Medial entorhinal cortex mediates learning of context-dependent interval timing behavior.

Episodic memory requires encoding the temporal structure of experience and relies on brain circuits in the medial temporal lobe, including the medial entorhinal cortex (MEC). Recent studies have identified MEC 'time cells', which fire at specific moments during interval timing tasks, collectively tiling the entire timing period. It has been hypothesized that MEC time cells could provide temporal information necessary for episodic memories, yet it remains unknown whether they display learning dynamics required for encoding different temporal contexts. To explore this, we developed a new behavio

Interactions between excitatory neurons and parvalbumin interneurons in V1 underlie neural mechanisms of amblyopia and visual stimulation treatment.

As the main cause of visual function deficits in children and adolescents worldwide, amblyopia causes serious impairment of monocular visual acuity and stereopsis. The recovery of visual functions from amblyopia beyond the critical period is slow and incomplete due to the limited plasticity of the mature cortex; notably, visual stimulation training seems to be an effective therapeutic strategy in clinical practice. However, the precise neural basis and cellular mechanisms that underlie amblyopia and visual stimulation treatment remain to be elucidated. Using monocular deprivation in juvenile m

Engineering a human P2X2 receptor with altered ligand selectivity in yeast.

P2X receptors are a family of ligand gated ion channels found in a range of eukaryotic species including humans but are not naturally present in the yeast Saccharomyces cerevisiae. We demonstrate the first recombinant expression and functional gating of the P2X2 receptor in baker's yeast. We leverage the yeast host for facile genetic screens of mutant P2X2 by performing site saturation mutagenesis at residues of interest, including SNPs implicated in deafness and at residues involved in native binding. Deep mutational analysis and rounds of genetic engineering yield mutant P2X2 F303Y A304W, wh

Aldosterone-induced salt appetite requires HSD2 neurons.

Excessive aldosterone production increases the risk of heart disease, stroke, dementia, and death. Aldosterone increases both sodium retention and sodium consumption, and increased sodium consumption may worsen end-organ damage in patients with aldosteronism. Preventing this increase could improve outcomes, but the behavioral mechanisms of aldosterone-induced sodium appetite remain unclear. In rodents, we previously identified aldosterone-sensitive neurons, which express the mineralocorticoid receptor and its prereceptor regulator, 11-β-hydroxysteroid dehydrogenase 2 (HSD2). In the present stu

Circuit mechanisms underlying sexually dimorphic outcomes of early life stress.

Stress during early life influences brain development and can affect social, motor, and emotional processes. We describe a striking sex difference in the effects of early life stress (ELS), which produces anhedonia and anxiety-like behaviors in female adolescent mice, as reported previously, but repetitive behavioral pathology and social deficits in male adolescent mice. Notably, this parallels sex differences seen in the prevalence of psychiatric symptoms: depression and anxiety disorders are more common in girls and women, whereas neurodevelopmental disorders like autism spectrum disorder an

Pramipexole Hyperactivates the External Globus Pallidus and Impairs Decision-Making in a Mouse Model of Parkinson's Disease.

In patients with Parkinson's disease (PD), dopamine replacement therapy with dopamine D2/D3 receptor agonists induces impairments in decision-making, including pathological gambling. The neurobiological mechanisms underlying these adverse effects remain elusive. Here, in a mouse model of PD, we investigated the effects of the dopamine D3 receptor (D3R)-preferring agonist pramipexole (PPX) on decision-making. PD model mice were generated using a bilateral injection of the toxin 6-hydroxydopamine into the dorsolateral striatum. Subsequent treatment with PPX increased disadvantageous choices char

Collateralizing ventral subiculum melanocortin 4 receptor circuits regulate energy balance and food motivation.

