Perennial allergic rhinitis is an inflammatory disorder characterized by symptoms of nasal congestion, rhinorrhea, sneezing, and itching. The prevalence of allergic rhinitis is quite common and affects 20% or more of various populations. Some patients with allergic rhinitis are symptomatic only during the pollen season, while many others are allergic to multiple allergens including indoor allergens such as house dust mites, animal dander, cockroaches, and fungi, which lead to perennial symptoms. Immunoglobulin (Ig)-E is the proximate cause of perennial allergic rhinitis. Circulating IgE antibo

In Sri Lanka, malaria is transmitted mainly by Anopheles culicifacies Giles sensu lato (Diptera: Culicidae). In India, this nominal taxon comprises sibling species A, B, C, D and E, distinguished by their chromosome morphology. Species B (identified by polytene chromosome sequence Xab, 2g1 + h1) is not such an efficient vector of malaria as other members of the An. culicifacies complex in India. All specimens of An. culicifacies s.l. examined from Sri Lanka possess Xab, 2g1 + h1 polytenes, previously interpreted as species B, despite their important vector status. Recently, species E was descr…

BACKGROUND: Insecticide resistance among malaria vectors remains a critical obstacle to effective vector control and malaria elimination across sub-Saharan Africa. This study assessed the phenotypic and genotypic resistance profiles of Anopheles coluzzii populations in three urban centres of Anambra State, Southeastern Nigeria: Awka, Onitsha and Nnewi.
METHODS: Larvae were collected from Awka, Onitsha and Nnewi and reared to adults for WHO insecticide susceptibility tests. Species identification, molecular form determination (PCR-RFLP using HhaI enzyme) and screening for kdr L1014F and ace-1 …

In the malaria vector Anopheles gambiae s.l. Toll and Imd immune pathways signal through Re1l and Rel2 NF-κB transcription factors, respectively. Toll/Rel1 and Imd/Rel2 have been characterized mostly in the context of defense against malaria parasites, exhibiting Plasmodium species-specific roles. There is also evidence associating Rel2 with mosquito resistance to bacterial infections, however, the role of Rel1 in this context remains questionable, especially that there have been no rigorous functional genetic studies assessing Rel1 and Rel2 contributions side-by-side to mosquito defense durin…

Large-scale surveillance and informed vector control approaches are urgently needed to ensure that national malaria programs remain effective in reducing transmission and, ultimately, achieving malaria elimination targets. In South America, Anopheles darlingi is the primary malaria vector and is responsible for the majority of Plasmodium species transmission. However, little is known about the molecular markers associated with insecticide resistance in this species. In this study, we developed a low-cost, high throughput amplicon sequencing ("amp-seq") panel, consisting of 11 amplicons targeti…

The physiological response of mosquitoes to cold is poorly understood, particularly for Anopheles mosquitoes, where overwintering is rarer than in Aedes and Culex. The effect of cold selection was examined in two laboratory strains of the major malaria vector An. arabiensis that differ in insecticide resistant phenotypes. This is due to previous studies that demonstrated an increased tolerance for high temperatures in insecticide resistant An. arabiensis. The insecticide unselected SENN strain and the insecticide-resistant SENN-DDT strain were reared at decreasing fluctuating temperatures for …

Sanaria® has pioneered production of aseptic, purified, vialed cryopreserved Plasmodium falciparum (Pf) sporozoites (SPZ) as vaccines and for controlled human malaria infections. More than 3,500 individuals have received more than 9,700 injections of PfSPZ, worldwide. The PfSPZ are manufactured in aseptically reared female Anopheles stephensi mosquitoes. Since PfSPZ vaccines are intended primarily for some of the most disadvantaged people in the world, keeping costs low is imperative. One approach to reducing cost of goods is to eliminate male mosquitoes from the production process, thereby do…

BACKGROUND: Anopheles mosquitoes and the malaria parasites they transmit remain a significant global health problem. Most genomic and functional genomic studies of mosquitoes have focused on the protein-coding genome, and comparatively little is known about the importance of noncoding transcriptional enhancers in controlling their gene expression and phenotypic variation. Here we evaluate nine enhancers previously identified in a STARR-seq screen and present in a genetic locus that was identified as a major influence on susceptibility to malaria infection in wild Anopheles coluzzii mosquitoes.…

BACKGROUND: Sri Lanka has been malaria-free since 2012, yet the risk of malaria re-introduction is high during the post-malaria elimination phase due to influx of infected individuals from endemic countries, the presence of competent Anopheles vectors, and suitable climatic conditions for vectors across Sri Lanka. This may lead to the re-emergence of autochthonous malaria in areas beyond the historically affected dry and intermediate zones. To sustain the malaria-free status, a better understanding of Anopheles species across Sri Lanka's climatic zones is essential. This information will suppo…

