Modulation of hippocampal protein expression by a brain penetrant biologic TNF-α inhibitor in the 3xTg Alzheimer's disease mice.

BACKGROUND: Biologic TNF-α inhibitors (bTNFIs) can block cerebral TNF-α in Alzheimer's disease (AD) if these macromolecules can cross the blood-brain barrier (BBB). Thus, a model bTNFI, the extracellular domain of type II TNF-α receptor (TNFR), which can bind to and sequester TNF-α, was fused with a mouse transferrin receptor antibody (TfRMAb) to enable brain delivery via BBB TfR-mediated transcytosis. Previously, we found TfRMAb-TNFR to be protective in a mouse model of amyloidosis (APP/PS1) and tauopathy (PS19), and herein we investigated its effects in mice that combine both amyloidosis and

Brain reserve contributes to distinguishing preclinical Alzheimer's stages 1 and 2.

BACKGROUND: In preclinical Alzheimer's disease, it is unclear why some individuals with amyloid pathologic change are asymptomatic (stage 1), whereas others experience subjective cognitive decline (SCD, stage 2). Here, we examined the association of stage 1 vs. stage 2 with structural brain reserve in memory-related brain regions. METHODS: We tested whether the volumes of hippocampal subfields and parahippocampal regions were larger in individuals at stage 1 compared to asymptomatic amyloid-negative older adults (healthy controls, HCs). We also tested whether individuals with stage 2 would sh

Etiologic fractions in patients of hepatocellular carcinoma in India with and without a background of cirrhosis: a multi-centric study.

BACKGROUND: Hepatocellular cancer (HCC) typically arises in the background of cirrhosis. The epidemiology of HCC has changed in recent years due to availability of newer antivirals, changing life-styles and greater possibility for early detection. We undertook a multicentric national sentinel surveillance for liver cirrhosis and HCC to assess the attributable risk factors for the development of HCC, both with and without a background of cirrhosis. METHODS: Data from January 2017 till August 2022 from hospital-based records of eleven participating centers were included. Diagnosed cases of cirr

Cytokine/chemokine patterns connect host and viral characteristics with clinics during chronic hepatitis C.

BACKGROUND: In chronic hepatitis C virus (HCV) infection, liver tissue pathology and HCV genotype are important determinants of clinical and/or treatment-related outcome. Although consistent epidemiological and/or molecular-biological clues derived from different studies on single virus-host interactions are meanwhile published, the in vivo transcriptional responses and cellular pathways affected in >1 key aspects of the disease or treatment process are far from being understood. METHODS: Microarray analysis was performed in peripheral whole blood (PB) samples from 36 therapy-naïve HCV-infect

The comparative economic burden of mild, moderate, and severe fibromyalgia: results from a retrospective chart review and cross-sectional survey of working-age U.S. adults.

BACKGROUND: Patients with fibromyalgia report persistent widespread pain, fatigue, and substantial functional limitations, which may lead to high health resource use (HRU) and lost productivity. Previous analyses of the U.S. population have not examined the direct and indirect costs of fibromyalgia by severity level. OBJECTIVES: To assess (a) HRU, direct and indirect costs associated with fibromyalgia in routine clinical practice in the United States using a patient-centric approach, and (b) the relationship of fibromyalgia severity level to HRU and costs. METHODS: This study recruited a non

When does sperm impact fertilization success? The incidence of sperm-related poor fertilization after ICSI in 13 632 matched oocyte donation cycles.

STUDY QUESTION: What is the true incidence of male factor-related poor fertilization in ICSI cycles when analyzed using a matched oocyte donation model to investigate the influence of sperm-related factors on fertilization outcomes? SUMMARY ANSWER: In ICSI cycles with donor oocytes, male factor-related poor fertilization occurs in 3.1% of cases and contributes to 84.1% of all poor fertilization outcomes (≤30% fertilization rate). WHAT IS KNOWN ALREADY: Opting for oocyte donation is a complex decision for couples, involving significant financial and long-term considerations. This decision is

Integrated multiomics reveals a clinically relevant CXCL8-centric protein network in pediatric postinfectious bronchiolitis obliterans.

BACKGROUND: Pediatric postinfectious bronchiolitis obliterans (PIBO) is a long-term sequela of severe lower respiratory tract infection. However, the underlying pathogenic mechanisms within the airways remain poorly understood. METHODS: Bronchoalveolar lavage fluid (BALF) from six children with PIBO and three control children was subjected to transcriptomic, proteomics and untargeted metabolomics analyses. Integrated analysis identified key differentially expressed candidates, among which protein candidates were subsequently validated by ELISAs. RESULTS: Transcriptomic analysis revealed alte

Fibroblast growth factor 10 attenuates advanced liver fibrosis through hepatocyte fibroblast growth factor receptor 2 signalling.

