The morphological analysis of autophagy in primary skeletal muscle cells infected with Toxoplasma gondii.

Toxoplasma gondii is an obligate intracellular protozoan parasite, the causative agent of toxoplasmosis, one of the most widespread zoonoses in the world. During the host immune response, tissue cysts are formed, allowing the maintenance of the parasite within the host cell. Autophagy, a degradation process of cellular components, is critical for cellular homeostasis. Recently, it has been proposed that autophagy participates in host-pathogen interactions. Autophagic inducers (rapamycin or glucose plus serum deprivation) inhibited infection and parasite proliferation in a clinically relevant m

Unlocking the Biological Enigma: Influence of Host Length and Infection Site on Parasite Abundance in Ompok bimaculatus.

PURPOSE: The influence of two key factors, host length and infection site, on the host-parasite interaction in Ompok bimaculatus (Butter catfish) from Mukutmanipur Dam Lake, were investigated. METHODS: Present study involved 192 specimens of Ompok bimaculatus with varying body lengths, subjected to diverse statistical analyses. One-way analysis of variance (ANOVA) was performed for the parasite numbers for three groups (cestode, nematode and trematode). Subsequently, we conducted one-way permutational multivariate analysis of variance (PERMANOVA) followed by pairwise test to assess parasite n

[Behavioural Changes Caused by Parasites: The Parasite Manipulation Hypothesis].

Bacteria, archaeae, fungi and viruses of the intestinal microbiome play an important role as symbionts in the complex human ecosystem. Parasites, which account for about 40 % of the earth's biodiversity, depend on hosts to complete their life cycle. This article explains how they use them and which signalling pathways play a role in this, using toxoplasmosis and malaria as examples. The parasitic manipulation hypothesis is based on impressive observations in the wild and under laboratory conditions, especially in invertebrates. For the assumption of an adaptive manipulation, every step from th

Helminth parasites and immune regulation.

Helminth parasites are complex metazoans that belong to different taxonomic families but that collectively share the capacity to downregulate the host immune response directed toward themselves (parasite-specific immunoregulation). During long-standing chronic infection, these helminths appear able to suppress immune responses to bystander pathogens/antigens and atopic, autoimmune, and metabolic disorders. Helminth-induced immunoregulation occurs through the induction of regulatory T cells or Th2-type cells (or both). However, secreted or excreted parasite metabolites, proteins, or extracellul

Solo Schistocephalus solidus tapeworms are nasty.

Trophically transmitted parasites must trade-off own growth on one hand and energy drain from the intermediate host on the other hand, since killing the host before transmission to the next host is a dead end for both parasites and hosts. This challenge becomes especially intriguing when multiple parasites find themselves within the same individual host. The tapeworm Schistocephalus solidus may gain more than 98% of its final body mass within few months infecting its three-spined stickleback (Gasterosteus aculeatus) intermediate host. During these months the tapeworms may achieve a mass even l

Gliding motility in apicomplexan parasites.

Apicomplexan parasites, including Plasmodium and Toxoplasma, employ a unique form of substrate-dependent locomotion known as gliding motility. In these obligate, intracellular parasites, gliding motility is used for migration through the tissues and cells of the host, for active penetration of the host cell, and, at times, for proactive egress from the host. Gliding motility is powered by an actin-myosin based motor apparatus, known as the glideosome, which is situated within the elaborate cortical domain of the parasite. In this system, myosin is anchored to an internal membrane complex and d

An evolutionary perspective on gastrointestinal nematodes of sheep.

The purpose of this paper was to discuss from an evolutionary perspective the interaction between domestic sheep (Ovis aries) and their gastrointestinal nematodes. Although evolution is the central theme of biology, there has been little attempt to consider how evolutionary forces have shaped and continue to shape the relationships between domestic animals and their parasite community. Mathematical modelling of the host-parasite relationship indicated that the system is remarkably robust to perturbations in its parameters. This robustness may be a consequence of the long coevolution of host an

Invasion of skin by schistosome cercariae: some neglected facts.

The process of skin invasion by Schistosoma cercariae was reviewed in a recent Trends Research Update, accompanied by a computer animation. Some aspects of that article were misleading and perpetuated misconceptions about parasite migration through the skin that should by now have been dispelled. This article sets out a different interpretation of events, taking account of the extensive data on migration and larval structural changes that have been documented for Schistosoma mansoni over the past 20 years.

Adaptive physiological processes in the host during gastrointestinal parasitism.

Parasite infection of the gastrointestinal tract with helminths or protozoa induces detrimental effects on host tissues and host physiology, which have been extensively studied and reviewed. However, parasitism of the digestive system is also associated with adaptive, compensatory phenomena based on changes in host physiology or structures and which tend to counterbalance the negative consequences. The objective of this review is to describe these adaptive processes and their possible underlying mechanisms. Different processes which tend to attenuate the effect of either the loss of appetite,

Pentastomid infections in cichlid fishes in the Kruger National Park and the description of the infective larva of Subtriquetra rileyi n. sp.

