Drug-inducible remote control of gene expression by probiotic Escherichia coli Nissle 1917 in intestine, tumor and gall bladder of mice.

The probiotic bacterium Escherichia coli Nissle 1917 (EcN) constitutes a prospective vector for delivering heterologous therapeutic molecules to treat several human disorders. To add versatility to this carrier system, bacteria should be equipped with expression modules that can be regulated deliberately in a temporal and quantitative manner. This approach is called in vivo remote control (IVRC) of bacterial vectors. Here, we have evaluated promoters P(araBAD), P(rhaBAD) and P(tet), which can be induced with L-arabinose, L-rhamnose or anhydrotetracycline, respectively. EcN harboring promoter c

How epigenetics and miRNA affect gene expression in dental pulp inflammation: A narrative review.

BACKGROUND: Pulpitis, an inflammatory condition of the dental pulp, typically arises due to caries. It can remain asymptomatic for extended periods, complicating its diagnosis. The inflammatory response induced by bacterial invasion encompasses both cell-mediated and humoural immunity, accompanied by neural and vascular changes. The primary aim of inflammation is to eradicate invading pathogens from the pulp. However, failure to eliminate pathogens may result in necrosis of the pulp. Before direct bacterial contact with cells occurs, the pulp initiates protective responses like the formation o

In vivo transcriptional profiling of Listeria monocytogenes and mutagenesis identify new virulence factors involved in infection.

Listeria monocytogenes is a human intracellular pathogen able to colonize host tissues after ingestion of contaminated food, causing severe invasive infections. In order to gain a better understanding of the nature of host-pathogen interactions, we studied the L. monocytogenes genome expression during mouse infection. In the spleen of infected mice, approximately 20% of the Listeria genome is differentially expressed, essentially through gene activation, as compared to exponential growth in rich broth medium. Data presented here show that, during infection, Listeria is in an active multiplicat

Dynamic cell type specificity of SRC-1 coactivator in modulating uterine progesterone receptor function in mice.

Regulation of gene transcription by the progesterone receptor (PR) in cooperation with coactivator/corepressor complexes coordinates crucial processes in female reproduction. To investigate functional relationships between PR and steroid receptor coactivators (SRCs) in distinct cell types of uterine tissue during gene transcription, we generated a new transgenic mouse model utilizing a Progesterone Receptor Activity Indicator (PRAI) system that could monitor PR activity in vivo. The PRAI system consists of a modified PR bacterial artificial chromosome (BAC) clone in which the DNA binding domai

RNA-Sequencing Reveals the Modulation of the NLRP3 Inflammasome by miR-223-3p in Extracellular Vesicles in Bacterial Endophthalmitis.

PURPOSE: Extracellular vesicles (EVs) are critical mediators of cell-cell communication via transfer of molecular cargo including miRNAs and regulate transcription in various physiological and pathological conditions. This study aimed to investigate the role of EV-derived-microRNAs (EV-miRNAs) in bacterial endophthalmitis, focusing on their regulatory impact on inflammation and host immune responses. METHODS: C57BL/6 mice were infected with Pseudomonas aeruginosa (P. aeruginosa) and Staphylococcus aureus (S. aureus) to induce endophthalmitis. EVs were isolated and characterized followed by mi

The Influence of Microwave Sterilization on the Ultrastructure, Permeability of Cell Membrane and Expression of Proteins of Bacillus Cereus.

Bacillus cereus was isolated from ready-to-serve brine goose, identified by 16S rRNA gene sequencing analysis and treated with a commercial microwave sterilization condition (a power of 1,800 W at 85°C for 5 min). The influence of microwaves on the morphology, the permeability of membrane and the expression of total bacterial proteins was observed. Microwave induced the clean of bacterial nuclear chromatin, increased the permeability and disrupted the integrity of membrane. Twenty-three proteins including 18 expressed down-regulated proteins and 5 expressed up-regulated proteins were identifie

Relationship between glycopeptide production and resistance in the actinomycete Nonomuraea sp. ATCC 39727.

