Transcriptome and Metabolome-Based Analysis Reveals the Molecular Mechanisms Underlying the Differences in Tanshinone and Salvianolic Acid Content Between Salvia miltiorrhiza Roots and Leaves.

Background/Objectives: Salvia miltiorrhiza (Danshen) is a well-known medicinal herb in traditional Chinese medicine. It produces tanshinones and salvianolic acids as key bioactive constituents in its roots, yet the molecular basis for their tissue-specific accumulation is still poorly understood. This study aims to identify candidate functional genes involved in the biosynthesis of tanshinones and salvianolic acids, and to reveal the molecular basis underlying their tissue-specific accumulation in S. miltiorrhiza. Methods: For this purpose, we compared transcriptomic and metabolomic difference

Design, synthesis, and antifungal activity of licochalcone A derivatives and mechanism of action against Phytopathogenic Fungi.

The exploration of natural products as primary scaffolds remains a valuable strategy for developing novel antifungal agents. Licochalcone A, a bioactive chalcone from licorice, served as a lead for developing novel antifungal agents. This study focused on designing and synthesizing a series of 26 derivatives and subsequently evaluating their antifungal activity against plant pathogens. In vitro antifungal screening against nine fungal strains indicated that most of the compounds demonstrated strong inhibitory activity against Valsa mali. Notably, compound 5b demonstrated significant inhibition

Combination of Isoliquiritigenin and Vancomycin Alleviates Staphylococcus aureus-Induced Bone Infection in Rats.

Staphylococcus aureus (S. aureus)-induced osteomyelitis presents therapeutic challenges due to antibiotic resistance. Isoliquiritigenin (ISL), a licorice-derived chalcone, exhibits antibacterial and anti-inflammatory properties. This study evaluated vancomycin (VAN) combined with ISL against methicillin-resistant S. aureus (MRSA)-induced implant-related osteomyelitis. A rat model was established by tibial MRSA inoculation with simultaneous Kirschner wire implantation. Four weeks postinfection, rats were divided into five groups: normal, model, VAN (50 mg/kg), ISL (100 mg/kg), and VAN + ISL. Af

Sofalcone attenuates neurodegeneration in MPTP-induced mouse model of Parkinson's disease by inhibiting oxidative stress and neuroinflammation.

BACKGROUND: Parkinson's disease (PD) is a progressive neurodegenerative disorder characterized by oxidative stress and neuroinflammation. Sofalcone (SFC), a chalcone derivative known for its antioxidative and anti-inflammatory properties, is widely used clinically as a gastric mucosa protective agent. However, its therapeutic potential in PD remains to be fully explored. In this study, we investigated the neuroprotective effects of SFC in a 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-induced PD mouse model. METHODS AND RESULTS: We found that SFC ameliorated MPTP-induced motor impairme

Phenolics as GABAA Receptor Ligands: An Updated Review.

Natural products can act as potential GABA modulators, avoiding the undesirable effects of traditional pharmacology used for the inhibition of the central nervous system such as benzodiazepines (BZD). Phenolics, especially flavonoids and phlorotannins, have been considered as modulators of the BZD-site of GABAA receptors (GABAARs), with sedative, anxiolytic or anticonvulsant effects. However, the wide chemical structural variability of flavonoids shows their potential action at more than one additional binding site on GABAARs, which may act either negatively, positively, by neutralizing GABAAR

NMR spectroscopy, excited states and relevance to problems in cell biology - transient pre-nucleation tetramerization of huntingtin and insights into Huntington's disease.

Chalcone synthase (CHS) is a pivotal enzyme in flavonoid biosynthesis involved in plant development, defense, and secondary metabolism. Xanthoceras sorbifolium (yellowhorn) is a medicinal and ornamental species with high resistance to environmental stresses, but its CHS gene family remains uncharacterized. We performed a pangenome-wide identification of CHS genes across five yellowhorn genomes (Xzs4, Xwf8, Xjg, Xg11, and Xzg2). Across the five yellowhorn genomes, 27 CHS genes were identified and classified into four core pangenes, present in all five genomes, and two dispensable genes, present

Structural studies of codeinone reductase reveal novel insights into aldo-keto reductase function in benzylisoquinoline alkaloid biosynthesis.

