The obese strain chicken as a model for human Hashimoto's thyroiditis.

The OS and the related Cornell C strain are interesting animal models for the study of the etiology of autoimmune thyroiditis. They are especially valuable for studying the etiologic role of thyroid abnormalities and abnormalities in the processing of iodine. Limitations of the model are the scarcity of immunological reagents for chickens, the absence of avian cloned thyroid-specific genes, minimal amounts of microsomal antibody and some differences between OS chickens and Hashimoto's patients in the types of thyroid abnormalities observed.

Development of T-lymphocyte subpopulations in the postnatal chicken oviduct.

Postnatal development of t-cell subpopulations in the oviduct was investigated in the Dekalb strain of the White Leghorn chicken by using an immunohistochemical method. T-lymphocytes first infiltrated the oviduct at 5 weeks. The number of T cells peaked at 15 weeks in the magnum, isthmus and uterus, and at 19 weeks in the infundibulum and vagina. The epithelium of the oviduct contained both granular and agranular lymphocytes. TcR1+ cells were predominant in the epithelium, whereas TcR2+ cells were more numerous than TcR1+ cells in the lamina propria. TcR3+ cells were absent from the epithelium

Sudden death of chicken embryos with hereditary riboflavin deficiency.

Riboflavin-binding protein (RfBP) mediates the deposition of riboflavin during the formation of eggs in birds. Hens of a strain of Single-Comb White Leghorn chickens, which are genetically unable to produce RfBP, lay eggs containing insufficient riboflavin to sustain embryogenesis beyond 13 or 14 d of incubation. Embryos in these eggs grow normally until the day of death, and their heart rate is normal to within an hour of death. The effects of riboflavin-deficiency first appear after d 10 of incubation when embryos become severely hypoglycemic and begin to accumulate intermediates of fatty ac…

Effect of Mycoplasma gallinarum on embryonated village chicken eggs.

The effect of Mycoplasma gallinarum isolated from village chickens on embryonated village chicken eggs was investigated. Seven-, 10-, 12- and 18-day-old embryos were inoculated with medium containing 10(6) colony forming units of M gallinarum or with uninfected medium and incubated at 37 degrees C until they hatched. There was no significant difference between the infected embryos which failed to hatch and those which hatched or between them and the group inoculated with medium only. A much higher percentage of the eggs inoculated as 18-day-old embryos hatched than of those inoculated at seven…

Molecular cloning and cellular localization of trkC in the chicken embryo.

Degenerate primers directed against conserved regions of the trk and trkB amino acid sequences were used in the polymerase chain reaction to isolate a 455 bp fragment from embryonic day 3 chicken cDNA encoding the trkC. This fragment was subsequently used to synthesize an anti-sense trkC cRNA probe which was used in a RNase protection assay of total RNA from chicken embryos. trkC mRNA was found in the E2 embryo with increasing levels later in development. In the E9 embryo highest levels were found in brain and spinal cord with intermediate levels in eye, heart, gut and muscle. Low levels were

Developmental process of Cryptosporidium in the intestine and bursa of Fabricius of chickens.

The developmental process of a Cryptosporidium isolated in Japan in the chicken intestine was investigated by scanning (SEM) and transmission electron microscopies (TEM). The parasites were detected in the ileum, cecum, colon, cloaca and bursa of Fabricius (BF). The intensity of infection tended to peak later in the BF than ileum. Trophozoites and schizonts were detected in all the portions of intestine, and were dominant in the developmental stages. Although macrogamonts were the secondary dominant stage, they were absent in the ileum and cecum at 60 hr postinoculation (PI). A few microgamont

Rapid enzyme immunoassay of chicken egg yolk IgG.

A simple and rapid enzyme immunoassay for specific antibodies in chicken egg yolk is described. As a model system, the levels of anti-Salmonella IgG in the yolk of eggs obtained from various produce retailers were compared. Polyester cloth coated with Salmonella typhimurium lipopolysaccharide was used to capture specific egg yolk antibodies, which were then detected using an anti-chicken IgG-peroxidase conjugate. This assay, requiring less than 30 min to complete, revealed considerable differences in the relative levels of anti-Salmonella IgG in the egg yolks. Anti-Salmonella IgG levels were g

Salmonella typhimurium outbreak in broiler chicken flocks in Mexico.