This study systematically evaluated the efficiency of rose bengal and methylene blue as photosensitizers against the immature aquatic stages of Anopheles pharoensis. Genetic identification using the COI partial sequence confirmed the species, and the obtained sequence was submitted to GenBank (Accession No. PQ346929). Both photosensitizers exhibited 100% mortality in larvae I within 24 h at their highest concentrations, demonstrating strong biocidal activity. LC50 values for rose bengal increased from 1.50 ppm (24 h) and 1.34 ppm (48 h) in larvae I to 3.83 ppm (24 h) and 3.12 ppm (48 h) in pup…

BACKGROUND: The efficacy of LLINs is compromised by the emergence of pyrethroid resistance in malaria vectors. New insecticides that target other sites of action in the mosquito, including pyriproxyfen (PPF), are needed to maintain this efficacy. METHODS: Various populations of An. gambiae s.l. collected from April to November 2022 in 4 Beninese communes and the susceptible strain (Kisumu) were used to assess their susceptibility to PPF via CDC bottle tests and to assess the bioefficacy of Royal guard® (RG) alpha-cypermethrin + pyriproxyfen-impregnated nets via WHO cone tests. BACKGROUND: Malaria, transmitted by the bite of infective female Anopheles mosquitoes, remains a global public health problem. The presence of invasive Anopheles stephensi, capable of transmitting Plasmodium vivax and Plasmodium falciparum, was first reported in Ethiopia in 2016. The ecology of this mosquito species differs from that of Anopheles arabiensis, the primary malaria vector in Ethiopia. This study aimed to evaluate the efficacy of selected insecticides, which are used in indoor residual spraying (IRS) and selected long-lasting insecticidal nets (LLINs) for malaria vector control aga…

BACKGROUND: The demonstration that the recently discovered Anopheles symbiont Microsporidia MB blocks malaria transmission in Anopheles arabiensis and undergoes vertical and horizontal transmission suggests that it is a promising candidate for the development of a symbiont-based malaria transmission-blocking strategy. The infection prevalence and characteristics of Microsporidia MB in Anopheles gambiae sensu stricto (s.s.), another primary vector species of malaria in Kenya, were investigated.
METHODS: Field-collected females were confirmed to be Microsporidia MB-positive after oviposition. E…

BACKGROUND: Malaria transmission in Africa is facilitated by multiple species of Anopheles mosquitoes. These vectors have different behaviors and vectorial capacities and are affected differently by vector control interventions, such as insecticide-treated nets and indoor residual spraying. This review aimed to assess changes in the contribution of different vector species to malaria transmission in east and southern Africa over 20 years of widespread insecticide-based vector control.
METHODS: We searched PubMed, Global Health, and Web of Science online databases for articles published betwee…

Anopheles (Cellia) dirus Peyton & Harrison and Anopheles baimaii Sallum & Peyton are sibling species within the Dirus complex belonging to the Leucosphyrus group, and have been incriminated as primary vectors of malaria in Thailand. In the present study, DNA barcoding and geometric morphometrics were used to distinguish between An. dirus and An. baimaii in the international border areas, Trat Province, eastern Thailand. Our results revealed that DNA barcoding based on the cytochrome c oxidase subunit I gene could not be used to distinguish An. dirus from An. baimaii. The overlapping values bet…

Malaria parasites require pantothenate from both human and mosquito hosts to synthesize coenzyme A (CoA). Specifically, mosquito-stage parasites cannot synthesize pantothenate de novo or take up preformed CoA from the mosquito host, making it essential for the parasite to obtain pantothenate from mosquito stores. This makes pantothenate utilization an attractive target for controlling sexual stage malaria parasites in the mosquito. CoA is synthesized from pantothenate in a multi-step pathway initiated by the enzyme pantothenate kinase (PanK). In this work, we manipulated A. stephensi PanK acti

BACKGROUND: Although malaria and Anopheles mosquito vectors are highly prevalent in Côte d'Ivoire, limited data are available to help understand the malaria vector density and transmission dynamics in areas bordering the country. To address this gap, the Anopheles mosquito species diversity, the members of the Anopheles gambiae complex and the transmission of malaria were assessed in four health districts along the borders of Côte d'Ivoire.
METHODS: From July 2016 through December 2016 and July 2017 through December 2017, adult Anopheles mosquitoes were collected in four health districts of C…
