N-WASP has the ability to compensate for the loss of WASP in macrophage podosome formation and chemotaxis.

Wiskott-Aldrich syndrome protein (WASP) and its homologue neural-WASP (N-WASP) are nucleation promoting factors that integrate receptor signaling with actin cytoskeleton rearrangement. While hematopoietic cells express both WASP and N-WASP, WASP deficiency results in altered cell morphology, loss of podosomes and defective chemotaxis. It was determined that cells from a mouse derived monocyte/macrophage cell line and primary cells of myeloid lineage expressed approximately 15-fold higher levels of WASP relative to N-WASP. To test whether N-WASP can compensate for the loss of WASP and restore a…

Polarized hyphal growth in Candida albicans requires the Wiskott-Aldrich Syndrome protein homolog Wal1p.

The yeast-to-hypha transition is a key feature in the cell biology of the dimorphic human fungal pathogen Candida albicans. Reorganization of the actin cytoskeleton is required for this dimorphic switch in Candida. We show that C. albicans WAL1 mutants with both copies of the Wiskott-Aldrich syndrome protein (WASP) homolog deleted do not form hyphae under all inducing conditions tested. Growth of the wild-type and wal1 mutant strains was monitored by in vivo time-lapse microscopy both during yeast-like growth and under hypha-inducing conditions. Isotropic bud growth produced round wal1 cells a…

Altered distribution of Sertoli cell vimentin and increased apoptosis in cryptorchid rats.

BACKGROUND/PURPOSE: Sertoli cell cytoskeletons play an important role in the process of spermatogenesis. The authors investigated the effects of cryptorchidism in immature rats on the distribution of Sertoli cell cytoskeletons and the incidence of testis germ cell apoptosis. METHODS: Immature 3-week-old rats were made cryptorchid unilaterally, and the distribution of Sertoli cell cytoskeletons by immunohistochemistry and germ cell apoptosis by TUNEL technique were evaluated at different day intervals. RESULTS: Immunohistochemical staining from control rats showed that the distribution of vim…

Ciprofloxacin causes cytoskeletal changes and detachment of human and rat chondrocytes in vitro.

Quinolones cause damage of articular cartilage in different species by forming chelate complexes with divalent cations and inducing magnesium deficiency. Cations are important for regular function of integrins, a group of transmembrane proteins which connect extracellular matrix proteins with the intracellular cytoskeleton. We have shown that cultivation of rat chondrocytes in ciprofloxacin (CFX)-supplemented and Mg(2+)-free medium led to pronounced changes in the cytoskeleton and decreased adhesion of cells to the culture dish. In order to test whether or not these effects are species-specifi…

Differential expression of the microspike-associated protein moesin in human tissues.

The protein moesin is a member of a gene family consisting of talin, ezrin, radixin, protein 4.1., and merlin. Proteins of this family are associated to the submenbranous cytoskeleton. Using monoclonal antibody 38/87 directed against moesin in immunochemical analysis, the 78 kDa moesin protein was demonstrated in endothelial cells and in cells of carcinoma, mesothelioma and lymphoid origin. Moesin was metabolically labeled by [32P]orthophosphate and reacted with an antibody against phosphotyrosine. Moesin also contains carbohydrate residues as demonstrated by immunostainings of digoxigenin-lab…

Hereditary spherocytosis of man. Defective cytoskeletal interactions in the erythrocyte membrane.

Hereditary spherocytosis (HS) is an inherited abnormality of red cell shape and results from defective interactions amongst the components of the cytoskeleton. It is known that spectrin/actin dissociates in low ionic strength media from ghosts and cytoskeletons at a rate which is slower for HS than normal preparations. Hybridization experiments have established that this behaviour is not due to a defective spectrin or actin but resides in a spectrin-binding component of the membrane [Hill, Sawyer, Howlett & Wiley (1981) Biochem. J. 201, 259-266]. In the present study erythrocyte shells have be…

Distinct mechanical regulation of PFK1 isoforms during cytoskeletal remodeling.

All cells experience mechanical forces that shape their behavior and organization. These forces are sensed at cell-cell adhesions and transduced through the actin cytoskeleton, triggering cytoskeletal reinforcement to withstand mechanical load. Such remodeling is energy-intensive and supported by increased glycolysis, yet the mechanisms linking mechanical stress to metabolic activation remain incomplete. Here, we identify phosphofructokinase-1 (PFK1) as a bridge coupling mechanotransduction and metabolism. Shear stress activates PFK1 and promotes its association with F-actin, enhancing reinfor…

Exploring the Role of Neuropeptide PACAP in Cytoskeletal Function Using Spectroscopic Methods.

The behavior and presence of actin-regulating proteins are characteristic of various clinical diseases. Changes in these proteins significantly impact the cytoskeletal and regenerative processes underlying pathological changes. Pituitary adenylate cyclase-activating polypeptide (PACAP), a cytoprotective neuropeptide abundant in the nervous system and endocrine organs, plays a key role in neuron differentiation and migration by influencing actin. This study aims to elucidate the role of PACAP as an actin-regulating polypeptide, its effect on actin filament formation, and the underlying regulato

SUMO system - a key regulator in sarcomere organization.

