Knockdown of Lcn2 alleviates pyroptosis in a neonatal rat model of necrotizing enterocolitis through IL-17A signaling pathway suppression.

BACKGROUND: Necrotizing enterocolitis (NEC) is a life-threatening gastrointestinal disease primarily affecting premature infants. However, the specific regulatory molecular mechanisms focused on pyroptosis remain unclear. METHODS: Lipocalin-2 (Lcn2) was identified as a pyroptosis-associated hub gene by bioinformatics methods. Neonatal rats were fed formula milk and subjected to hypoxia (100 % N₂, 70 s) and hypothermia (4°C, 10 min). Rat intestinal epithelial cells-6 (IEC-6) and human intestinal epithelial cells-6 (HIEC-6) were stimulated with lipopolysaccharide (LPS) to mimic NEC-induced infl

Human Bone Marrow-Derived Mesenchymal Stromal Cells Reduce the Severity of Experimental Necrotizing Enterocolitis in a Concentration-Dependent Manner.

Necrotizing enterocolitis (NEC) is a devastating gut disease in preterm neonates. In NEC animal models, mesenchymal stromal cells (MSCs) administration has reduced the incidence and severity of NEC. We developed and characterized a novel mouse model of NEC to evaluate the effect of human bone marrow-derived MSCs (hBM-MSCs) in tissue regeneration and epithelial gut repair. NEC was induced in C57BL/6 mouse pups at postnatal days (PND) 3-6 by (A) gavage feeding term infant formula, (B) hypoxia/hypothermia, and (C) lipopolysaccharide. Intraperitoneal injections of PBS or two hBM-MSCs doses (0.5 ×

Peroxisome proliferator-activated receptor-γ agonist pioglitazone reduces the development of necrotizing enterocolitis in a neonatal preterm rat model.

BACKGROUND: Factors affecting innate immunity and acting as inflammatory regulators, such as the nuclear peroxisome proliferator-activated receptors (PPAR) could be crucial in the pathogenesis of necrotizing enterocolitis (NEC). We hypothesized that the PPARγ agonist pioglitazone (PIO) might be effective in preventing the development of NEC and/or reducing its severity. METHODS: We studied preterm rats in which NEC was induced using the hypoxia-hypothermia model. The treatment group (TG; n = 30) received enteral PIO (10 mg/kg/d) for 72 h and the control group (CG; n = 30) did not. Animals wer

Cyclosporine treatment improves mesenteric perfusion and attenuates necrotizing enterocolitis (NEC)-like intestinal injury in asphyxiated newborn piglets during reoxygenation.

PURPOSE: Asphyxia-related intestinal injury in neonates may present similar to necrotizing enterocolitis (NEC) and is partially associated with hypoxia-reoxygenation injury. Cyclosporine has been shown to reduce myocardial cell death following ischemia-reperfusion. We hypothesize that cyclosporine treatment may attenuate NEC-like intestinal injury in asphyxiated newborn piglets during reoxygenation. METHODS: Twenty piglets (1-4 days old) were acutely anesthetized and instrumented for continuous monitoring of systemic hemodynamics and superior mesenteric arterial (SMA) flow. After stabilizatio

Role of pyroptosis-related hub genes in necrotizing enterocolitis: a study based on bioinformatics analysis and clinical cohort validation.

OBJECTIVE: To characterize the expression patterns and clinical significance of pyroptosis-related key genes in necrotizing enterocolitis (NEC), and to identify potential biomarkers with adjunctive diagnostic value. METHODS: Pyroptosis-related candidate genes associated with NEC were screened through transcriptomic differential expression analysis, weighted gene co-expression network analysis (WGCNA), and immune infiltration analysis, followed by validation in an NEC mouse model and an LPS-induced IEC-6 cell model. Furthermore, a clinical cohort study was conducted to compare serum ADIPOQ (ad

Degree of Uncertainty in Reporting Imaging Findings for Necrotizing Enterocolitis: A Secondary Analysis from a Pilot Randomized Diagnostic Trial.

Diagnosis of necrotizing enterocolitis (NEC) relies heavily on imaging, but uncertainty in the language used in imaging reports can result in ambiguity, miscommunication, and potential diagnostic errors. To determine the degree of uncertainty in reporting imaging findings for NEC, we conducted a secondary analysis of the data from a previously completed pilot diagnostic randomized controlled trial (2019-2020). The study population comprised sixteen preterm infants with suspected NEC randomized to abdominal radiographs (AXRs) or AXR + bowel ultrasound (BUS). The level of uncertainty was determi

Deciphering the role of TNF‑α‑induced protein 8‑like 2 in the pathogenesis of necrotizing enterocolitis in neonatal rats.

Neonatal necrotizing enterocolitis (NNEC) is a disease characterized by intestinal inflammation and ischemic necrosis. Despite progress having been made during decades of research, details regarding its pathophysiology remain to be elucidated. It is known that abnormal expressions of TNF-α-induced protein 8-like 2 (TIPE2) can be observed in several diseases. However, the expression of TIPE2 in necrotizing enterocolitis (NEC) rats has not been examined before. The present study aimed to describe the expression pattern of TIPE2 and its role in NNEC pathogenesis. An NEC rat model was generated an

Intestinal Resection is More Likely to be Effective in Necrotizing Enterocolitis Extending to Colon than in Disease Limited to the Small Intestine.

BACKGROUND: The prognosis in surgical necrotizing enterocolitis (NEC) has focused on the total length of the resected bowel; the relative impact of small intestinal vs colonic resection is not well studied. OBJECTIVE: We hypothesized that intestinal resections may reduce mortality and length of hospital stay (LOS) more likely in infants who have NEC extending into the colon than in those with disease limited to the small intestine. We also investigated the relationship between gestational maturation and NEC-related mortality. METHODS: A retrospective study of 153 patients compared demographi

Mortality in micro-premature infants with necrotizing enterocolitis treated by primary laparotomy is independent of gestational age and birth weight.

Previous reports indicate that mortality in micro-premature infants with surgical necrotizing enterocolitis (NEC) is inversely proportional to gestational age and birth weight. We have observed that patterns of survival in micro-premature infants with NEC appear to be changing and may be influenced by the use of primary laparotomy (PL). Thirty-one infants <1,500 g who underwent surgery for NEC were classified into two subgroups: extremely low birth weight infants <1,000 g (ELBW, N = 17) and very low birth weight infants 1,000-1,500 g (VLBW, N = 14) and the groups were compared. Data were retro

Necrotizing enterocolitis in a premature infant as the presenting symptom of familial dysautonomia in the neonatal period: case report and review of the literature.

We present a case of a premature infant with early-onset necrotizing enterocolitis as the presenting symptom of familial dysautonomia. The diagnosis of familial dysautonomia in the neonatal period is usually rare and difficult, because many of the symptoms may be mild or nonspecific, whereas other characteristic signs and symptoms appear only later in life. The neonatologist should be aware of this rare diagnosis in the neonatal period, especially in the offspring of Ashkenazi Jews. The diagnosis is genetic. Preconception counseling to future parents belonging to populations at risk should inc
