Selection of a potent inhibitor of trihydroxynaphthalene reductase by sorting disease control data.

Compounds that control rice blast, but not other crop diseases, were selected for testing as inhibitors of trihydroxynaphthalene reductase of the fungal melanin biosynthetic pathway. A potent inhibitor of the enzyme (2) (Ki = 25 nM) was identified. An X-ray structure of the enzyme-NADPH-2 complex was determined at 2.1 A resolution.

Specialized pro-resolving mediators regulate in vitro human airway epithelial resistance to Aspergillus fumigatus.

Cystic fibrosis (CF) is caused by mutations in the CFTR gene resulting in alterations in the immune functions of the airway epithelium, including reduced biosynthesis of the specialized pro-resolving lipid mediators (SPMs). Aspergillus fumigatus is the most isolated filamentous fungus in patients with CF. This study explore whether specialized pro-resolving lipid mediators (SPMs) regulate the barrier function of airway epithelium in vitro during colonization by A. fumigatus. We have shown that mature A. fumigatus hyphae disrupt tight junctions in primary cultures of nasal epithelium from CF an

Implications of PM2.5 chemical composition in modulating microbial community dynamics during spring in Seoul.

Particulate matter with an aerodynamic diameter of 2.5 μm or less (PM2.5) harbors a diverse microbial community. To assess the ecological dynamics and potential health risks associated with airborne microorganisms, it is crucial to understand the factors influencing microbial communities within PM2.5. This study investigated the influence of abiotic parameters, including air pollutants, PM2.5 chemical composition (water-soluble ions and organics), and meteorological variables, on microbial communities in PM2.5 samples collected in Seoul during the spring season. Results revealed a significant

Indoor microbial communities: Influence on asthma severity in atopic and nonatopic children.

BACKGROUND: Allergic and nonallergic asthma severity in children can be affected by microbial exposures. OBJECTIVE: We sought to examine associations between exposures to household microbes and childhood asthma severity stratified by atopic status. METHODS: Participants (n = 196) were selected from a cohort of asthmatic children in Connecticut and Massachusetts. Children were grouped according to asthma severity (mild with no or minimal symptoms and medication or moderate to severe persistent) and atopic status (determined by serum IgE levels). Microbial community structure and concentration

IgE-mediated and T cell-mediated autoimmunity against manganese superoxide dismutase in atopic dermatitis.

BACKGROUND: Autoreactivity of patients with atopic dermatitis (AD) to human proteins has been postulated as a decisive pathogenetic factor for AD. OBJECTIVE: In this study, it was investigated whether the stress-inducible enzyme manganese superoxide dismutase (MnSOD) of human and fungal origin might act as an autoallergen in atopic dermatitis. METHODS: Patients with AD (n = 69; mean SCORAD [SCORing Atopic Dermatitis], 27) and other inflammatory skin diseases as well as with inhalant allergies were investigated. The presence of specific IgE against recombinant MnSOD of fungal and human origin

Targeting adhesion in fungal pathogen Candida albicans.

Fungal infections with increasing resistance to conventional therapies are a growing concern. Candida albicans is a major opportunistic yeast responsible for mucosal and invasive infections. Targeting the initial step of the infection process (i.e., C. albicans adhesion to the host cell) is a promising strategy. A wide variety of molecules can interfere with adhesion processes via an assortment of mechanisms. Herein, we focus on how small molecules disrupt biosynthesis of fungal cell wall components and membrane structure, prevent the localization of GPI-anchor proteins, inhibit production of

Role of FKS Gene in the Susceptibility of Pathogenic Fungi to Echinocandins.

Echinocandins are antifungal agents that specifically inhibit the biosynthesis of 1,3-β-D-glucan, a major structural component of fungal cell walls. Echinocandins are recommended as first-line or alternative/salvage therapy for candidiasis and aspergillosis in antifungal guidelines of various countries. Resistance to echinocandins has been reported in recent years. The mechanism of echinocandin resistance involves amino acid substitutions in hot spot regions of the FKS gene product, the catalytic subunit of 1,3-β-D-glucan synthase. This resistance mechanism contributes to not only acquired res

Common Child and Adolescent Cutaneous Infestations and Fungal Infections.

