug-delivery strategies. Arimoclomol is a co-inducer of the endogenous heat-shock response with the capacity to amplify stress-dependent proteostasis, whereas Pluronic F127 is an amphiphilic PEO-PPO-PEO copolymer that forms drug-associated mi

Routine childhood vaccination rates in an academic family health team before and during the first wave of the COVID-19 pandemic: a pre-post analysis of a retrospective chart review.

BACKGROUND: There has been concern about declining routine vaccination rates during the COVID-19 pandemic. We evaluated the impact of the COVID-19 pandemic on early childhood vaccination rates at 2 sites of an academic family health team in the Greater Toronto Area, Ontario, serving both an urban and suburban patient population. METHODS: We conducted a pre-post analysis of vaccination records from Jan. 1, 2018, to Nov. 30, 2020, for a cohort of children born between Jan. 1, 2018, and Aug. 31, 2020, from the electronic medical record (EMR) of the Mount Sinai Academic Family Health Team (includ

Adult-onset CNS myelin sulfatide deficiency is sufficient to cause Alzheimer's disease-like neuroinflammation and cognitive impairment.

BACKGROUND: Human genetic association studies point to immune response and lipid metabolism, in addition to amyloid-beta (Aβ) and tau, as major pathways in Alzheimer's disease (AD) etiology. Accumulating evidence suggests that chronic neuroinflammation, mainly mediated by microglia and astrocytes, plays a causative role in neurodegeneration in AD. Our group and others have reported early and dramatic losses of brain sulfatide in AD cases and animal models that are mediated by ApoE in an isoform-dependent manner and accelerated by Aβ accumulation. To date, it remains unclear if changes in speci

Follicular dendritic cell sarcoma: clinicopathologic study of 15 cases with emphasis on novel expression of MDM2, somatostatin receptor 2A, and PD-L1.

Follicular dendritic cell sarcoma (FDCS) is a rare low-grade neoplasm with the phenotype of FDC cells. This rare sarcoma has been well known for being mistaken for a variety of neoplasms (mainly meningioma), particularly at extranodal sites. Diagnosis of FDCS mainly relies on characteristic histologic appearance supplemented by immunohistochemistry and electron microscopy. In this study, we reviewed 15 FDCSs retrieved from our consultation files and stained them for newly reported or novel markers (PD-L1, Rb1, MDM2, and somatostatin receptor 2A [SSTR2A]) in addition to conventional FDC markers

Metal-Chelating Polymers (MCPs) with Zwitterionic Pendant Groups Complexed to Trastuzumab Exhibit Decreased Liver Accumulation Compared to Polyanionic MCP Immunoconjugates.

Metal-chelating polymers (MCPs) can amplify the radioactivity delivered to cancer cells by monoclonal antibodies or their Fab fragments. We focus on trastuzumab (tmAb), which is used to target cancer cells that overexpress human epidermal growth factor receptor 2 (HER2). We report the synthesis and characterization of a biotin (Bi) end-capped MCP, Bi-PAm(DET-DTPA)36, a polyacrylamide with diethylenetriaminepentaacetic acid (DTPA) groups attached as monoamides to the polymer backbone by diethylenetriamine (DET) pendant groups. We compared its behavior in vivo and in vitro to a similar MCP with

Surveillance for human West Nile virus disease - United States, 1999-2008.

PROBLEM/CONDITION: West Nile virus (WNV) is an arthropod-borne virus (arbovirus) in the family Flaviviridae and is the leading cause of arboviral disease in the United States. An estimated 80% of WNV infections are asymptomatic. Most symptomatic persons develop an acute systemic febrile illness that often includes headache, myalgia, arthralgia, rash, or gastrointestinal symptoms. Less than 1% of infected persons develop neuroinvasive disease, which typically presents as encephalitis, meningitis, or acute flaccid paralysis. REPORTING PERIOD: 1999-2008. DESCRIPTION OF SYSTEM: WNV disease is a

Addressing Clinical Challenges of Platinum Anticancer Drugs through Rational Chemical Design.

Platinum (Pt)-based anticancer drugs have been a cornerstone of chemotherapy for decades, yet their clinical application remains constrained by dose-limiting systemic toxicity and drug resistance. In this Account, we summarize our systematic efforts to address these challenges through two complementary strategies: 1) functionalization of Pt(IV) prodrugs and 2) spatially controlled targeted delivery. The kinetic inertness and octahedral geometry of Pt(IV) complexes offer a versatile platform for axial functionalization, allowing the integration of diverse bioactive ligands that are released upo

A systematic review of paediatric-specific risk factors for nosocomial infections in paediatric intensive care units, 2005-2024.