During 1995, studies were conducted on the pentastome fauna of the cichlid fishes Tilapia rendalli and Oreochromis mossambicus in the Kruger National Park. The prevalence of infective pentastome larvae was 40.5% in T. rendalli and 9.2% in O. mossambicus. Encapsulated nymphs of Leiperia cincinnalis were taken from the mesentery, while Sebekia wedli was either encapsulated or free-living in the swim bladder. The subtriquetrids moved about freely in the swim bladder. L. cincinnalis was present in 0.5% of T. rendalli and 0.8% of O. mossambicus and additional descriptions and measurements of the ny

Parasitic infection and autoimmunity.

The studies summarized in this paper indicate that parasitic infections can serve as a trigger factor of autoimmune reactivity by several mechanisms. The relationship between parasites and autoimmunity could be manifested by the presence of autoantibodies or T-cells with autoreactivity. In spite of the evidence that has accumulated, the specific association between infection and autoimmunity is still obscure. The reasons for tissue damage in parasitic diseases are controversial. Some believe it is the result of pathogenic autoantibodies or autoreactive T-cells. Others argue against the causati

An experimental Schistosoma mattheei infection in man.

Certain aspects of the immune response of a male experimentally infected with 3-day old cercariae of a pure field strain of Schistosoma matheei were investigated. Among others, aspects such as the reaction of eosinophils, neutrophils and blood platelets after infection, were included in the study. The involvement of IgG and the cross reaction between these antibodies and S. haematobium and S. mansoni were also investigated. The phenomenon that the cercariae were, 3 days after shedding, still capable of penetrating the skin causing an inflammatory response was studied. The results lend some sup

Strongyloides stercoralis: loss of ability to disseminate after repeated passage in laboratory beagles.

A strain of Strongyloides stercoralis originally isolated from a south-east Asian patient and maintained by passage in monkeys for four years was introduced into parasite-free laboratory-reared beagles. After the second passage in these dogs, the prepatent period shortened from an average of 12-14 d to 8-9 d. Up to the third generation, immunosuppressive treatment of the dogs with oral corticosteroids caused hyperinfection with dissemination, albeit limited, to extra-intestinal organs. From the fourth generation onward, neither hyperinfection nor extra-intestinal migration could be induced eve

Roles of helminth extracellular vesicle-derived let-7 in host-parasite crosstalk.

Helminth infections are a major public health problem as they can cause long-term chronic infections in their hosts for which there is no effective vaccine. During the long-term interaction between helminths and their hosts, helminth-derived extracellular vesicles (EVs) can participate in host immunomodulatory processes by secreting bioactive molecules (BMAs). Growing data suggests that microRNAs (miRNAs) in helminth EVs have a significant impact on the host's immune system. The let-7 family is highly conserved among helminth EVs and highly homologous in the host, and its function in host-para

Lectin-Mediated Bacterial Modulation by the Intestinal Nematode Ascaris suum.

Ascariasis is a global health problem for humans and animals. Adult Ascaris nematodes are long-lived in the host intestine where they interact with host cells as well as members of the microbiota resulting in chronic infections. Nematode interactions with host cells and the microbial environment are prominently mediated by parasite-secreted proteins and peptides possessing immunomodulatory and antimicrobial activities. Previously, we discovered the C-type lectin protein AsCTL-42 in the secreted products of adult Ascaris worms. Here we tested recombinant AsCTL-42 for its ability to interact wit

Sapecin B alters kinetic properties of rapidly inactivating K(+) channels in rat pituitary GH(3) cells.

BACKGROUND: Rhizocephalan interaction with their decapod hosts is a superb example of host manipulation. These parasites are able to alter the host's physiology and behavior. Host-parasite interaction is performed, presumably, via special modified rootlets invading the ventral ganglions. METHODS: In this study, we focus on the morphology and ultrastructure of these special rootlets in Polyascus polygeneus (Lützen & Takahashi, 1997), family Polyascidae, invading the neuropil of the host's nervous tissue. The ventral ganglionic mass of the infected crabs were fixed, and the observed sites of th

Parasites alter host community structure in a natural experiment.

Parasites can profoundly alter host communities. However, the impact of parasites can vary from one community to another. Understanding why the impact of parasites varies across communities is challenging because it requires (i) separating the direct effects of the parasite on the host species from the indirect effects it exerts through the ecological interactions among the hosts and (ii) identifying how the presence of one host alters rates of infection in another. Freshwater fish communities in Trinidad have recently been invaded by a generalist parasitic nematode. This event, combined with
