[Genotype and phenotype characteristics of DFNB67 hearing loss patients with LHFPL5 variants].

The current study aimed to analyze the genotype and phenotype characteristics of families with hearing loss caused by LHFPL5 gene variants. The clinical and molecular genetic data of two Han Chinese families with heredity hearing loss who visited Tianjin Medical University General Hospital and Chinese PLA General Hospital from May to November 2023 were retrospectively analyzed. The probands from two families are a 35-year-old female deaf-mute patient and a 5-year-old boy with hearing loss. Whole exome sequencing was used to identified candidate pathogenic genes in probands, and variant loci we

PLA2G6-associated neurodegeneration presenting as a complicated form of hereditary spastic paraplegia.

PLA2G6-associated neurodegeneration (PLAN) comprises heterogeneous neurodegenerative disorders, including infantile neuroaxonal dystrophy, neurodegeneration with brain iron accumulation 2B, and Parkinson disease 14 (PARK14). In addition, very recently, PLA2G6 mutations have been reported to represent a phenotype of hereditary spastic paraplegia (HSP). In this study, we screened 383 HSP families to clarify the frequency of PLA2G6 mutations in the Japan Spastic Paraplegia Research Consortium, and revealed the clinical characteristics of HSP with PLA2G6 mutations. We found three families with com

Does early family risk and current quality of care predict indiscriminate social behavior in institutionalized Portuguese children?

The current study extends research on the effects of institutionalization, most notably by evaluating the influence of proximal relationship processes over and above prenatal and pre-institutional family experiences. By focusing on current quality of institutional care and the child's early family background, it examines the influence of variations in the institutionalization experience on displays of indiscriminate social behavior, after taking into account potentially confounding pre-admission experiences. Seventy-four Portuguese children (11-30 months) placed in 17 residential institutions

Benign familial infantile convulsions: phenotypic variability in a family.

Benign familial infantile convulsion is an autosomal dominant epilepsy syndrome characterized by seizures starting from 3 to 12 months and a favorable outcome. We present a Turkish family with benign familial infantile convulsions and report the clinical variability associated with this syndrome in three generations. All 11 affected members had benign infantile seizures, which were primarily generalized in all but one patient, who had partial seizures with secondary generalization. The seizures started within the first year and were accompanied by normal neurologic development and a good respo

Neuronal ubiquitinated intranuclear inclusions in familial and non-familial frontotemporal dementia of the motor neuron disease type associated with amyotrophic lateral sclerosis.

Ubiquitinated cytoplasmic inclusions (Ub-CIs) in superficial frontal cortex and dentate gyrus neurons are the hallmark of frontotemporal degeneration of the motor neuron disease-type (FTD-MND-type). To date, 2 reports have described intranuclear ubiquitinated inclusions (Ub-INIs) in 9 cases of familial FTD-MND-type (without clinical or pathologic motor neuron disease, MND). In the current study we found an additional 11 cases with Ub-INIs. We have identified for the first time among these cases 2 with a negative family history and 3 that have concomitant amyotrophic lateral sclerosis (ALS). Th

Levodopa-responsive dystonia. GTP cyclohydrolase I or parkin mutations?

Autosomal dominant DOPA-responsive dystonia (DRD) is usually caused by mutation in the gene encoding guanosine triphosphate-cyclohydrolase I (GTPCH I). We studied 22 families with a phenotype of levodopa-responsive dystonia by sequencing the six coding exons, the 5'-untranslated region and the exon-intron boundaries of the GTPCH I gene. Eleven heterozygous mutations were identified, including five missense mutations, one splice site mutation, two small deletions and two nonsense mutations, in 12 families that included 27 patients and 13 asymptomatic carriers. Six mutations were new and five ha

What Goes in the Bag: Emergency Time and the Ethics of Retrospective Proof.

A "go-bag" assumes an emergency with a beginning, a peak, and an end. This editorial asks what a family needs to keep for the years after that ending - not to survive the emergency, but to survive its administrative afterlife. Drawing on a decade of research with families living with congenital Zika syndrome in Northeast Brazil, and on Alagoas's compensation law, we examine how states convert the diagnostic limits of an emergency into an evidentiary burden borne by individual families. A decade after exposure, families are asked to reconstruct causation from medical fragments never designed to

Mutations of the LMNA gene can mimic autosomal dominant proximal spinal muscular atrophy.

The molecular basis of autosomal dominant spinal muscular atrophy (AD-SMA) is largely unknown. Because the phenotypic spectrum of diseases caused by LMNA mutations is extremely broad and includes myopathies, neuropathies, and cardiomyopathies designated as class 1 laminopathies, we sequenced the LMNA gene in index patients with the clinical picture of proximal SMA, who had a family history suggestive of autosomal dominant inheritance. Among the 19 families investigated, two showed pathogenic mutations of the LMNA gene, resulting in the diagnosis of a class 1 laminopathy in about 10% of our ser

Peering into proteolysis with presenilins.