BACKGROUND: Children admitted to a paediatric intensive care unit (PICU) are at markedly higher risk of nosocomial infections (NIs), which correlate with elevated mortality, extended hospital stay, and amplified healthcare burden. Children exhibit distinct immunological and physiological traits compared with adult patients, highlighting an urgent need for paediatric-specific evidence to guide infection prevention. The latest existing systematic review on this topic was published in 2009, and did not cover emerging multi-drug-resistant infections (MDRIs) and modern paediatric critical care mana

Nitrated Nuclear Receptor Coactivator 4 Contributes to Ferritinophagy in Hyperhomocysteinemia-Induced Liver Injury.

AIMS: Hyperhomocysteinemia (HHcy) has been established as a significant risk factor for liver diseases, and its impact on nitrative stress and protein nitration has also been recognized. Nuclear receptor coactivator 4 (NCOA4) is crucial in maintaining iron homeostasis and regulating ferroptosis. However, limited studies are available regarding the post-translational modifications (PTMs) of NCOA4 in liver injury. RESULTS: Mice fed with a diet containing 2.5% methionine and water with 1.8 g/L DL-Hcy were used in this study. The HHcy-induced liver injury phenotype was assessed through serum aspa

Calcium-enhanced COF platforms for synergistic photothermal-antibacterial therapy: Unlocking calcium-mediated membrane disruption and ion overload for bacterial and biofilm eradication in wound care and clinical nursing.

The escalating threat of antibiotic-resistant bacteria and biofilm-associated infections necessitates the development of non-antibiotic therapeutic strategies with novel mechanisms of action. Here, we report a synergistic antibacterial platform based on a calcium-functionalized covalent organic framework (BQ-Ca-COF), designed to exploit the dual role of calcium ions (Ca2+) as both structural modulators and bioactive effectors. Using a solvothermal approach, we synthesized a series of COFs incorporating a dipyrazino [2,3-f:2',3'-h] quinoxalin (DPQ) linkage, enabling precise regulation of both f

Perceptions, Barriers and Pathways Forward: A Scoping Review of CALD Participation in Australian Clinical Trials.

BACKGROUND: Equitable participation in clinical trials is essential for ensuring that research evidence is scientifically robust, ethically just and representative of the populations it is intended to serve. In Australia, culturally and linguistically diverse (CALD) populations remain underrepresented in clinical trials despite increasing national policy attention to equity and inclusion. This review synthesized empirical evidence and Australian policy and practice documents to examine factors influencing CALD participation in clinical trials. OBJECTIVES: To synthesize evidence on CALD attitu

Detection of Sarcocystis hominis, Sarcocystis bovifelis, Sarcocystis cruzi, Sarcocystis hirsuta and Sarcocystis sigmoideus sp. nov. in carcasses affected by bovine eosinophilic myositis.

Bovine eosinophilic myositis is an inflammatory myopathy characterized by multiple focal or diffuse grey to green patches leading to condemnation of affected carcasses. Although its etiology is still uncertain, there is evidence that Sarcocystis species may play a role in the development of eosinophilic myositis. The goal of the present study was to identify Sarcocystis spp. in intralesional and extralesional tissues of condemned cattle carcasses, in order to evaluate the possible role of different bovine Sarcocystis spp. in the etiology of bovine eosinophilic myositis. Muscle samples (n = 100

Presence of an ultra-small microbiome in fermented cabbages.

BACKGROUND: Ultramicrobacteria (UMB), also known as ultra-small bacteria, are tiny bacteria with a size less than 0.1 µm3. They have a high surface-to-volume ratio and are found in various ecosystems, including the human body. UMB can be classified into two types: one formed through cell contraction and the other that maintains a small size. The ultra-small microbiome (USM), which may contain UMB, includes all bacteria less than 0.2 µm in size and is difficult to detect with current methods. However, it poses a potential threat to food hygiene, as it can pass through sterilization filters and

Extraction of Information Related to Drug Safety Surveillance From Electronic Health Record Notes: Joint Modeling of Entities and Relations Using Knowledge-Aware Neural Attentive Models.

BACKGROUND: An adverse drug event (ADE) is commonly defined as "an injury resulting from medical intervention related to a drug." Providing information related to ADEs and alerting caregivers at the point of care can reduce the risk of prescription and diagnostic errors and improve health outcomes. ADEs captured in structured data in electronic health records (EHRs) as either coded problems or allergies are often incomplete, leading to underreporting. Therefore, it is important to develop capabilities to process unstructured EHR data in the form of clinical notes, which contain a richer docume