Hippocampal dysfunction is associated with major depressive disorder, a serious mental illness characterized by not only depressed mood but also appetite disturbance and dysregulated body weight. However, the underlying mechanisms by which hippocampal circuits regulate metabolic homeostasis remain incompletely understood. Here we show that collateralizing melanocortin 4 receptor (MC4R) circuits in the ventral subiculum (vSUB), one of the major output structures of the hippocampal formation, affect food motivation and energy balance. Viral-mediated cell type- and projection-specific input-outpu

Sleep-wake patterns are altered with age, Prdm13 signaling in the DMH, and diet restriction in mice.

Old animals display significant alterations in sleep-wake patterns such as increases in sleep fragmentation and sleep propensity. Here, we demonstrated that PR-domain containing protein 13 (Prdm13)+ neurons in the dorsomedial hypothalamus (DMH) are activated during sleep deprivation (SD) in young mice but not in old mice. Chemogenetic inhibition of Prdm13+ neurons in the DMH in young mice promotes increase in sleep attempts during SD, suggesting its involvement in sleep control. Furthermore, DMH-specific Prdm13-knockout (DMH-Prdm13-KO) mice recapitulated age-associated sleep alterations such a

Neuroimmune crosstalk in the gut and liver.

It has long been assumed that the nervous system exerts distinct effects on immune functions, given the large number of immune disorders that are affected by mental stress. In fact, many different immune cells have been shown to possess a wide variety of neurotransmitter receptors and receive signals from various neurotransmitters, including acetylcholine and noradrenaline. Compared with the findings on local neuroimmune interactions, limited experimental techniques have so far failed to capture a comprehensive overview of neuroimmune interactions between distant organs and the autonomic nervo

Stress-encoding neurons in the lateral habenula promote humoral immunity under eustress.

The protective effects of initial stress responses are associated with stress-induced immune enhancement. The lateral habenula (LHb) is a key brain region involved in stress regulation; however, its role in stress-induced humoral immunomodulation remains unclear. We aimed to determine whether stress-responsive neurons in the LHb mediate eustress-induced enhancement of humoral immunity and to elucidate the underlying neural mechanisms. Under conditions of eustress that promote humoral immune responses, c-Fos expression and calcium signaling in LHb neurons were significantly increased. Chemogene

Differential contributions of striatal dopaminergic pathways to visuomotor conditional learning.

RATIONALE: Midbrain dopamine (DA) neurons project principally towards the striatum, serving as key regulators of movement, motivation, and cognition. Different striatal dopaminergic (DA) pathways may regulate different aspects of cognition. OBJECTIVES: We investigated how different striatal DA pathways contribute to a known function of the striatum, visuomotor conditional learning. METHODS: Using fiber photometry, we recorded DA transients in these regions as mice learned the touchscreen Visuo-Motor Conditional Learning task. RESULTS: DA transients in all regions dynamically tracked task ev

The MAP6-NTS axis in hippocampal CA1 regulates synaptic plasticity and memory.

Microtubule-associated Protein 6 (MAP6) is critical for maintaining microtubule stability and synaptic plasticity, and its dysfunction is a key driver of cognitive impairment. However, the molecular mechanisms linking MAP6 deletion to cognitive deficits remain unclear. Here, we generated a novel Map6 knockout (KO, Map6-/-) mouse model using CRISPR/Cas9-mediated genome editing. Behavioral tests confirmed that Map6-/- mice exhibited prominent cognitive impairments, primarily in long-term memory and spatial learning. Hippocampal transcriptome profiling identified marked downregulation of neuroten

Dichotomous Regulation of Social Behaviors by Nucleus Accumbens Neurons in Female and Male Syrian Hamsters.

Social relationships, including dominance status, are powerful regulators of mental health, and social experiences impact the continued expression of these behaviors. However, very little is known about the neuronal mechanisms that regulate the positive reinforcing effects of aggression that underlie social dominance, including potential sex differences. To address this gap, we used viral-mediated gene expression in Syrian hamsters to investigate how nucleus accumbens neurons regulate behavioral adaptations caused by aggression and other social interactions in both males and females. We found

Social isolation disrupts prefrontal excitatory-inhibitory balance to impair social novelty memory in adult male mice.