Benzylisoquinoline alkaloids (BIAs) are a class of specialized metabolites with a diverse range of chemical structures and physiological effects. Codeine and morphine are two closely related BIAs with particularly useful analgesic properties. The aldo-keto reductase (AKR) codeinone reductase (COR) catalyzes the final and penultimate steps in the biosynthesis of codeine and morphine, respectively, in opium poppy (Papaver somniferum). However, the structural determinants that mediate substrate recognition and catalysis are not well defined. Here, we describe the crystal structure of apo-COR dete

An uncovered XIII century icon: particular use of organic pigments and gilding techniques highlighted by analytical methods.

The restoration of a panel painting depicting a Madonna and Child listed as an unknown Tuscan artist of the nineteenth century, permitted the hidden original version, a XIII century Medieval icon to be uncovered. It is discovery provided the opportunity for an extensive in situ campaign of non-invasive analytical investigations by portable imaging and spectroscopic techniques (infrared, X-ray fluorescence and diffraction, UV-Vis absorption and emission), followed by aimed micro-destructive investigations (Raman and SEM-EDS). This approach permitted characterization of the original ground and p

Butein, a tetrahydroxychalcone, suppresses cancer-induced osteoclastogenesis through inhibition of receptor activator of nuclear factor-kappaB ligand signaling.

Osteoclastogenesis is associated with aging and various age-related inflammatory chronic diseases, including cancer. Receptor activator of nuclear factor-kappaB (NF-κB) ligand (RANKL), a member of the tumor necrosis factor superfamily, has been implicated as a major mediator of bone resorption, suggesting that agents that can suppress RANKL signaling might inhibit osteoclastogenesis, a process closely linked to bone resorption. We therefore investigated whether butein, a tetrahydroxychalcone, could inhibit RANKL signaling and suppress osteoclastogenesis induced by RANKL or tumor cells. We foun

Discovery of 7-ether substituted homoisoflavonoids as selective FXR modulators attenuating MASH via the FXR and AMPK pathways.

Farnesoid X receptor (FXR) modulation represents a promising therapeutic strategy for metabolic dysfunction-associated steatohepatitis (MASH, formerly NASH); however, the development of MASH drugs from FXR full and partial agonists remains unsucessful. In this study, we employed ring cyclization of our previously identified chalcone FXR antagonists to homoisoflavonoid skeleton and to which attached various 7-ester and ether substituents. Among them, 4l was identified as most potent compound exhibiting high binding affinity and potent cellular antagonistic activity with an IC50 value of 0.70 μM

Novel isocoumarin-pyrazole hybrids: synthesis, characterization, and investigation of their carbonic anhydrase inhibitory activities.

AIM: Selective inhibition of tumor-associated carbonic anhydrase (CA) isoforms IX and XII is a key challenge in anticancer drug discovery, as most inhibitors cause off-target inhibition of cytosolic enzymes. We addressed this by designing and synthesizing a series of isocoumarin - pyrazole hybrids from isocoumarin - chalcone scaffolds and evaluating their potential as selective CA IX/XII inhibitors. MATERIALS AND METHODS: Hybrids (5a-5o) were prepared via cyclization with substituted phenylhydrazines under reflux. Structures were confirmed by FT-IR and NMR analyses. Inhibition of hCA I, II, I

Soluble epoxide hydrolase inhibitory constituents from the Heartwood of Toxicodendron vernicifluum: isolation, kinetic characterization, molecular modeling, and quantitative analysis.

Soluble epoxide hydrolase (sEH) is a pivotal therapeutic target for inflammation management, as it regulates the hydrolysis of anti-inflammatory epoxy fatty acids (EpFAs). In this study, we investigated the sEH inhibitory potential of Toxicodendron vernicifluum (Stokes) F.A. Barkley heartwood. Bioactivity-guided fractionation of the ethanol extract identified the ethyl acetate-soluble fraction as the most active fraction (~ 60% inhibition at 50 µg/mL). Subsequent purification yielded 11 polyphenols, all of which are known compounds previously reported from this species. While these are known p
