ATP Synthase Inhibitory Factor-1 Deficiency Attenuates Doxorubicin Cardiotoxicity by Preserving Mitochondrial Structure and Function.

Doxorubicin (DOX) remains an effective chemotherapeutic agent, but its clinical use is limited by dose-dependent cardiotoxicity. Mitochondrial dysfunction and metabolic remodeling are central features of DOX-induced cardiac injury. ATP synthase inhibitory factor-1 (IF1) is an endogenous inhibitor of the hydrolytic activity of mitochondrial ATP synthase and has emerged as an important regulator of cellular bioenergetics. Cardiac IF1 expression is increased in multiple pathological conditions; however, its role in chemotherapy-induced cardiotoxicity remains unclear. Here, we investigated the con…

A Cardio-Oncological Mice Model of Anthracycline-Induced Cardiotoxicity in Breast Tumor-Bearing Immunocompetent Mice.

The recent update from the European Society of Cardiology on preclinical models of cancer therapy-related cardiac dysfunction highlights a critical need for more representative and translational in vivo models. It pinpoints the need to develop models that employ immunocompetent, tumor-bearing animals, incorporating multiple strains and both sexes, to more accurately recapitulate the heterogeneity observed in clinical populations. Therefore, a clinically relevant model of anthracycline-induced cardiotoxicity (AIC) in breast tumor-bearing animals was built. According to mice' genetic background,…

Chicory Extract Alleviates Anthracycline-Induced Cardiotoxicity by Inhibiting Mitochondrial Damage via the UCP2/NLRP3 Pathway.

Doxorubicin (Dox)-induced cardiotoxicity (DIC) was characterized by severe myocardial damage that might progress to irreversible heart failure. There were limited options available for the prevention and treatment of DIC. Chicory (Cichorium intybus L.) has demonstrated notable cardioprotective effects. However, its potential to mitigate DIC remains unexplored. This study aimed to assess the therapeutic potential of chicory in alleviating DIC and elucidate its active ingredients and potential molecular mechanism. Male Sprague-Dawley (SD) rats were used to construct DIC models. The rats were pro…

Acteoside ameliorates doxorubicin-induced cardiotoxicity by suppressing ferroptosis through activation of the KEAP1/NRF2 pathway.

Doxorubicin (DOX)-induced cardiotoxicity (DIC) is a major limitation to the clinical use of DOX, highlighting the need for effective cardioprotective strategies. Although acteoside (ACT), a natural compound with antioxidant properties, has shown potential cardioprotective effects, its role in DIC, particularly in the regulation of ferroptosis, remains unclear. This study therefore investigated the protective effects of ACT against DIC and the underlying molecular mechanisms. Using C57BL/6 mice in vivo and H9c2 cells in vitro, we evaluated cardiac function, myocardial injury and histopathologic…

Infection-associated lipopolysaccharide aggravates doxorubicin-induced cardiotoxicity via modulation of the FOXO1-GPX4 ferroptosis pathway.

Doxorubicin (DOX) is an effective chemotherapeutic agent, but its clinical utility is limited by dose-dependent cardiotoxicity. However, the mechanisms by which infection exacerbates DOX-induced myocardial injury remain unclear. This study investigated the role of ferroptosis in lipopolysaccharide (LPS)-potentiated DOX-mediated myocardial damage. Clinical data were analyzed to evaluate the association between infection and myocardial injury in patients receiving DOX-based chemotherapy. In vitro and in vivo experiments were performed to assess ferroptosis and cardiac injury following DOX and/or…

DDR2 deficiency attenuates doxorubicin-induced cardiotoxicity by reducing cardiac dysfunction and oxidative stress in mice.

Doxorubicin (DOX) is a highly effective anthracycline chemotherapeutic drug. However, the clinical application is limited due to its dose-dependent cardiotoxicity. Discoidin domain receptor 2 (DDR2), the main collagen-binding receptor tyrosine kinase is a major player of various degrees of fibrosis or tissue restructuring in pathological conditions and unknown its role for DOX-induced heart injuries. We investigated the impacts of DDR2 deficiency from acute DOX cardiotoxicity in vivo, by utilizing DDR2-knockout (DDR2KO) and wild-type (Wt) mice. Genetic deletion of DDR2 significantly reduced DO…

Exploring the Potential Mechanism of Polyethylene Terephthalate Associated Cardiotoxicity through Network Toxicology and Molecular Docking.

As polyethylene terephthalate (PET) is one of the most widely used plastics and a pervasive environmental contaminant, growing evidence links micro/nanoplastic exposure to cardiovascular dysfunction; however, the underlying mechanisms remain unclear. Here we aimed to explore the potential cardiotoxicity of polyethylene terephthalate (PET) using an integrative computational strategy combining network toxicology, molecular docking, and molecular dynamics simulations. This fragment-based approach examined the interactions between PET monomers, terephthalic acid (TPA) and ethylene glycol (EG) with…

Overcoming hERG Cardiotoxicity in Psammaplysene D via Rational Design to Discover a Safe Antifibrotic Lead.