Social isolation is a pervasive stressor that disrupts social cognition, yet its neural mechanisms in adulthood remain unclear. Here, we showed that four weeks of social isolation in adult male mice selectively impaired social novelty memory without affecting general sociability, object recognition, working memory, or anxiety- and depression-like behaviors. This deficit was sex-specific, as female mice remained unaffected. Mechanistically, c-Fos mapping and in vivo calcium imaging revealed that social isolation induced a maladaptive shift in prefrontal excitatory-inhibitory (E/I) balance, char

Acute activation of Gq-signaling in pancreatic islet macrophages inhibits insulin secretion through AMPK-sphingolipid axis.

Obesity-associated inflammation impairs pancreatic β-cell function, yet the mechanisms by which immune cells acutely regulate insulin secretion remain poorly defined. Here, we identify myeloid Gq signaling as an immunometabolic node linking macrophage lipid sensing to impaired insulin secretion. Using chemogenetic DREADD-mediated activation of myeloid Gq, we show that acute macrophage Gq activation impairs glucose-stimulated insulin secretion (GSIS) in vivo, whereas myeloid Gαq ablation enhances GSIS. Mechanistically, Gq activation rapidly induced AMPK phosphorylation and sphingolipid remodeli

A novel role for the infralimbic cortex in conditioned fear responding.

Freezing during Pavlovian fear conditioning is the most commonly used indicator of learned fear in rodents. Although freezing and its neural underpinnings have been widely studied, the field has largely ignored other potential indicators of learned fear. Darting, an escape-like conditioned fear response, occurs more frequently in females and is reliably predicted by a distinct behavioral phenotype: heightened shock response and decreased freezing compared to rodents that do not engage in darting. The experiments in this study sought to expand the field's understanding of conditioned fear by in

Norepinephrine neurons in the locus coeruleus and the nucleus of the solitary tract drive different stress-related behavioral outputs in mice.

Norepinephrine (NE) is one of the main neuromodulators that regulate the stress response. NE-expressing neurons in the locus coeruleus (LC) and the nucleus of the solitary tract (NTS) project to the corticotropin-releasing hormone (CRH) neurons in the hypothalamic paraventricular nucleus (PVN) to regulate the hypothalamic-pituitary-adrenal (HPA) axis response to stress. NE neurons also project to other brain regions to regulate stress outputs, including throughout the limbic system and the medial prefrontal cortex. The NE circuits, therefore, are well-positioned to regulate an integrated stres

G protein-coupled receptor 75 mediates depression-like behaviors and HPA axis hyperactivity through an excitatory arcuate-paraventricular nucleus circuit.

Metabolic dysregulation is a core pathological feature of and a major risk factor for depression. In this study, the role of G protein-coupled receptor 75 (GPR75), a brain-enriched metabolic sensor, in the pathogenesis of depression was explored and its potential role as a therapeutic target was evaluated. In chronic social defeat stress (CSDS) models, GPR75 expression was specifically upregulated in the arcuate nucleus glutamatergic (ARCVglut2) neurons of susceptible mice, a finding corroborated by analyses of postmortem brains from patients with depression. Conditional (GPR75-cKO) or local (

Targeting the KCNC3-Dependent Anterior Insular-Lateral Orbitofrontal Cortex Glutamatergic Circuit Attenuates Post-Traumatic Anxiety.

Traumatic brain injury (TBI) is a prevalent cause of secondary neuropsychiatric disorders, with post-traumatic anxiety significantly worsening long-term outcomes. However, the key neural circuits and molecular mechanisms underlying post-TBI anxiety remain poorly understood. Here, we show that TBI patients with frontal lobe injuries and anxiety exhibited increased activity and enhanced functional connectivity in the anterior insular cortex (aIC) and lateral orbitofrontal cortex (lOFC). Mechanistically, we established a mouse model of moderate frontal lobe TBI, categorizing animals into anxious
