Computational investigation of MHD stagnation point flow of Eyring-Powell nanofluid with microorganisms over a cylinder under Cattaneo-Christov heat and mass flux.

This study investigates magnetohydrodynamic stagnation point flow with non-Fourier heat and mass transfer in an Eyring-Powell nanofluid containing gyrotactic microorganisms over an inclined porous cylindrical surface. The model simultaneously incorporates the Buongiorno nanofluid formulation, nonlinear thermal radiation, Arrhenius activation energy, Brownian motion, thermophoresis, Darcy porous medium resistance, and dual Cattaneo-Christov heat and mass flux model, thereby establishing a unified framework for analysing coupled momentum, thermal, concentration, and bioconvective transport pheno

[Molecular Mechanisms of Antitumor Effect in Targeted Alpha-radionuclide Therapy].

Targeted α-radionuclide therapy (TAT) is a systemic therapy for cancer with cancer-targeting compounds conjugated with α-emitters. Since the linear energy transfer (LET) of α-particles are high, relative biological effectiveness is higher than low LET radiation, such as X-rays and β-particles, and the damage to the normal tissues surrounding the tumor is minimal because path length of α-particle is short (a few cells). TAT has demonstrated remarkable therapeutic effects in patients and has attracted considerable attention from physicians and researchers worldwide. In particular, a study publis…

Lighting up FLASH and beyond: TLD response across four orders of magnitude in proton dose rate.

BACKGROUND: Thermoluminescent dosimeters (TLDs) are widely used for proton therapy dosimetry and remote audits. Yet their response under pulsed delivery and the ultrahigh dose rates relevant to proton FLASH remains incompletely characterized. In proton beams, apparent dose-rate effects can be masked by changes in linear energy transfer (LET), so we designed measurements to separate true dose-rate dependence from LET-driven changes in TLD sensitivity. PURPOSE: To characterize TLD-100 (LiF:Mg,Ti) detector response to synchrotron-based proton beams across in-pulse dose rates spanning approximate…

Evaluation of Complex Formation between Aminated Polyrotaxane and Proteins Using FRET.

We previously developed an intracellular carrier, an aminated polyrotaxane (amino-PRX), that recognizes and deforms in response to the shape … ot only the fluorescence of the carrier and drug themselves within cells but also Förster resonance energy transfer (FRET)-derived fluorescence between the two components was observed to examine the intracellular behavior of the complex. First, fluorescently capped PRX was prepared by capping polypseudorotaxane, consisting of α-cyclodextrin (α-CyD) and polyethyle …

Protein function evolution through the lens of conformational dynamics: A single-molecule perspective.

Understanding how proteins evolve requires moving beyond the classical sequence-structure paradigm toward a framework that incorporates conformational dynamics as a substrate of innovation. Within this framework, function emerges not only from single structures but from an ensemble encoded in the sequence. In this context, single-molecule Förster resonance energy transfer, which resolves heterogeneous and transient conformations inaccessible to ensemble methods, has shown that dynamics can tune activity, specificity, and regulation. Importantly, disordered proteins - which maintain biochemical…

Optimization of Fluorescent Nanoparticles Architecture for Efficient Brightness, FRET Efficiency and Biosensing.

Precise control over fluorophore localization within polymer nanoparticles remains a major challenge, limiting the rational optimization of brightness, Förster Resonance Energy Transfer (FRET), and biosensing performance. Here, we report architecturally controlled donor core-shell-crown fluorescent polymer nanoparticles synthesized via a controlled radical polymerization in miniemulsion through a one-pot process performed entirely in water at high solid content. The polymerizable BODIPY monomers are covalently incorporated into the polymer backbone, preventing dye leakage while enabling spatia…

A near-infrared crosstalk-free FRET fluorescent probe for highly specific detection of sulfite and viscosity as well as its multi-applications.

Sulfur dioxide (SO2) serves as a crucial signaling molecule in biological systems and a common preservative in food products, while viscosity variations reflect microenvironment alterations in pathological states and plant stress responses. To address detection challenges in these dual analytes, a near-infrared FRET probe PPTC was architected by the hybridization of a phenanthroimidazole donor and a piperazine-benzene-thiophene-cyanine acceptor strcture. The engineered architecture achieved a significant 250 nm emission separation (510/760 nm) effectively preventing spectral crosstalk, while i

Exploration of precision coregulator TR-FRET identifies diverse signatures for LXR ligands relevant to discovery of nonlipogenic ABCA1 inducers.

APOE4, the major genetic risk factor for Alzheimer's disease (AD), and ATP-binding cassette-A1 (ABCA1), required for lipidation of APOE are gene products of the liver X receptor (LXR) receptor. LXR agonists have been validated in animal models as therapeutics for AD, atherosclerosis, and many other diseases. Clinical progress has been thwarted by unwanted hepatic lipogenesis. Structurally diverse LXR ligands were profiled in coregulator TR-FRET (CRT) assays analyzing ligand-induced coactivator recruitment, coactivator selectivity, corepressor dissociation, and LXR isoform selectivity. A multip

Development of an assay to quantify tranexamic acid levels in plasma.