Glycopeptides and β-lactams inhibit bacterial peptidoglycan synthesis in Gram-positive bacteria; resistance to these antibiotics is studied intensively in enterococci and staphylococci because of their relevance to infectious disease. Much less is known about antibiotic resistance in glycopeptide-producing actinomycetes that are likely to represent the evolutionary source of resistance determinants found in bacterial pathogens. Nonomuraea sp. ATCC 39727, the producer of A40926 (the precursor for the semisynthetic dalbavancin), does not harbor the canonical vanHAX genes. Consequently, we invest

malT knockout mutation invokes a stringent type gene-expression profile in Actinobacillus pleuropneumoniae in bronchoalveolar fluid.

BACKGROUND: Actinobacillus pleuropneumoniae causes contagious pleuropneumonia, an economically important disease of commercially reared pigs throughout the world. To cause this disease, A. pleuropneumoniae must rapidly overcome porcine pulmonary innate immune defenses. Since bronchoalveolar fluid (BALF) contains many of the innate immune and other components found in the lungs, we examined the gene expression of a virulent serovar 1 strain of A. pleuropneumoniae after exposure to concentrated BALF for 30 min. RESULTS: In reverse transcription PCR differential display (RT-PCR DD) experiments,

Inhibition of ascorbic acid on Pseudomonas bubulae under super-chilled storage.

To explore the inhibitory effect of 1% ascorbic acid on spoilage bacteria under super-chilled storage, tilapia fillets either with natural microbiota or inoculated with Pseudomonas bubulae P1, a dominant spoiler derived from tilapia, were employed in this research. Results showed that ascorbic acid significantly inhibited bacterial growth, delayed sensory spoilage, and reduced the levels of total volatile basic nitrogen (TVB-N) in both types of fillets stored at -1.5 °C. Further analysis indicated that the reduction in TVB-N was attributed to the diminished ammonia production by P1. Transcript

Haemophilus influenzae tryptophan biosynthesis is required for lung infection.

BACKGROUND: Tryptophan plays a key role in regulating human lung homeostasis and immunity through the indoleamine 2,3-dioxygenase (IDO) and aryl hydrocarbon receptor (AhR) signalling pathways. In patients with chronic obstructive pulmonary disease (COPD), altered IDO and AhR activity is observed, potentially favouring infection by pathogens capable of synthesizing their own tryptophan. In this study, we investigated the contribution of tryptophan availability to lung infection by Haemophilus influenzae, a tryptophan synthesizing pathobiont associated with COPD exacerbations. METHODS: A chemic

Fructose activates a stress response shared by methylglyoxal and hydrogen peroxide in Streptococcus mutans.

Fructose catabolism by Streptococcus mutans is initiated by three phosphotransferase (PTS) transporters yielding fructose-1-phosphate (F-1-P) or fructose-6-phosphate. Deletion of one such F-1-P-generating PTS, fruI, was shown to reduce the cariogenicity of S. mutans in rats fed a high-sucrose diet. Moreover, a recent study linked fructose metabolism in S. mutans to a reactive electrophile species methylglyoxal. Here, we conducted a comparative transcriptomic analysis of S. mutans treated briefly with 50 mM fructose, 50 mM glucose, 5 mM methylglyoxal, or 0.5 mM hydrogen peroxide (H2O2). The res

Optimization of the BoNT/A-Hc expression in recombinant Escherichia coli using the Taguchi statistical method.

The use of the recombinant BoNT/A-Hc (carboxylic domain of the Clostridium botulinum neurotoxin heavy chain) has been proposed as a vaccine candidate for botulism. This fragment contains the principle protective antigenic determinant. In the present study, in order to maximize recombinant protein expression, after verification of recombinant BoNT/A-Hc by Western blotting, modified M9 medium was selected as a simple medium, and the operational and medium-composition variables together with their interactions were optimized by using the Taguchi statistical method. Pyelonephritis decreases serum cholesterol and mitigates atherosclerosis severity despite systemic inflammation.

