Substance abuse treatment for "hazardous users": an early intervention.

A six-session cognitive behavioral protocol has been developed for substance abusers who meet the description "hazardous users." This category includes individuals evidencing mild to moderate use of alcohol or other drugs, whose lifestyles are minimally disrupted, or who are displaying signs of problem use or abuse, but are unwilling to enter intensive treatment. The treatment model is nonconfrontational and is designed to motivate the individual to recognize the problems associated with his or her substance use and initiate treatment-seeking behavior. The intervention may be particularly usef

Social and Behavioral Factors in Sickle Cell Disease: Employment Predicts Decreased Health Care Utilization.

to determine frequency of health care encounters among people with sickle cell disease (SCD) seeking treatment for a vaso-occlusive crisis (VOC). Health care encounters are categorized by visit type (day hospital, ED visit, hospitalization), prevalence of self-reported behavioral and social factors, and any associations between behavioral and social factors and health care encounters. Ninety-five people with SCD were enrolled in a prospective descriptive study in North Carolina. Patients were interviewed concerning behavioral-social factors, and a report of health care encounters was produced,

Moving Beyond Self-Report in Characterizing Drug Addiction: Using Drug-Biased Behavior to Predict Treatment Completion and Dropout in Heroin-Primary, Medication-Maintained Opioid Use Disorder.

BACKGROUND: Drug addiction is accompanied by enhanced salience attributed to drug over nondrug cues. This bias can be objectively measured and is reliable but underutilized in informing clinical end points, where self-report measures are most commonly used, with limited success. METHODS: We investigated whether behavioral picture choice (laboratory-simulated measure of drug seeking) and verbal fluency (drug and nondrug words generated) revealed drug-biased processing in 59 individuals with opioid use disorder (iOUDs) compared with 29 healthy control (HC) individuals; assessed twice, we also i

The role of the nucleus accumbens OXR1 in cocaine-induced locomotor sensitization.

Re-exposure to drug or drug-associated cues after withdrawal can induce behavioral sensitization expression in animals or increase in the expected effect to drug in humans, which mean an enhanced drug seeking/taking motivation to trigger relapse after abstinence. The Nucleus accumbens (NAc) is known to play a key role in mediating this motivation. Recently, it has been shown that systemic administration of orexin receptor 1 (OXR1) antagonist attenuates animals' motivation behavior to take drug by self-administration paradigm, which is more effectively than orexin receptor 2 (OXR2) antagonist.

Concomitant behavioral and prefrontal cortex neuronal responses following acute and chronic methylphenidate exposure in adolescent and adult rats.

There is growing concern that the psychostimulant Methylphenidate (MPD) is being abused for cognitive enhancement and recreation by healthy adults and adolescents seeking to improve their work or academic performance. This study concomitantly recorded the behavioral and prefrontal cortex (PFC) neuronal activity in freely behaving animals exposed to acute and chronic MPD doses (0.6, 2.5, and 10.0 mg/kg MPD) in order to compare MPD effects on adult and adolescent rats. The PFC is one of the primary brain areas affected by MPD and the drug of choice for treating ADHD. Moreover, the PFC is one of

Heroin Regulates Orbitofrontal Circular RNAs.

The number of drug overdose deaths involving opioids continues to rise in the United States. Many patients with opioid use disorder (OUD) that seek treatment still experience relapse. Perseverant opioid seeking behaviors represent a major challenge to treating OUD and additional therapeutic development will require insight into opioid-induced neurobiological adaptations. In this study, we explored the regulation of a novel class of RNAs, circular RNAs (circRNAs), by the addictive opioid heroin in the rat orbitofrontal cortex (OFC), a brain region that mediates behavioral responses to rewarding

Discovery of [(3-Phenylphenyl)carbamoylamino]benzenesulfonyl Fluoride (RTICBM-303), a Cannabinoid Receptor Type 1 Allosteric Modulator that Suppresses Cue- and Drug-Primed Methamphetamine Seeking in Rats.

