Appearance and distribution of dendritic cells and macrophages in dental pulp during early postnatal morphogenesis of mouse mandibular first molars.

Dendritic cells and macrophages were examined in dental pulp during the postnatal development of mouse mandibular first molars, by immuno- and enzyme histochemistry. F4/80 antibody against dendritic cells and macrophages demonstrated labeled cells predominantly in and around the odontoblastic layer during tooth development from postnatal day 0 (PN0) to PN5. Labeling with Mac-1, Mac-2, and MOMA-2 antibodies against macrophages showed varied distribution patterns. Mac-1-positive cells were not detected in the dental pulp. Mac-2-positive cells appeared in the dental pulp at PN0, but not in or aro

A 3D pooled PBMC-organoid co-culture platform for profiling immune susceptibility and PD-1 blockade response in gastric cancer.

BACKGROUND: Peritoneal metastatic gastric cancer (PMGC) is associated with a dismal prognosis and limited benefit from immune checkpoint inhibitors (ICIs). Although PD-1/PD-L1 blockade has improved outcomes in selected patients, therapeutic responses remain highly heterogeneous, even among PD-L1-high tumors. The lack of preclinical models that functionally recapitulate tumor-immune interactions in a three-dimensional (3D) context has hindered the investigation of immune susceptibility and resistance mechanisms in PMGC. METHODS: We established a standardized allogeneic 3D co-culture platform i

Biosilica: Molecular Biology, Biochemistry and Function in Demosponges as well as its Applied Aspects for Tissue Engineering.

Biomineralization, biosilicification in particular (i.e. the formation of biogenic silica, SiO(2)), has become an exciting source of inspiration for the development of novel bionic approaches following 'nature as model'. Siliceous sponges are unique among silica-forming organisms in their ability to catalyze silica formation using a specific enzyme termed silicatein. In this study, we review the present state of knowledge on silicatein-mediated 'biosilica' formation in marine demosponges, the involvement of further molecules in silica metabolism and their potential applications in nano-biotech

α-Galactosyl residues on Trypanosoma cruzi trypomastigote glycoconjugates contribute to host-cell invasion via Galectin-3 and Annexin A2-S100A10 complex.

Linear, nonreducing terminal a-galactopyranosyl (a-Gal) epitopes are displayed on the major surface glycosylphosphatidylinositol (GPI)-anchored glycoconjugates of infective Trypanosoma cruzi trypomastigotes, the agent of Chagas disease (CD). These immunodominant epitopes elicit abundant (3-5% of circulating IgG), trypanolytic CD anti-a-Gal antibodies, yet their role on the parasite surface remains elusive. Here, affinity purification of macrophage extracts on the immobilized Galα1-3Galβ1-4GlcNAcβ (Galili's aGal epitope) identified Galectin-3 (Gal-3) and the annexin A2-S100A10 heterotetramer (A

Cathepsin S Deficiency Prevents Chronic Stress-Related Renal Remodeling and Dysfunction in a Mouse 5/6 Nephrectomy Injury Model.

Chronic psychological stress exacerbates chronic kidney disease (CKD). Given that lysosome cathepsins participate in human pathobiology, we investigated the roles of cathepsin S (CatS) in chronic stress-related renal damage and dysfunction in mice subjected to a 5/6 nephrectomy surgery. A mouse model combining 5/6 nephrectomy (5/6Nx) surgery and chronic restrain stress (CRS) was applied to wild-type (CatS+/+) and CatS knockout (CatS-/-) mice. The CRS exacerbated the 5/6Nx-induced renal injury, blood-pressure elevation, glomerulosclerosis, fibrosis, podocyte damage, and inflammatory macrophage

Pathophysiology and emerging biomarkers of cardiovascular-renal-hepato-metabolic syndrome.

Cardiovascular-Renal-Hepatic-Metabolic (CRHM) syndrome characterizes a complex, interrelated disease framework that encompasses cardiovascular disease, chronic kidney disease (CKD), metabolic dysfunction-associated steatotic liver disease, and metabolic disorders such as obesity, type 2 diabetes mellitus, dyslipidemia, and hypertension. The syndrome extends the concept of cardiovascular-Kidney-Metabolic syndrome by incorporating the liver's pivotal role in systemic metabolic dysfunction. This syndrome progresses through a cycle of chronic inflammation, insulin resistance, oxidative stress, and

Diagnostic and Prognostic Utility of the Extracellular Vesicles Subpopulations Present in Pleural Effusion.

Extracellular vesicles (EVs), comprising exosomes, microvesicles, and apoptotic bodies, are released by all cells into the extracellular matrix and body fluids, where they play important roles in intercellular communication and matrix remodeling in various pathological conditions. Malignant pleural mesothelioma (MPM) is a primary tumor of mesothelial origin, predominantly related to asbestos exposure. The detection of MPM at an early stage and distinguishing it from benign conditions and metastatic adenocarcinomas (AD) is sometimes challenging. Pleural effusion is often the first available bio

Altered Glycosylation Contributes to Placental Dysfunction Upon Early Disruption of the NK Cell-DC Dynamics.