Hypercholesterolemia and inflammation are main causes of cardiovascular disease. Urinary tract infections are common and frequently recur. We here tested how pyelonephritis affects atherosclerotic plaque development and lipid levels. LDL receptor-deficient (Ldlr-/-) and wild-type mice were infected with uropathogenic Escherichia coli. Renal and systemic inflammation, lipid levels, and atherosclerotic plaque development were assessed. Gene regulation was studied in pyelonephritis and in human cells in vitro. In patients admitted with urinary tract infections, serum lipids and disease severity w

Identification and functional characterization of NifA variants that are independent of GlnB activation in the photosynthetic bacterium Rhodospirillum rubrum.

The activity of NifA, the transcriptional activator of the nitrogen fixation (nif) gene, is tightly regulated in response to ammonium and oxygen. However, the mechanisms for the regulation of NifA activity are quite different among various nitrogen-fixing bacteria. Unlike the well-studied NifL-NifA regulatory systems in Klebsiella pneumoniae and Azotobacter vinelandii, in Rhodospirillum rubrum NifA is activated by a direct protein-protein interaction with the uridylylated form of GlnB, which in turn causes a conformational change in NifA. We report the identification of several substitutions i

Inflammation and LPS-Binding Protein Enable the Stimulatory Effect of Endotoxin on Prolactin Secretion in the Ovine Anterior Pituitary: Ex Vivo Study.

Prolactin is a hormone that plays an important role in the regulation of many physiological processes including lactation, reproduction, fat metabolism, and immune response. The secretion of prolactin could be disturbed by an immune stress commonly accompanying infection. This study was designed to determine the influence of bacterial endotoxin-lipopolysaccharide (LPS)-on prolactin gene (PRL) expression and prolactin release from the ovine anterior pituitary (AP) explants collected from saline- and LPS-treated ewes in the follicular phase. The expressions of toll-like receptor 4 (TLR4) and pro

The vitamin D-antimicrobial peptide pathway and its role in protection against infection.

Vitamin D deficiency has been correlated with increased rates of infection. Since the early 19th century, both environmental (i.e., sunlight) and dietary sources (cod liver) of vitamin D have been identified as treatments for TB. The recent discovery that vitamin D induces antimicrobial peptide gene expression explains, in part, the 'antibiotic' effect of vitamin D and has greatly renewed interest in the ability of vitamin D to improve immune function. Subsequent work indicates that this regulation is biologically important for the response of the innate immune system to wounds and infection a

Octopamine modulates the innate immune response in Drosophila melanogaster.

BACKGROUND: Octopamine, the functional equivalent of noradrenaline in invertebrates, is a key neuromodulator that also influences immune functions. However, the receptor‑specific roles of octopamine in regulating innate immunity in Drosophila melanogaster remain incompletely understood. METHODS: Using loss‑of‑function mutants for the two major octopamine receptors Octβ1R and Octβ2R, combined with systemic bacterial infection models, bacterial clearance assays, in vivo and in vitro phagocytosis assays, and quantification of antimicrobial peptide (AMP) gene expression, we dissected the contribu

Vitamin B12-associated interactions between Mesorhizobium sp. TaiHu and Synechococcus sp. PCC 7002 revealed by multi-omics analysis.

The marine cyanobacterium Synechococcus sp. PCC 7002 (Syn7002) is a model organism that lacks the gene cluster required for vitamin B12 biosynthesis, necessitating cooperative interactions with other microbes. In this study, we established a synthetic microbial consortium by co-culturing Syn7002 with a bloom-forming Microcystis community, followed by purification, and subsequently investigated the interactions between Syn7002 and the associated microbial community. Electron microscopy revealed numerous rod-shaped bacteria clustered around Syn7002 cells, indicating close spatial associations be