BACKGROUND: Metabolic dysfunction-associated steatotic liver disease (MASLD) and advanced fibrotic stages are significant contributors to cirrhosis and liver-related mortality, yet no therapies directly target fibrosis in the later stages of the disease. Fibroblast growth factor 10 (FGF10) facilitates epithelial repair, yet its function and epithelial receptor requirements in chronic liver fibrogenesis are unclear. METHODS: We quantified hepatic FGF10 and fibroblast growth factor receptor 2 (FGFR2) expression across fibrosis stages in biopsies from patients with MASLD and mouse models. We the

Construction of lactate-myeloid-related prognostic risk model and prediction of immune-excluded microenvironment in gastric cancer via integrated scRNA-Seq and bulk RNA-Seq.

BACKGROUND: Gastric cancer (GC) progression entails profound epithelial-stromal remodeling, yet how epithelial metabolic programs are linked to myeloid-dominated immune exclusion and prognostic risk remains unclear. This study aimed to integrate single-cell RNA sequencing (scRNA-seq) and bulk transcriptomic analyses to characterize a lactate-myeloid-related risk axis in GC. METHODS: We integrated six scRNA-seq datasets comprising 116 samples and 355,091 cells spanning paratumor (PT), chronic atrophic gastritis (CAG), moderate intestinal metaplasia (MIM), severe intestinal metaplasia (SIM), ea

A dual-level Bayesian network meta-analysis of first-line immunotherapy in advanced hepatocellular carcinoma: strategy-level and regimen-level evidence.

BACKGROUND: Phase III trials of first-line immunotherapy-based combinations for advanced hepatocellular carcinoma (HCC) have yielded heterogeneous results, and head-to-head comparisons remain scarce. We evaluated mechanism-based strategy classes and individual regimens using a dual-level Bayesian network meta-analysis to provide a structured interpretation of the current evidence. METHODS: We systematically searched the literature up to December 28, 2025, and identified 15 phase III randomised controlled trials. A dual-level Bayesian network meta-analysis was performed using separate models a

Association of multimorbidity patterns with potential out-of-hospital clinical service needs: results from a nationally representative sample of older Chinese.

BACKGROUND: The rising prevalence of multimorbidity strains hospital-centric healthcare. Urgent attention is needed to understand potential out-of-hospital health service needs and inform policy for effective public health practices. METHODS: Utilizing data from the China Health and Retirement Longitudinal Study (CHARLS), we first employed latent class analysis (LCA) to identify distinct patterns of multimorbidity. Subsequently, basic characteristics associated with each identified multimorbidity pattern were investigated using logistic regression models. Third, employing logistic mixed-effec

A large, multi-center survey assessing health, social support, literacy, and self-management resources in patients with heart failure.

BACKGROUND: Most patients with heart failure (HF) have multimorbidity which may cause difficulties with self-management. Understanding the resources patients draw upon to effectively manage their health is fundamental to designing new practice models to improve outcomes in HF. We describe the rationale, conceptual framework, and implementation of a multi-center survey of HF patients, characterize differences between responders and non-responders, and summarize patient characteristics and responses to the survey constructs among responders. METHODS: This was a multi-center cross-sectional surv

Sensory and Choice Responses in MT Distinct from Motion Encoding.

The macaque middle temporal (MT) area is well known for its visual motion selectivity and relevance to motion perception, but the possibility of it also reflecting higher-level cognitive functions has largely been ignored. We tested for effects of task performance distinct from sensory encoding by manipulating subjects' temporal evidence-weighting strategy during a direction discrimination task while performing electrophysiological recordings from groups of MT neurons in rhesus macaques (one male, one female). This revealed multiple components of MT responses that were, surprisingly, not inter

Comparative genomics reveals synteny-guided remodeling of carbohydrate-foraging and intrinsic resistance in an ocular Chryseobacterium isolate, NG-CE01.

BACKGROUND: Chryseobacterium spp. are environmentally widespread bacteria increasingly recognized as opportunistic pathogens in human and veterinary medicine, yet genome-scale determinants that may support persistence in host-associated microenvironments remain poorly defined. Isolates from anatomically specialized sites such as the ocular surface are underrepresented in comparative genomic datasets. METHODS: We performed integrated phenotypic and genome-resolved characterization of Chryseobacterium sp. NG-CE01, isolated from a bovine corneal infection. A high-quality hybrid assembly was analy