Cutaneous infections and infestations are common among children and adolescents. Ectoparasitic infestations affect individuals across the globe. Head lice, body lice, scabies, and infestations with bed bugs are seen in individuals who reside in both resource poor areas and in developed countries. Superficial cutaneous and mucosal candida infections occur throughout the life cycle. Dermatophyte infections of keratin-containing skin and skin structures result in tinea capitis (scalp), tinea corporis (body), tinea pedis (foot), and tinea unguium (nails). Less frequent endemic fungal infections su

Development of a groundwater fungal strain as a tool for toxicity assessment.

Despite the significant ecological role played by fungi in groundwater, their value as indicators of groundwater pollution is not known, as fungi are notoriously challenging to assess for toxicity because of their varied morphological components, in particular their filamentous structures. Hence they are rarely used in environmental risk assessments and monitoring programs for aquatic systems. A rapid, low cost, 1-step static toxicity test targeting the hyphal components of fungi was developed that subsequently demonstrated the sensitivity of a ubiquitous and dominant groundwater fungal strain

[Serious bacterial and fungal infections in intravenous drug addicts].

Invasive infections caused by bacteria and fungi are common complications of intravenous drug abuse. Various vital organs and structures may be affected, e.g. the cardiac valves, the larger arteries, the bones, the joints and the central nervous system. However, due to the high frequency of low-virulent microbes of skin and oral origin, the clinical picture may be atypical with subacute course and few focal signs and symptoms. The complexity of this problem is illustrated by eight cases of serious bacterial and fungal infections recently diagnosed at our hospitals. All patients were HIV negati

[Residual histoplasmosis of the lung and bronchopulmonary lymph nodes in autopsies carried out in Uberaba, MG].

Lungs and broncho-pulmonary lymph nodes were studied from 127 sequential adult autopsies done in Uberaba, MG, Brazil through the years 1992 and 1993. Calcified lung nodules and/or calcified lymph nodes were found in 39 (30.7%) cases. For 27 (69.2%) out of those 39 autopsies, fungi morphologically compatible with Histoplasma capsulatum were found within those calcified structures. Fite-Faraco stain for acid-fast bacilli was negative for all cases. It is concluded that the majority of calcified pulmonary nodes and calcified regional lymph nodes found at autopsies in Uberaba are related to infect

Pneumocystis carinii choroidopathy. A new clinical entity.

A 43-year-old black woman with acquired immunodeficiency syndrome developed bilateral multifocal choroidopathy characterized by slowly enlarging round to oval lesions in the posterior pole and midperiphery. Systemic evaluation revealed no evidence of mycobacterial, fungal, or spirochetal disease. Fluorescein angiography of the lesions showed early hypofluorescence with late staining of the lesions, which appeared deep to the retinal circulation. There was no evidence of retinal involvement. Over a 4-month period of observation, the lesions appeared to enlarge slowly, with no evidence of vitreo

Twenty-five years' experience treating cerebro-rhino-orbital mucormycosis.

Mucormycosis is an acute fungal disease with high mortality exhibiting craniofacial, pulmonary, cutaneous, gastrointestinal and disseminated forms. Cerebro-rhino-orbital (CRO) mucormycosis is the type that most frequently involves the structures of the head and neck. While usually non-pathogenic, these saprophytic organisms can be particularly lethal in chronically debilitated patients. Between the years 1957 and 1982, 18 cases of CRO mucormycosis were seen at the Medical College of Virginia Hospital and the McGuire VA Medical Center. Four illustrative cases of CRO mucormycosis are presented t

Differences in rhizosphere microbial communities between Fusarium wilt-resistant and susceptible watermelon cultivars.

To elucidate genotype-associated differences in rhizosphere microbial community assembly, this study compared the microbiomes of three Fusarium wilt-resistant and three susceptible watermelon cultivars using amplicon sequencing. Results revealed distinct bacterial and fungal community structures between the two groups. Notably, resistant cultivars harbored a higher number of unique operational taxonomic units and displayed greater fungal richness compared to their susceptible counterparts. Beyond taxonomic composition, co-occurrence network analysis demonstrated that the fungal community withi

Bioactive potential of Eugenia luschnathiana essential oil and extract: antifungal activity against Candida species isolated from oncological patients.

Immunosuppressed individuals, including those undergoing cancer treatment, are more vulnerable to fungal infections, such as oral candidiasis, impacting their quality of life. Given the limitations of current therapies, the discovery of new antifungal agents, including those of natural origin, is crucial for the proper managing of these infections. We investigated the phytochemical profile and antifungal activity of both the essential oil and crude ethanolic extract (CEE) obtained from Eugenia luschnathiana against reference strains and clinical isolates of Candida from oncology patients. Toxi
