Growth hormone-releasing hormone: past and present.

The discovery of hypothalamic hypophysiotropic factors confirmed the hypothesis of Green and Harris in the late 1940s. These hormones were isolated from their eutopic site of production (the hypothalamus) with the exception of growth hormone (GH)-releasing hormone (GHRH), which was isolated from an ectopic, tumoral site of production and found to be responsible for acromegaly. Following the isolation, characterization and synthesis of human GHRH, clinical studies were performed and are described below. Circulating levels of GHRH can be measured and provide the basis for the diagnosis of acrome…

[Bone and hormones. Effects of parathyroid hormone on the bone].

ACTION OF PTH: Parathyroid hormone (PTH) stimulates both bone formation and bone resorption. It also stimulates synthesis of 1,25-dihydroxyvitamin D3 which increases intestinal absorption of calcium and phosphorus and stimulates bone resorption. In bone tissue, PTH acts mainly on osteoblasts which, unlike osteoclasts, carry PTH receptors. PTH also exerts an indirect action on osteoclasts, increasing their number and activity. IMPACT OF INSUFFICIENT PTH: In the absence of PTH, bone remodeling is greatly reduced. Increased bone density has been observed in patients with hypoparathyroidism studie

Origin of luteinizing hormone-releasing hormone neurons.

Neurons expressing luteinizing hormone-releasing hormone (LHRH), found in the septal-preoptic nuclei and hypothalamus, control the release of gonadotropic hormones from the anterior pituitary gland and facilitate reproductive behaviour. LHRH-expressing neurons are also found in the nervus terminalis, a cranial nerve that is a part of the accessory olfactory system and which projects directly from the nose to the septal-preoptic nuclei in the brain. During development, LHRH-immunoreactivity is detected in the peripheral parts of the nervus terminalis before it is found in the brain. Using a com…

Growth hormone-releasing hormone: therapeutic perspectives.

Growth hormone-releasing hormone (GHRH), the hypothalamic peptide which stimulates the release of growth hormone (GH) from the pituitary, has been the center of intense study since its structure was described in 1982. It is in the area of growth hormone deficiency (GHD) that GHRH has its greatest clinical potential. Several studies have addressed the efficacy of GHRH as an alternative therapy in GHD. It appears that GHRH can be equally effective, whether given in a pulsatile manner or as discrete injections several times a day; i.e. the total dose may be more crucial than how it is given. Whil

Testing for growth hormone deficiency in adults: doing without growth hormone-releasing hormone.

PURPOSE OF REVIEW: This article summarizes recent advances in testing for growth hormone deficiency (GHD) in adults, focusing on critical appraisal of existing growth hormone (GH) provocative tests as well as newer tests in development. RECENT FINDINGS: The diagnosis of GHD can be challenging and often requires the use of GH provocative testing. The most widely validated of these is insulin-induced hypoglycemia (ITT), which requires close supervision and has significant contraindications and side-effects. The arginine-growth hormone-releasing hormone (GHRH) test had become widely used as a sa

[Bone and bone related biochemical examinations. Hormone and hormone related substances. Parathyroid hormone].

Parathyroid hormone (PTH) is the most important hormone to maintain serum calcium levels. PTH acts directly on bone by stimulating both bone resorption and bone formation with the reactive excess in the former when it acts continuously. PTH also acts on renal tubule to promote urinary excretion of phosphate as well as production active vitamin D (1,25 [OH] 2D) . 1,25 [OH] 2D has been also suggested to have some unique and direct action on the bone. Measurement of intact-PTH is essential for optimal management of bone abnormalities in patients with primary hyperparathyroidism, secondary hyperpa

Growth hormone response to growth hormone-releasing hormone varies with the hypothalamic-pituitary abnormalities.

We determined growth hormone (GH) and insulin-like growth factor I (IGF-I) levels after a 3 h infusion of escalating doses of growth hormone-releasing hormone (GHRH(1-29)) followed by a bolus injection in hypopituitary patients with marked differences in pituitary features at magnetic resonance imaging (MRI) in order to evaluate further the contribution of MRI in the definition of pituitary GH reserve in GH-deficient patients. Twenty-nine patients (mean age 14.5 +/- 4.0 years) were studied. Group I comprised 13 patients: seven with isolated GH deficiency (IGHD) (group Ia) and six with multiple

Inhibitory effect of amylin on growth hormone responsiveness to growth-hormone-releasing hormone in the rat.

The effects of intracerebroventricular (i.c.v.) or intracarotid (i.a.) administration of amylin (AMY) on growth hormone (GH) release induced by GH-releasing hormone (GHRH) were examined in conscious male rats. Amylin (25 ng-5 micrograms/rat, i.c.v. or 10 micrograms/rat, i.a.) was administered 10 min before GHRH (2 micrograms/kg, i.a.). I.c.v. administration of AMY dose-dependently inhibited GH secretion induced by GHRH but when given peripherally, AMY did not modify the GH response to GHRH. Amylin (10(-8)-10(-6) M) had no direct effect on the rat anterior pituitary in vitro either alone or whe

Therapeutic use of gonadoliberin (follicle-stimulating hormone/luteinizing hormone-releasing hormone) in women.

Certain conclusions may be drawn from the present review: 1. Synthetic FSH/LH-RH may induce ovulation; therefore, a therapeutic effect has been established in some cases of anovulatory infertility, but it is still difficult to assess the correct dose of FSH/LH-RH because of individual variations in response. 2. Gonadoliberin may also be used to induce ovulation after follicular maturation has been evoked by other agents. FSH/LH-RH can be utilized for supplementing the LH surge after clomiphene therapy in cases of clomiphene failure. When associated with HMG, the synthetic decapeptide may be he

Hormonal profiles in anovulatory patients treated with gonadotropins and synthetic luteinizing hormone releasing hormone.