The development of effective pharmacotherapies for methamphetamine (METH) use disorder remains a critical challenge, prompting interest in strategies to modulate the endocannabinoid system because of its involvement in reward regulation. We hereby report the identification and characterization of a negative allosteric modulator (NAM) of the CB1 receptor, RTICBM-303 (11), which displayed good potencies in several CB1 in vitro assays, concentration dependently reduced the Emax of the agonist CP55,940, and increased [3H]CP55,940 binding. Alcohol-seeking associations with resting state functional connectivity of the amygdala.

BACKGROUND: Prolonged alcohol use is associated with neuroadaptations that may contribute to continued alcohol consumption, even in the presence of adverse consequences. Although well studied in preclinical models, the neural mechanisms of aversion-resistant alcohol-seeking in humans are understudied. PURPOSE: The goal of this study was to examine whether altered amygdala resting state functional connectivity (rsFC) is a mechanism for how lifetime drinking history and alcohol use disorder symptoms relate with objectively measured neutral and aversion-resistant alcohol-seeking in the human lab

Simultaneous cocaine and ethanol self-administration promotes a sex-dimorphic pattern in drug-seeking behaviour and alters plasma amino acid profile related to glutamate homeostasis in young adult rats.

The concurrent use of cocaine and alcohol is highly prevalent in Western countries and carries a substantial risk of relapse in recovering process. Craving, characterized as a strong desire for consuming drugs, is a core feature of cocaine and ethanol use disorders and presents a significant challenge to maintaining abstinence and preventing relapse. The primary objective of this study was to explore the behavioural patterns associated with the incubation of drug-seeking for a cocaine-ethanol combination and, secondarily, to examine the plasma amino acid profile that might be associated with t

A ventral pallidal glutamatergic aversive network encodes abstinence from and reexposure to cocaine.

Relapse to drugs of abuse can occur after long periods of abstinence. The ventral pallidum (VP) is central to drug addiction, and its glutamatergic neurons (VPGlu), whose activation drives aversion, inhibit drug seeking. However, it remains unknown whether these neurons encode the abstinence from and relapse to drugs. We show here that VPGlu projections specifically to the aversion-related lateral habenula (LHb) and ventral tegmental area gabaergic (VTAGABA) neurons show plasticity induced by abstinence from and reexposure to cocaine or cocaine cues. Both these pathways potentiate during absti

lnc15qD3 expression in the NAc shell regulates the reconsolidation of opioid reward memories.

Following memory retrieval, permanent drug memories become unstable and easily disrupted, followed by reconsolidation; this pattern holds promise for treating substance use disorder (SUD). Recent research has demonstrated that epigenetic modifications, including long noncoding RNA (lncRNA) regulation, participate in learning and memory. However, the role of lncRNAs in drug-memory reconsolidation remains unclear. Here, we found that in conditioned place preference (CPP) and self-administration (SA) mouse models of drug abuse, lnc15qD3 expression was substantially upregulated after contextual cu

Maternal ADHD Diagnoses Before and After Childbirth: A Danish Population-Based Cohort Study.

OBJECTIVE: The rates of women of reproductive age diagnosed with ADHD have increased significantly. However, little is known about how pregnancy, childbirth, and the postpartum period influence ADHD related problems, help-seeking behaviors or the timing of ADHD diagnosis. This study aimed to investigate incident ADHD diagnoses among women from 5 years before to 5 years after childbirth. METHOD: The population-based cohort study utilized Danish nationwide registers to identify mothers of all childbirths between 2010 and 2018. ADHD diagnoses, defined as ICD-10 codes (F90 or F98.8) or redemption

Differential Modulation of Glutamate Transporter-1 by Cocaine and Oxycodone and the Efficacy of MC-100093 to Reduce Reinstatement of Self-Administration.

INTRODUCTION: Despite the widespread impact of opioid use disorder, pharmacological options for treatment remain limited. Recent studies find that cocaine exposure decreases the expression of the glutamate transporter GLT-1 in the nucleus accumbens (NAc) and that treatment with the beta-lactam antibiotic ceftriaxone rescues this loss of expression and reduces cue-induced reinstatement to cocaine self-administration. The novel beta-lactam derivative MC-100093 (093) lacks antimicrobial properties but crosses the blood-brain barrier more rapidly and retains the beneficial effects of ceftriaxone f

Insular cortex subregions have distinct roles in cued heroin seeking after extinction learning and prolonged withdrawal in rats.