Skeletal muscles constitute roughly 40% of human body mass. Muscles are specialized tissues that generate force to drive movements through ATP-driven cyclic interactions between the protein filaments, namely actin and myosin filaments. The filaments are organized in an intricate structure called the 'sarcomere', which is a fundamental contractile unit of striated skeletal and cardiac muscle, hosting a fine assembly of macromolecular protein complexes. The micrometer-sized sarcomere units are arranged in a reiterated array within myofibrils of muscle cells. The precise spatial organization of s

Anchoring fibrils in a spindle cell squamous carcinoma.

To the best of our knowledge anchoring fibrils in tumours have been reported to occur only in cutaneous cylindromas and squamous cell carcinomas of oral mucosa and larynx. In this paper we describe a difficult-to-diagnose malignant spindle cell tumour where the striking feature was the presence of large numbers of anchoring plaques and anchoring fibrils in the tumour matrix. On the basis of light microscopic and immunohistochemical studies several diagnoses were preferred, such as malignant fibrous histiocytoma, neurofibrosarcoma, malignant schwannoma, desmoplastic melanoma and spindle cell sq

Morphological alterations of salivary gland parenchyma in chronic sialadenitis.

Ten cases of chronic nonspecific sialadenitis of the parotid and submandibular glands were studied by a combination of immunohistochemistry (to detect cytoplasmic filaments), morphometry, and electron microscopy in order to assess histological modifications that might reflect on the concepts for the induction of neoplasia in these glands. Histologically, the gradual atrophy of the glandular parenchyma results in the presence of many small ductules. Although structural alterations were apparent at all levels of the secretory/excretory glandular components, and in all types of cells, the fact th

Tubulofilaments in negatively stained scrapie-infected brains: relationship to scrapie-associated fibrils.

A simple method was devised for negative-stain transmission electron microscopy of brain infected with the agent of scrapie. Brains of infected hamsters contained large masses of tubulofilamentous structures with irregular fuzzy surfaces. Brains of mice infected with Creutzfeldt-Jakob disease agent contained similar tubulofilaments in smaller numbers. The abnormal tubulofilaments resembled but were distinguished from normal microtubules. On grids soaked in sodium dodecyl sulfate the abnormal tubulofilaments were found in stages of fragmentation, an outer coat appearing t0 strip from the surfac

Actin cytoskeletal defects in immunodeficiency.

The importance of the cytoskeleton in mounting a successful immune response is evident from the wide range of defects that occur in actin-related primary immunodeficiencies (PIDs). Studies of these PIDs have revealed a pivotal role for the actin cytoskeleton in almost all stages of immune system function, from hematopoiesis and immune cell development, through to recruitment, migration, intercellular and intracellular signaling, and activation of both innate and adaptive immune responses. The major focus of this review is the immune defects that result from mutations in the Wiskott-Aldrich syn…

Colchicine inhibits macrophage motility and matrix degradation through microtubule disruption.

BACKGROUND: Atherosclerosis is a chronic inflammatory process wherein macrophages play a central role in the evolution of arterial wall plaques in extracellular matrix. Colchicine, a drug used most commonly to treat gout, reduces immune cell motility and recruitment to inflamed joints by targeting microtubules. Colchicine also reduces cardiovascular events in patients with recent acute or chronic coronary disease, but the mechanisms are incompletely understood. AIMS: To characterise the effect of low-dose colchicine on human and mouse macrophages, focusing on the microtubule cytoskeleton and …

Role of Metabolic Sensor GPR81/HCAR1 in Diabetic Podocytes: Downregulated Lipolysis Results in the Deterioration of Glomerular Filtration Barrier.

The effacement of podocyte foot processes, which form slit diaphragms, are common features of proteinuria. Exploring podocyte energy metabolism, especially under diabetic conditions, may offer insights into the pathogenesis of diabetic kidney disease. Lipid accumulation is recognized as a cause of podocyte cytoskeleton remodeling and insulin resistance. Thus, the role of the metabolic sensor G-protein-coupled receptor 81 (GPR81) was examined in the molecular pathway of lipid accumulation in podocytes under hyperglycemic conditions. It was discovered that hyperglycemia downregulated the cyclic …

Antagonistic effects of actin-specific toxins on Salmonella Typhimurium invasion into mammalian cells.

Competition between bacterial species is a major factor shaping microbial communities. In this work, we explored the hypothesis that competition between bacterial pathogens can be mediated through antagonistic effects of bacterial effector proteins on host systems, particularly the actin cytoskeleton. Using Salmonella Typhimurium invasion into cells as a model, we demonstrate that invasion is inhibited if the host actin cytoskeleton is disturbed by any of the four tested actin-specific toxins: Vibrio cholerae MARTX actin crosslinking and Rho GTPase inactivation domains (ACD and RID, respective…
