Serum samples from 22 patients with proven typhoid fever, 60 febrile nontyphoidal patients, and 120 healthy subjects were tested for immunoglobulin A (IgA), IgG, and IgM anti-Salmonella typhi lipopolysaccharide antibodies by an enzyme-linked immunosorbent assay. The levels of all three classes of immunoglobulin anti-lipopolysaccharide were higher in typhoid patients than in controls; the test for IgM anti-lipopolysaccharide gave the best discrimination between typhoid and nontyphoidal sera. A micro enzyme-linked immunosorbent assay utilizing antigen dotted onto nitrocellulose filter discs (Dot-ELISA) was developed for the rapid diagnosis of visceral leishmaniasis. Leishmania donovani promastigotes applied to filter discs in volumes of 1 microliter were placed in 96-well microtiter plates, blocked with bovine serum albumin, then incubated with 4-fold dilutions of patient sera. After incubation with peroxidase-conjugated anti-human antibody, washing and addition of precipitable substrate, positive reactions appeared as blue dots on a white background which were easily read by eye.

A sensitive and technically simple enzyme-linked immunosorbent assay (ELISA) was developed to demonstrate circulating IgA- and IgG-class antibodies to gliadin, a component of wheat gluten. The ELISA was used to measure circulating antigen and antibody in four baboons of which three were treated. The circulating antigen appeared earlier after infection than the antibody which eventually, however, reached a higher level. Both antigen and antibody levels increased slightly after treatment and thereafter declined to reach background levels eight weeks later. It is concluded that the ELISA has a potentially useful role in detecting both antibody and circulating antigen and that it may be successfully used in evaluating the efficacy of schistosomicides.

Antigens of plaque-purified Rickettsia tsutsugamushi strains Gilliam, Karp, and Kato were separated by sodium dodecyl sulfate-polyacrylamide gel electrophoresis and were analyzed by an enzyme-linked immunosorbent assay. The sera of 25 patients with bacteriological diagnosis of typhoid fever were analyzed to identify and measure the antibodies to S. typhi somatic antigens using the enzyme-linked immunosorbent assay technique (ELISA). The results were positive in all the serologic tests, with titers varying from 1:600 to 1:2500. From sera of highly parasitised mice and dogs with Babesia rodhaini, B. galagolata and B. canis ectoantigens were isolated by column-chromatography and tested in the ELISA for their serological properties. Hyperimmunsera against the three Babesia species were prepared in rabbits, mice and dogs. Serologic cross-reactions occurred between the investigated Babesia species although distinct titer differences could be observed between B. canis on one hand and B. rodhaini and B. galagolata on the other hand. The ELISA appears to be suitable for serological field surveys of babesiosis.