Evidence indicates that the anterior (aIC), but not posterior (pIC), insular cortex promotes cued reinstatement of cocaine seeking after extinction in rats. It is unknown whether these subregions also regulate heroin seeking and whether such involvement depends on prior extinction learning. To address these questions, we used baclofen and muscimol (BM) to inactivate the aIC or pIC bilaterally during a seeking test after extinction or prolonged withdrawal from heroin. Male Sprague-Dawley rats in the extinction groups underwent 10+ days of heroin self-administration, followed by 6+ days of extin

Translating preclinical models of alcohol seeking and consumption into the human laboratory using intravenous alcohol self-administration paradigms.

Preclinical models of alcohol use disorder (AUD) have advanced theoretical, mechanistic, and pharmacological study of the human condition. "Liking" and "wanting" behaviors reflect core processes underlying several models of AUD. However, the development and application of translational models of these preclinical approaches are at an incipient stage. The goal of this study was to examine how intravenous free-access and progressive-ratio, operant-response human alcohol self-administration paradigms can be used as translational human model parallels of preclinical "liking" and "wanting." Impairments in reversal learning following short access to cocaine self-administration.

BACKGROUND: Cocaine use disorder is characterized by compulsive drug-seeking that persists long into abstinence. Work using rodent models of cocaine addiction has found evidence for reversal learning deficits 21 days after non-contingent cocaine administration and 60 days after self-administration. Here we sought to determine if a deficit in reversal learning is present 3-4 weeks after cessation of cocaine self-administration, when relapse to cocaine-seeking is robust. Conversely, we hypothesized that reversal learning training would protect against relapse, similar to other forms of environme

Intentional Misuse and Abuse of Loperamide: A New Look at a Drug with "Low Abuse Potential".

BACKGROUND: Despite its opioid properties, loperamide has long been thought to have low abuse potential due to its poor absorption from the gastrointestinal tract and limited potential to cross the blood-brain barrier. A recent patient reportedly taking loperamide to avoid heroin withdrawal symptoms, at doses approximately 100 times those recommended, directed our attention to this issue. OBJECTIVES: 1) Investigate number of cases of intentional loperamide abuse and misuse reported to poison centers between 2009 and 2015; 2) Compile reports of clinical effects of loperamide abuse; and 3) Sear

Prenatal exposure to vanilla or alcohol induces crawling after these odors in the neonate rat: The role of mu and kappa opioid receptor systems.

Rat fetuses can perceive chemosensory stimuli derived from their mother's diet, and they may learn about those stimuli. In previous studies we have observed that prenatal exposure to alcohol during the last days of gestation increases the acceptance and liking of an alcohol flavor in infant and adolescent rats. While these results were not found after prenatal exposure to vanilla, cineole or anise, suggesting that the pharmacological properties of alcohol, mediated by the opioid system, underlie the effects observed with this drug. Considering that other studies report enhanced acceptance of n

Sex differences in oral oxycodone self-administration and stress-primed reinstatement in rats.

The opioid epidemic has become a severe public health problem, with approximately 130 opioid-induced deaths occurring each day in the United States. Prescription opioids are responsible for approximately 40% of these deaths. Oxycodone is one of the most commonly abused prescription opioids, but despite its prevalent misuse, the number of preclinical studies investigating oxycodone-seeking behaviors is relatively limited. Furthermore, preclinical oxycodone studies that include female subjects are even more scarce, and it is critical that future work includes both sexes. Additionally, the oral r

Exposure to environmental enrichment attenuates addiction-like behavior and alters molecular effects of heroin self-administration in rats.

Environmental factors profoundly affect the addictive potential of drugs of abuse and may also modulate the neuro-anatomical/neuro-chemical impacts of uncontrolled drug use and relapse propensity. This study examined the impact of environmental enrichment on heroin self-administration, addiction-related behaviors, and molecular processes proposed to underlie these behaviors. Male Sprague-Dawley rats in standard and enriched housing conditions intravenously self-administered similar amounts of heroin over 14 days. However, environmental enrichment attenuated progressive ratio, extinction, and r