Dysregulations of blood clot breakdown (fibrinolysis) during vascular trauma can lead to excessive blood loss. Tranexamic acid (TXA) is an inhibitor of fibrinolysis that works by blocking the interaction between plasminogen and fibrin degradation products (FDPs) - a key step in fibrinolysis. Despite the widespread usage, there are no tests available in a clinical setting to monitor TXA levels. We developed a fluorescence resonance energy transfer (FRET)-based assay to quantify TXA concentrations in plasma by using 1) fluorescently labeled plasminogen, and 2) FDPs labeled with a fluorescence qu…

Mobile genetic elements associated with utilization of dichloromethane and methanol as energy sources in Cupriavidus metallidurans.

Cupriavidus metallidurans strain PD11 isolated from laboratory waste drainage can use C1 compounds, such as dichloromethane (DCM) and methanol, as a sole carbon and energy source. However, strain CH34 (a type-strain) cannot grow in the medium supplemented with DCM. In the present study, we aimed to unravel the genetic elements underlying the utilization of C1 compounds by strain PD11. The genome subtraction approach indicated that only strain PD11 had several genes highly homologous to those of Herminiimonas arsenicoxydans strain ULPAs1. Moreover, a series of polymerase chain reaction (PCR) to

Molecular Mechanism of a FRET Biosensor for the Cardiac Ryanodine Receptor Pathologically Leaky State.

Ca2+ leak from cardiomyocyte sarcoplasmic reticulum (SR) via hyperactive resting cardiac ryanodine receptor channels (RyR2) is pro-arrhythmic. An exogenous peptide (DPc10) binding promotes leaky RyR2 in cardiomyocytes and reports on that endogenous state. Conversely, calmodulin (CaM) binding inhibits RyR2 leak and low CaM affinity is diagnostic of leaky RyR2. These observations have led to designing a FRET biosensor for drug discovery targeting RyR2. We used FRET to clarify the molecular mechanism driving the DPc10-CaM interdependence when binding RyR2 in SR vesicles. We used donor-FKBP12.6 (D

An electronic tongue based on conjugated polymers for the discrimination and quantitative detection of tetracyclines.

A fluorescent sensor array has been developed based on conjugated polymers (CPs) having six different skeletons for the detection of tetracyclines (TCs), which are known as environmental pollutants. CPs were synthesized from confined nanoreactors in a controlled manner. The fluorescent response occurs through the fluorescence resonance energy transfer (FRET) effect. By utilizing linear discriminant analysis (LDA), effective differentiation of TCs was accomplished with a very low detection concentration (66 nM). Moreover, the sensor array exhibited a highly sensitive ability to quantitatively d…

A FRET-based assay for the quantitation of the thrombin-factor XI interaction.

BACKGROUND: Thrombin-mediated activation of FXI supports clot stability and protects the clot from fibrinolysis. This generally poor activation could be enhanced to stimulate coagulation, which might serve patients experiencing bleeding, for example due to FXI deficiency. OBJECTIVES: To establish a reliable assay that can monitor FXI-thrombin binding and is suitable for high throughput screening. METHODS: A time-resolved fluorescence resonance energy transfer assay was set up to measure binding between FXI and thrombin in a dose-dependent manner. This assay was subjected to varying concentra…

Tunable multiplexed fluorescence biosensing platform for simultaneous and selective detection of paraquat and carbendazim pesticides.

The monitoring of multiple pesticides commonly used in food is a prerequisite for public health safety. Herein, a multiplexed biosensor based on fluorescence resonance energy transfer (FRET) from multicolor upconversion nanoparticles (UCNPs)to single black phosphorus nanosheets (BPNSs) was successfully developed for simultaneous and selective detection of paraquat and carbendazim pesticides. Due to the strong π-π stacking interactions, aptamers functionalized UCNPs may adsorb on the BPNSs surface, allowing strong upconversion fluorescence quenching. In the presence of paraquat and carbendazim,…

Intracellular imaging of metmyoglobin and oxygen using new dual purpose probe EYFP-Myoglobin-mCherry.

The biological relevance of nitric oxide (NO) and reactive oxygen species (ROS) in signaling, metabolic regulation, and disease treatment has become abundantly clear. The dramatic change in NO/ROS processing that accompanies a changing oxygen landscape calls for new imaging tools that can provide cellular details about both [O2 ] and the production of reactive species. Myoglobin oxidation to the met state by NO/ROS is a known sensor with absorbance changes in the visible range. We previously employed Förster resonance energy transfer to read out the deoxygenation/oxygenation of myoglobin, crea…

Site-Specific Three-Color Labeling of α-Synuclein via Conjugation to Uniquely Reactive Cysteines during Assembly by Native Chemical Ligation.

Single-molecule fluorescence is widely used to study conformational complexity in proteins, and has proven especially valuable with intrinsically disordered proteins (IDPs). Protein studies using dual-color single-molecule Förster resonance energy transfer (smFRET) are now quite common, but many could benefit from simultaneous measurement of multiple distances through multi-color labeling. Such studies, however, have suffered from limitations in site-specific incorporation of more than two dyes per polypeptide. Here we present a fully site-specific three-color labeling scheme for α-synuclein, …

Dosimetric and microdosimetric analyses for blood exposed to reactor-derived thermal neutrons.

Thermal neutrons are found in reactor, radiotherapy, aircraft, and space environments. The purpose of this study was to characterise the dosimetry and microdosimetry of thermal neutron exposures, using three simulation codes, as a precursor to quantitative radiobiological studies using blood samples. An irradiation line was designed employing a pyrolytic graphite crystal or-alternatively-a super mirror to expose blood samples to thermal neutrons from the National Research Universal reactor to determine radiobiological parameters. The crystal was used when assessing the relative biological effe
