Spectrum of mutations in single-stranded DNA phage M13mp2 exposed to sunlight: predominance of G-to-C transversion.

Sunlight is regarded to be a cause of skin cancer, though the mechanisms underlying the causation are still unclear. The genotoxic effects of sunlight are believed to be induced by pyrimidine photoproducts produced by the action of the UV portion of sunlight. However, it is not clear whether these pyrimidine modifications are the sole sources for the mutations. In the present study, we have analyzed the mutagenic potential of sunlight on the lacZ alpha region of single-stranded DNA phage M13mp2 using an SOS-deficient recA- strain and an SOS-induced rec+ strain of Escherichia coli as hosts. Discovery of a Potent APOBEC3B Inhibitor to Augment Antitumor Immunity and Reduce Inflammatory Toxicity.

APOBEC3B (A3B), a key driver of cancer mutagenesis, has emerged as a promising therapeutic target to be ablated to enhance antitumor immunity while reducing intestinal inflammation. Starting from SMC247 (3, 5-diiodotyrosine), a kelp-derived natural product, we identified the optimized analogue SMC247-9 as a potent A3B inhibitor, with an IC50 of 50 pM and KD value of 300 pM. SMC247-9 reduced cellular A3B abundance and promoted IL-15 expression in tumor cells, thereby relieving tumor cell-mediated suppression of CD8+ T cells in an IL-15-dependent manner. In immunocompetent mice, SMC247-9 suppres

Detection of developmental toxicity of the anti-COVID-19 drug molnupiravir using gastruloid-based in vitro assays.

In pharmaceutical drug development, animal tests are traditionally required to conduct comprehensive toxicity assessments before initiating human clinical trials. However, animal tests are time-consuming and can hinder the rapid development of drugs needed to combat urgent health crises, such as the COVID-19 pandemic. Therefore, faster non-animal alternatives are critical to accelerating preclinical toxicity assessments. Molnupiravir, an antiviral medication authorized for emergency use to treat COVID-19, is an oral pro-drug that is metabolized into its active form, N4-hydroxycytidine (NHC). T…

Pharmacokinetics of Molnupiravir in Cats with Naturally Occurring Feline Infectious Peritonitis.

Antiviral drugs like EIDD-2801 (molnupiravir; MPV) have been successfully used in the treatment of feline infectious peritonitis (FIP). The previous study of the pharmacokinetics of MPV in healthy cats showed promise for its use and safety. The objective was to determine the pharmacokinetics of molnupiravir in cats with naturally occurring FIP by measuring MPV and EIDD-193 (β-D-N4-hydroxycytidine; NHC) serum levels. Blood was collected from seven cats diagnosed with naturally occurring FIP treated at 1, 2, 4, 6 and 12 h post oral MPV administration and at 12 h post pill administration 7 days l…

SARS-CoV-2 infection rebound among patients receiving antiviral agents, convalescent plasma, or no treatment: a systematic review with meta-analysis.

BACKGROUND: There is some evidence showing rebound of COVID-19 infections in patients treated with nirmatrelvir-ritonavir between 2 and 8 days following cessation of the antiviral treatment. COVID-19 rebound is not unique to patients treated with nirmatrelvir-ritonavir, but is also observed in molnupiravir recipients, in patients who did not receive any antiviral treatment and in patients who received convalescent plasma (CP). MATERIALS AND METHODS: This was a systematic review with meta-analysis of clinical trials evaluating rates of virologic and clinical rebound in COVID-19 patients receiv

Dysregulation of choline metabolism and therapeutic potential of citicoline in Huntington's disease.

Huntington's disease (HD) is associated with dysregulated choline metabolism, but the underlying mechanisms remain unclear. This study investigated the expression of key enzymes in this pathway in R6/2 HD mice and human HD postmortem brain tissues. We further explored the therapeutic potential of modulating choline metabolism for HD. Both R6/2 mice and HD patients exhibited reduced expression of glycerophosphocholine phosphodiesterase 1 (GPCPD1), a key enzyme in choline metabolism, in the striatum and cortex. The striatum of R6/2 mice also showed decreased choline and phosphorylcholine, and in

Synthesis and biological evaluation of new β-D-N4-hydroxycytidine analogs against SARS-CoV-2, influenza viruses and DENV-2.

Drug repurposing approach was applied to find a potent antiviral agent against RNA viruses such as SARS-CoV-2, influenza viruses and dengue virus with a concise strategy of small change in parent molecular structure. For this purpose, β-D-N4-hydroxycytidine (NHC, 1) with a broad spectrum of antiviral activity was chosen as the parent molecule. Among the prepared NHC analogs (8a-g, and 9) from uridine, β-D-N4-O-isobutyrylcytidine (8a) showed potent activity against SARS-CoV-2 (EC50 3.50 μM), Flu A (H1N1) (EC50 5.80 μM), Flu A (H3N2) (EC50 7.30 μM), Flu B (EC50 3.40 μM) and DENV-2 (EC50 3.95 μM)…

APOBEC3-dependent kataegis and TREX1-driven chromothripsis during telomere crisis.

Chromothripsis and kataegis are frequently observed in cancer and may arise from telomere crisis, a period of genome instability during tumorigenesis when depletion of the telomere reserve generates unstable dicentric chromosomes1-5. Here we examine the mechanism underlying chromothripsis and kataegis by using an in vitro telomere crisis model. We show that the cytoplasmic exonuclease TREX1, which promotes the resolution of dicentric chromosomes4, plays a prominent role in chromothriptic fragmentation. In the absence of TREX1, the genome alterations induced by telomere crisis primarily involve

The salvage of deoxycytidine into dCDP-diacylglycerol by macrophages and lymphocytes.

Extracellular deoxycytidine (CdR) was previously shown to be salvaged into water soluble [1] and also into lipidic [2] precursors of phospholipids in stimulated lymphocytes and in lymphoma cells [3]. In this paper we have described that non-dividing murine macrophages salvaged not only 5-3H-CdR but also tritiated thymidine (3H-TdR) mainly into the pools as nucleotides. Chlorpromazine shifted the CdR salvage into a lipidic compound of the cells which was identified as 3H-dCDP-diacylglycerol (dCDP-DAG). After 5-3H-CdR labeling the lipid/DNA ratio was eleven times higher in macrophages than in to…

Development of gemcitabine-modified multimodal miR-129 mimic as a novel therapeutic in non-small cell lung cancer.

Resistance to chemotherapy and tyrosine kinase inhibitor (TKI)-based therapy is the main reason for treatment failure in non-small cell lung cancer (NSCLC). Therefore, there is an urgent need to develop novel strategies to combat resistance. In this study, we designed and developed a novel, multi-targeted microRNA (miRNA) approach to overcome resistance to EGFR inhibitors. Specifically, the guide strand of the tumor suppressor miR-129 was modified by replacing cytidine with the nucleoside analog gemcitabine (Gem). The therapeutic potential of Gem-miR-129 was investigated in both in vitro and i…

The Innate Antiviral Factors APOBEC3G and APOBEC3H Interact with the Nucleocapsids of Human Coronaviruses in an RNA-Dependent Manner.

Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) evolution has been marked by the rapid accumulation of mutations, among which cytosine-to-uracil (C-to-U) transitions represent a major proportion of observed genomic changes. These mutations have been proposed to result from the activity of host APOBEC3 cytidine deaminases, innate immune enzymes capable of editing viral RNA. However, the molecular mechanisms underlying APOBEC3 involvement in SARS-CoV-2 biology remain poorly understood. Here, we systematically investigated physical interactions between APOBEC family members and the S…