Immune cells [e. g., dendritic cells (DC) and natural killer (NK) cells] are critical players during the pre-placentation stage for successful mammalian pregnancy. Proper placental and fetal development relies on balanced DC-NK cell interactions regulating immune cell homing, maternal vascular expansion, and trophoblast functions. Previously, we showed that in vivo disruption of the uterine NK cell-DC balance interferes with the decidualization process, with subsequent impact on placental and fetal development leading to fetal growth restriction. Glycans are essential determinants of reproduct

Hyaline globules and papillary fragments in cytologic smears from two intra-abdominal tumors (ovarian and hepatic) in female patients: A diagnostic pitfall with histologic correlation.

Hyaline globules and papillary fragments in cytologic samples from two intra-abdominal tumors in young females are presented including the cytological features and the correlation with the histopathologic and immunohistochemical findings. In the first case a cytologic study from an ovarian mass showed papillary structures and isolated tumor cells with epithelioid morphology, irregular reniform-like nuclear contour, pale or vacuolated cytoplasm, abundant hyaline globules and occasional glomeruloid structures resembling Schiller-Duval bodies. Yolk sac tumor (YST) was the diagnosis on the histolo

IL-34 and macrophage colony-stimulating factor are overexpressed in hepatitis C virus fibrosis and induce profibrotic macrophages that promote collagen synthesis by hepatic stellate cells.

UNLABELLED: Chronic hepatitis C virus (HCV) infection is characterized by progressive hepatic fibrosis, a process dependent on monocyte recruitment and accumulation into the liver. The mediators expressed in chronically injured liver that control the differentiation of human monocytes into profibrotic macrophages (Mφ) remain poorly defined. We report that chronically HCV-infected patients with high fibrosis stages have higher serum levels of macrophage colony-stimulating factor (M-CSF) and interleukin (IL)-34 than HCV-infected patients with lower fibrosis stages and healthy subjects. New Cardiovascular Risk Biomarkers in Rheumatoid Arthritis: Implications and Clinical Utility-A Narrative Review.

Rheumatoid arthritis (RA) is a chronic autoimmune disease that not only causes joint inflammation but also significantly increases the risk of cardiovascular disease (CVD), leading to a higher morbidity and mortality. RA patients face an accelerated progression of atherosclerosis, attributed to both traditional cardiovascular risk factors and systemic inflammation. This review focuses on emerging biomarkers for cardiovascular risk assessment in RA, aiming to enhance early detection and treatment strategies. Specifically, we examine the roles of interleukin-32 (IL-32), Dickkopf-1 (DKK-1), galec

Genetic alterations in papillary thyroid cancer: clinicopathological correlations and diagnostic implications.

INTRODUCTION: Papillary thyroid carcinoma (PTC) is the most common thyroid malignancy, encompassing distinct histological variants and a wide spectrum of clinical behaviors. Advances in molecular diagnostics have identified key genetic alterations - particularly BRAFV600E, RAS mutations, RET/PTC fusions, and TERT promoter mutations - that are strongly linked to tumor aggressiveness and prognosis. This study aimed to determine the prevalence of these alterations and evaluate their clinicopathological significance in a Saudi Arabian patient cohort. MATERIALS AND METHODS: A retrospective analysi

Sulfur dioxide inhibits mast cell degranulation by sulphenylation of galectin-9 at cysteine 74.

OBJECTIVES: Mast cell (MC) degranulation is a key process in allergic reactions and inflammatory responses. Aspartate aminotransferase 1 (AAT1)-derived endogenous sulfur dioxide (SO2) is an important regulator of MC function. However, the mechanism underlying its role in MC degranulation remains unclear. This study aimed to investigate the mechanism by which endogenous SO2 controlled MC degranulation. METHODS: HMC-1 and Rat basophilic leukemia cell MC line (RBL-2H3) were used in the cell experiments. SO2 content was detected by in situ fluorescent probe. MC degranulation represented by the re

Candida albicans Promotes Oral Cancer via IL-17A/IL-17RA-Macrophage Axis.

The association between Candida albicans (C. albicans) and oral cancer (OC) has been noticed for a long time, but the mechanisms for C. albicans promoting OC are rarely explored. In this study, we determined that C. albicans infection promoted OC incidence in a 4-nitroquinoline 1-oxide (4NQO)-induced mouse tongue carcinogenesis model as well as promoted OC progression in a tongue tumor-bearing mouse model (C3H/HeN-SCC VII). We then demonstrated that tumor-associated macrophage (TAMs) infiltration was elevated during C. albicans infection. Meanwhile, the attracted TAMs polarized into M2-like ma