Liver fibrosis is a major global health challenge with limited treatment options. The marine natural product Psammaplysene D (PD) exhibits promising antifibrotic activity but suffers from significant hERG channel inhibition (>99% at 5 μM) and moderate pharmacokinetics. To address this cardiotoxicity, we employed rational optimization guided by computational toxicity prediction (ADMETlab 2.0), leading to the design and synthesis of derivatives and identification of compound C11. Patch-clamp studies confirmed C11 eliminated hERG liability (IC50 > 100 μM). C11 retained antifibrotic efficacy compa…

BHT and Its Metabolites Disrupt β2-Adrenergic Signaling: A Biotransformation-Dependent Cardiotoxicity Mechanism in Zebrafish.

Butylated hydroxytoluene (BHT) is a widely used synthetic phenolic antioxidant, ubiquitously detected in aquatic environments. While traditionally considered safe based on in vitro data, BHT adversely affects aquatic organisms at environmentally relevant concentrations. Here, we demonstrate that BHT-induced cardiotoxicity is primarily mediated by its metabolites through disruption of β2-adrenergic receptor (β2AR) signaling. In zebrafish larvae exposed to 0.01-1 μM BHT for 6 days, metabolites BHT-CHO, BHT-COOH, and BHT-Q accumulated prominently, inducing cardiac morphological and histological a…

Low-Intensity Exercise Attenuates Immune Checkpoint Inhibitor-Induced Cardiotoxicity via Regulation of Metabolism and Autophagy.

BACKGROUND: Immune checkpoint inhibitors (ICIs) are a new anti-cancer therapy that have improved survival rates in many aggressive cancers. However, while rare, a significant number of patients develop ICI-induced cardiotoxicity. Clinical manifestations are non-specific and underlying cellular mechanisms remain unknown, making diagnosis and treatment of these ICI-induced cardiac side effects difficult. Exercise has shown protective effects against chemotherapy-induced cardiotoxicity but has not been investigated in combination with ICIs. High-intensity exercise has shown greatest cardioprotect…

MTF-hERG: A Multi-Type Features Fusion-Based Framework for Predicting hERG Cardiotoxicity of Compounds.

The human ether-a-go-go-related gene (hERG) cardiac toxicity of a compound refers to its inhibitory effect on the hERG potassium channel. The hERG channel is crucial for cardiac depolarization, and its blockage can lead to prolongation of the QT interval, triggering arrhythmias and posing life-threatening risks. Therefore, assessing hERG cardiac toxicity is a vital consideration in drug development. Traditional assessment methods are complex and have low throughput, making the development of deep learning models to predict this toxicity essential for enhancing drug development efficiency, redu

Mitigation of Doxorubicin Cardiotoxicity With Synergistic miRNA Combinations Identified Using Combinatorial Genetics en masse (CombiGEM).

BACKGROUND: Cardiomyocyte loss occurs in acute and chronic cardiac injury, including cardiotoxicity due to chemotherapeutics like doxorubicin, and contributes to heart failure development. There is a pressing need for cardiac-specific therapeutics that target cardiomyocyte loss, preventing chemotherapy complications without compromising chemotherapeutic efficacy. OBJECTIVES: The authors employed massively parallel combinatorial genetic screening to find microRNA (miRNA) combinations that promote cardiomyocyte survival. METHODS: CombiGEM (combinatorial genetics en masse) screening in a cardio…

Enhanced Antitumor Efficacy and Reduced Cardiotoxicity of Ultrasound-Mediated Doxorubicin Delivery by Microbubble-Liposome Complexes.

OBJECTIVE: Doxorubicin (Dox) is standard of care for treatment of sarcomas, but cumulative dosing is often limited by cardiotoxicity. We hypothesized that ultrasound targeted microbubble (MB) cavitation (UTMC) of a liposomal doxorubicin (LDox) conjugated polymer microbubble complex (DoxLPX) would enhance tumor inhibition and limit Dox cardiotoxicity. METHODS: DoxLPX was intravenously injected in MCA205 sarcoma-bearing mice and concurrent ultrasound was delivered to the tumor site (DoxLPX + UTMC). Other mice received equivalent dosages of free Dox, LDox, or LDox + MB co-administration with UTM…

Targeting LTBP2 Reveals a Novel Anti-Cardiac Remodeling Mechanism of Finerenone Against Doxorubicin-Induced Cardiotoxicity.

Despite the clinical efficacy of doxorubicin (DOX), effective strategies to prevent its cardiotoxicity are still lacking. Finerenone, a nonsteroidal mineralocorticoid receptor antagonist (MRA), has demonstrated cardioprotective properties; however, its role and mechanism in DOX-induced cardiotoxicity (DIC) remain unclear. In this study, Finerenone treatment was found to significantly alleviate DOX-induced cardiac dysfunction and pathological remodeling in both mouse models and cultured cells. Mechanistically, molecular docking suggests that Finerenone may directly bind to Latent Transforming G…

Unmasking 5-fluorouracil cardiotoxicity: The clinical utility of dynamic myocardial CT perfusion: A case report.

We report the first documented case in Peru of 5-fluorouracil (5-FU)-induced cardiotoxicity assessed using combined coronary computed tomography angiography (CCTA) and dynamic myocardial computed tomography perfusion (CTP). A 57-year-old male with metastatic pancreatic cancer undergoing FOLFOX chemotherapy developed chest pain during the second cycle. Coronary computed tomography angiography (CCTA) revealed non-obstructive calcified plaques, ruling out significant coronary artery disease. Due to a history of active asthma, adenosine stress was contraindicated; therefore, a hyperventilation-apn…
