Sorafenib analogues exhibit dual Colistin-potentiating and anti-MRSA activity.

In response to the antimicrobial resistance (AMR) global health crisis, physicians have been forced to employ antibiotics of last resort such as colistin, which had previously been removed from clinics due to toxicity concerns. The increase in colistin usage has in turn resulted in the increased incidence of colistin-resistant isolates, making novel therapeutic options for infections caused by these pathogens a priority. We previously reported that sorafenib, a eukaryotic kinase inhibitor, is a colistin adjuvant, augmenting colistin activity against colistin-resistant Klebsiella pneumoniae and

Genotypic features of antimicrobial resistance, virulence determinants, and clonal lineages of Enterococcus spp. reported in Brazil: a scoping review with marked regional asymmetry.

PURPOSE: Vancomycin-resistant Enterococcus faecium is listed in the 2024 WHO Bacterial Priority Pathogens List as a high-priority agent of hospital-acquired infections. Its persistence is supported by marked genomic plasticity across human, animal, food, and environmental sources. We aimed to map antimicrobial resistance genes, virulence determinants, and clonal lineages of Enterococcus spp. reported in Brazil across these four sampling contexts. METHODS: Scoping review following JBI methodological guidance and reported per PRISMA-ScR. Four databases (PubMed, Embase, SciELO, LILACS) were sear

The role of masks and respirators in preventing respiratory infections in healthcare and community settings.

The covid-19 pandemic saw frequent changes and conflicts in mask policies and politicization of masks. On reviewing the evidence, including studies published after the pandemic, the data suggest respirators are more effective than masks in healthcare, but must be continuously worn to be protective. Healthcare and aged care settings amplify outbreaks, so protection of patients and staff is paramount. Most guidelines assume risk is only present during close contact or aerosol generating procedures, but studies show intermittent use of respirators is not protective. New research in aerosol scienc

Dynamics of Antimicrobial Susceptibility and Risk Factors Associated with Infections Caused by Colistin-Resistant Bacteria: A Study from the Northern Region of Haryana, India.

Antimicrobial resistance poses a significant threat to global health, with colistin as a last-resort antibiotic against multidrug-resistant (MDR) microorganisms. The present study aimed to investigate the dynamics of antimicrobial susceptibility patterns and risk factors associated with infections caused by colistin-resistant bacteria in the Northern region of Haryana, India. Clinical samples (n = 12,652) collected from a single hospital in Haryana were subjected to microbiological analysis for five months. Among the total samples (n = 12,652) processed, 24% (n = 3,061) showed growth of pathog

Ensuring progress on sustainable access to effective antibiotics at the 2024 UN General Assembly: a target-based approach.

Rising antimicrobial resistance (AMR) is a global health crisis for countries of all economic levels, alongside the broader challenge of access to antibiotics. As a result, development goals for child survival, healthy ageing, poverty reduction, and food security are at risk. Preserving antimicrobial effectiveness, a global public good, requires political will, targets, accountability frameworks, and funding. The upcoming second high-level meeting on AMR at the UN General Assembly (UNGA) in September, 2024, is evidence of political interest in addressing the problem of AMR, but action on targe

Design and synthesis of novel arylurea derivatives of aryloxy(1-phenylpropyl) alicyclic diamines with antimicrobial activity against multidrug-resistant Gram-positive bacteria.

The alarming increase in the resistance of bacteria to the currently available antibiotics necessitates the development of new effective antimicrobial agents that are active against bacterial pathogens causing major public health problems. For this purpose, our in-house libraries were screened against a wide panel of clinically relevant Gram-positive and Gram-negative bacteria, based on which compound I was selected for further optimization. Synthetic efforts in a group of arylurea derivatives of aryloxy(1-phenylpropyl) alicyclic diamines, followed with an in vitro evaluation of the activity a

Management of Crown-root Fracture with 180-degree Rotation Replantation: a Report of 2 Cases.

Intentional replantation involves a combination of periodontics, endodontics, prosthodontics and oral surgery. Crown-root fracture management is still complicated nowadays. A fracture line extending longitudinally to the subgingival area and intruding bioogical width could affect infection control, gingival health and crown restoration. In the present study, we present two cases. Case 1 involved a 23-year-old man who presented at our hospital with crown-root fracture of the maxillary left central incisor. A radiographic image of the tooth revealed a fracture line under the alveolar crest. The

Putative mobilized colistin resistance genes in the human gut microbiome.

BACKGROUND: The high incidence of bacterial genes that confer resistance to last-resort antibiotics, such as colistin, caused by mobilized colistin resistance (mcr) genes, poses an unprecedented threat to human health. Understanding the spread, evolution, and distribution of such genes among human populations will help in the development of strategies to diminish their occurrence. To tackle this problem, we investigated the distribution and prevalence of potential mcr genes in the human gut microbiome using a set of bioinformatics tools to screen the Unified Human Gastrointestinal Genome (UHGG

Facemask Alternatives in Veterinary Medicine in the Context of COVID-19 Shortages.

The COVID-19 pandemic has caused a widespread shortage of facemasks and other personal protective equipment in veterinary medicine without clear, research-based guidance on alternatives to FDA-certified disposable surgical masks. In the absence of detailed veterinary research, an in-depth review of the human medical literature was conducted to evaluate the viability of reusable, sterilizable cloth, medical textile, or other material alternatives that may be quickly manufactured and used by veterinarians. The results at the time of publication support the AVMA, CDC, and WHO recommendations to e

Infections Caused by Carbapenem-Resistant Enterobacteriaceae: An Update on Therapeutic Options.

Carbapenems are considered as last-resort antibiotics for the treatment of infections caused by multidrug-resistant Gram-negative bacteria. With the increasing use of carbapenems in clinical practice, the emergence of carbapenem-resistant pathogens now poses a great threat to human health. Currently, antibiotic options for the treatment of carbapenem-resistant Enterobacteriaceae (CRE) are very limited, with polymyxins, tigecycline, fosfomycin, and aminoglycosides as the mainstays of therapy. The need for new and effective anti-CRE therapies is urgent. Here, we describe the current understandin