A Salmonella typhimurium outbreak in 1-to-2-week-old broiler flocks in Mexico is reported. Clinical signs were growth retardation, blindness, twisted necks, and lameness. Gross lesions consisted of hypopyon, panophthalmitis, hepatomegaly with necrotic foci, enlarged spleen, pericarditis, coagulated and unabsorbed yolks, and purulent arthritis. Mortality and cull rates in different flocks ranged from 1.7% to 10.6% during the first two weeks of age. All internal organs, eyes, and hock joints of diseased chickens that were cultured were Salmonella-positive. The bacteria were also isolated from th…

Megalencephaly in the epileptic chicken: a morphometric study of the adult brain.

The epileptic chicken is a genetic model of generalized epilepsy in which epilepsy is combined with megalencephaly. We have performed a morphometric study of the brains of adult epileptic hens, using heterozygous carrier hens as controls. There is no obvious disorder of cell form or of architectural arrangement in the megalencephalic brains. We have found that the enlargement of the epileptic brain is not uniform: it is most marked in the telencephalon, and is present to a lesser degree in the cerebellum, but neither the optic tectum nor the diencephalic nucleus rotundus shows a significant in

Bilirubin and heme as growth inhibitors of chicken embryos in ovo.

The increased morbidity during pregnancies complicated by hematologic or liver disease has generally been attributed to the metabolic abnormalities of the illness itself. Because tetrapyrrole concentrations are elevated in these conditions, we introduced bilirubin or heme (prepared as 10 mM solutions) into the air sac of fertilized chicken eggs to study their effect on the growth of normal chicken embryos. In 9-d eggs, the injection of 0.06 mL heme resulted in significant embryo growth inhibition as indicated by overall wt (91 +/- 3% versus control), tibia length (84 +/- 2%), tibia wt (81 +/-

Cholinergic neurons of the chicken ciliary ganglion contain somatostatin.

Somatostatin immunoreactivity was studied in the avian ciliary ganglion by immunocytochemistry and radioimmunoassay. Immunoreactivity was localized to small diameter cell bodies of neurons from embryos, newly-hatched and adult preparations. Immunostaining of ganglia with a mixture of antisera to substance P and monoclonal antibody to somatostatin indicated that a number of somatostatin-immunoreactive neurons were surrounded by substance P-immunoreactive boutons, which characteristically terminate on choroidal neurons. Staining with a mixture of antisera to choline acetyltransferase and antibod

Immunization, abrasion, and sunburn as localizing factors in chicken pox.

Three children had striking localization of varicella lesions to skin overlying a diphtheria-pertussis-tetanus toxoid (DPT) immunization injection site, a facial abrasion, and a sunburn of the trunk, respectively. Presumably, during the viremic stage of varicella, the virus passed from damaged blood vessels into the skin. A disabling myalgia in the thigh at the DPT site in one child may also have been the result of localization of the varicella virus in the muscle during the inapparent viremic stage. This suggests that an injection given during an occult viremia could trigger an unexpected rea

Pop-eye: an inherited Z-linked keratoglobus in the chicken.

A Z-linked recessive mutation in the domestic fowl is reported that results in a keratoglobus of both eyes. It does not appear to have any effect on the vision of affected individuals. The keratoglobus is not noticeable at hatching, but becomes obvious by 5-6 weeks of age. The mutant has been designated pop-eye, gene symbol pop. Initial studies suggest linkage with the late feathering (K) locus, and with the NM 7092 t (Z;1) translocation, which had previously been shown to be closely linked with Z-linked silver (S). There appear to be no other detrimental effects associated with this mutant. P

Ultrastructure of cryptosporidial life cycle in chicken host cells.

Ultrastructural studies were conducted on cryptosporidia in various stages of the life cycle, attached to the epithelial cell surface of the bursa of Fabricius, caecum and trachea in six 5 and 6-week-old chickens fed faeces containing cryptosporidial oocysts. The stages observed were sporozoites or merozoites penetrating the host cells, trophozoites, schizonts, macrogametocytes, microgametocytes and oocysts. The cryptosporidia in the chickens were morphologically and developmentally similar to those reported previously in other animals and humans. All the life cycle stages were recognised in e…

Cyto-, dendro- and fibro-architectonic studies on the chicken hypothalamus.

Based on the chicken hypothalamic analysis using the Nissl, Golgi-Cox and silver staining methods, a first attempt to make the cyto-, dendro- and fibro-architectonic atlases of the frontal planes at the same level was performed. In the preparations stained with Nissl method, the hypothalamus was subdivisible basically into the preoptic, rostral and caudal hypothalamic regions, each of which was composed of medial and lateral parts respectively. A well-defined boundaries between the preoptic and rostral hypothalamic regions was decided at the level of the rostral margin of the commissura rostra