Presenilins were originally identified because they cause a neurodegenerative disease, but the impact of these proteins on the neuron appears to extend beyond neurodegeneration and into fundamental concepts of cell biology and development. Presenilins were identified as a result of the steady march of molecular genetics which first lead to identification of loci on chromosomes 1 and 14 that segregated with the phenotype of early onset familial Alzheimer Disease (AD), and then to discovery of the genes coding for PS1 and PS2.

Chromosome 12 and late-onset Alzheimer's disease.

Alzheimer's disease is a complex neurodegenerative disorder, characterized by cognitive decline and distinctive neuropathology. Using large extended families with multiple affected, we found that three markers on chromosome 12 were linked with late-onset Alzheimer's disease. These markers were downstream from the gene for alpha-2 macroglobulin. It is likely that multiple genes will be identified either as risk factors or as causative agents for late-onset Alzheimer's disease.

Psychosomatic aspects of Menière's disease.

The author presents a study of the personality structure, youth development and the role of interhuman conflicts in the work situation and the family, in the causation and course of Menière's disease, the somatopsychic reactions of the patients to the illness, and the principles of a form of psychotherapy which was combined with the regular E.N.T. check-ups and treatment. The results confirm and extend previous work by other researchers. A testable hypothesis about the specific psychosomatic etiology of Menière's disease is formulated and presented.

Communicating Genetic Information in Families with Inherited Late-Onset Neurodegenerative Diseases: A Scoping Review.

Communicating genetic information within families living with inherited genetic conditions (IGCs), especially incurable and progressive late-onset neurodegenerative diseases (LONDs), presents significant challenges. To date, no literature review has specifically addressed this issue in families with LONDs. To fill this gap, a scoping review was conducted following PRISMA and JBI guidelines. Four databases (Scopus, Web of Science, PubMed, and PsycInfo) were searched, resulting in 31 articles for analysis. Six categories were identified: (i) from whom to whom: communication transmitters and rece

Variants in the ZNF469 gene in families with Brittle cornea syndrome and keratoconus.

BACKGROUND: Brittle cornea syndrome 1 (BCS1) is a rare autosomal recessive disorder characterized by corneal and sclera thinning and fragility that is caused by zinc finger protein 469 (ZNF469) gene mutation. Keratoconus is another disease related to corneal thinning. Several reports have linked ZNF469 variants and keratoconus. We recruited a four-generation BCS1 family and two keratoconus families to explore pathogenic ZNF469 variants. METHODS: This study included 11 members from a family with BCS1, 2 families with keratoconus, 368 sporadic keratoconus patients and 325 unrelated healthy cont

The Role of Food in the Family Relationships of Adolescents With Anorexia Nervosa and Bulimia in Northeastern Brazil: A Qualitative Study Using Photo Elicitation.

Family components can play roles both as protective factors and maintenance mechanisms of eating disorders. We aimed to investigate the role of food in the family relationships of adolescents with anorexia nervosa and bulimia in northeastern Brazil. Using photo elicitation, a visual narrative method that gives insight into the participants' perspective through photograph, we conducted semi-structured interviews with 26 people: four teenage girls with anorexia, four with bulimia, eight mothers, four fathers, five grandmothers, and one sister. Data were analyzed using the principles of Interpret

Novel mutations in the EDAR gene in two Pakistani consanguineous families with autosomal recessive hypohidrotic ectodermal dysplasia.

BACKGROUND: Hypohidrotic ectodermal dysplasia (HED) is a human heritable disorder characterized by sparse hair, a lack of sweat glands and malformation of teeth. There are X-linked, autosomal recessive and autosomal dominant forms of this disorder. Mutations in the EDA gene cause X-linked HED and mutations in either the EDAR or the EDARADD genes cause autosomal forms of HED. OBJECTIVES: To identify pathogenic mutations in two consanguineous Pakistani families (A and B) with 11 affected individuals demonstrating the autosomal recessive form of HED. METHODS: Genotyping of 17 members of the two

Effects of Hydration on Transthyretin Conformational Dynamics and Oligomerization.

Transthyretin (TTR) is a 56 kDa tetrameric protein complex that transports thyroxine and retinol but can misfold, causing amyloid diseases, such as senile systemic amyloidosis, familial amyloid cardiomyopathy, and familial amyloid polyneuropathy. Previous studies have found that TTR aggregation is initiated when tetramers disassemble into monomers, dimers, and trimers, which misfold and assemble into heterogeneous oligomers. These oligomers are thought to be cytotoxic, yet their formation and composition remain poorly understood. To investigate monomer misfolding, ion mobility-mass spectrometr