Ten patients with hypothalamic anovulation weretreated with a "retard" preparation of synthetic luteinizing hormone releasing hormone (LHRH) after an HMG stimulation in order to induce ovulation and pregnancy. Four of the patient ovulated after intramuscular administration of the LHRH preparation. This study suggests that is is possible to induce ovulation with LHRH in patients pretreated with HMG, and that LHRH has advantages over HCG since it does not induce hyperstimulation even in the presence of exagerated follicular growth. Nevertheless, the optimal conditions for the use and monitoring

Multiple sclerosis occurrence with gut microbial metabolites and sex hormones.

Multiple sclerosis (MS) is a chronic immune and neurodegenerative condition that affects the central nervous system (CNS). Multiple factors, both genetic (including the HLA gene locus) and environmental (Epstein-Barr virus, low vitamin D levels), may contribute to MS development. Growing evidence supports the importance of the gut-brain axis and hormones in disease pathogenesis and progression. This review focuses on identifying the role of specific gut microbial products (short-chain fatty acids, tryptophan derivatives, bile acids, and polyamines) in this process and how they affect the immun…

Indications for Dose Adjustment of Thyroid Hormones in Older Patients.

BACKGROUND: Thyroid dysfunction is common in older people (persons aged 65 and above). A meta-analysis of data from European countries revealed a 4.1% prevalence of undiagnosed subclinical hypothyroidism. It was found in a primary care study from the United Kingdom that approximately 10 percent of people over age 70 were under treatment for hypothyroidism. METHODS: This narrative review is based on findings in relevant publications retrieved by a PubMed search and on the guidelines of the European Thyroid Association and the American Thyroid Association. RESULTS: Overtreatment with levothyro

Perimenopause and metabolic vulnerability: hormones, body composition and lifestyle changes.

Perimenopause is a transitional phase characterized by substantial hormonal fluctuations, particularly declining estradiol levels, accompanied by age-related metabolic changes. Increasing evidence suggests that this period constitutes a critical window of metabolic vulnerability, marked by adverse alterations in body composition, energy metabolism and cardiometabolic health. Notably, these changes may develop even without significant increases in body weight, potentially delaying their recognition in clinical settings. A narrative review was conducted using the PubMed/MEDLINE, Scopus, Web of S

Ovarian hormones moderate systolic hypertension in female Eln haploinsufficient mice.

Hypertension is a hallmark of cardiovascular abnormalities associated with Williams syndrome (WS), a rare genetic disorder involving microdeletion of genes on human chromosome 7, including the elastin gene (ELN). Heterozygous deletion of Eln (Eln+/-) in mice recapitulates hypertension and arteriopathy associated with WS. Previously, differences in blood pressure elevation and sensitivity to dietary sodium were found to be less profound in female Eln+/- mice. Here, we determined whether ovarian hormones play a role in sex-related differences in blood pressure elevation resulting from Eln haploi…

Sex hormones and their receptors in osteoarthritis: current research progress.

Osteoarthritis (OA) is the most prevalent degenerative joint disease worldwide and a leading cause of pain, disability, and reduced quality of life, with pronounced sex-specific differences in incidence and clinical presentation. Epidemiological evidence shows a sharp increase in OA prevalence in women after menopause, whereas in men, the elevated risk at older age is more closely associated with declines in bone mass, muscle strength, and biomechanical stability. Increasing experimental and clinical studies indicate that sex hormones-including estrogen, androgens, and progesterone-and their r…

Impact of intestinal resections on the secretion of gastrointestinal hormones.

BACKGROUND AND AIMS: Intestinal resection changes nutrient exposure of enteroendocrine cells and may alter gastrointestinal hormone secretion. This study aims to characterize basal and postprandial gastrointestinal hormone profiles across distinct remnant intestinal anatomies after intestinal resection. METHODS: In this single-center, cross-sectional study, 31 patients were grouped according to remnant anatomy: end-jejunostomy (Group 1A, n = 8), ileostomy (1B, n = 8), jejuno-colonic anastomosis (2, n = 7) and jejuno-ileal anastomosis (3, n = 8) and compared with 8 healthy controls. The extragonadal biology of gonadotropin hormones: should we rename these?

Over recent years, gonadotropins and their receptors have been shown to exert non-traditional actions that bypass the classical hypothalamic-pituitary-gonadal axis. Findings of the expression of receptors for follicle-stimulating hormone (FSH) and luteinizing hormone (LH) in skeletal, fat, and immune cells and in neurons suggest that their functions are much broader than their classical roles. FSH, once believed solely to regulate procreation-gonadal development and maturation at puberty and gamete production during the fertile phase, has been found to regulate body composition, bone metabolis

Associations between circulating sex hormones and anterior urethral stricture disease.

BACKGROUND: The pathophysiology of anterior urethral stricture disease (aUSD) is unknown (idiopathic) in the majority of men. Testosterone is required for the in utero development of the male urethra and has been shown to aid in urethral healing. Thus, the purpose of this study was to understand the relationship between circulating sex hormones and aUSD. METHODS: This study included 154 males with aUSD undergoing urethroplasty who were enrolled in a prospective, multicenter study evaluating the relationship between inflammation and urethral strictures. This study represents a secondary analys…
