Forensic microbiology reveals that Neisseria animaloris infections in harbour porpoises follow traumatic injuries by grey seals.

Neisseria animaloris is considered to be a commensal of the canine and feline oral cavities. It is able to cause systemic infections in animals as well as humans, usually after a biting trauma has occurred. We recovered N. animaloris from chronically inflamed bite wounds on pectoral fins and tailstocks, from lungs and other internal organs of eight harbour porpoises. Gross and histopathological evidence suggest that fatal disseminated N. animaloris infections had occurred due to traumatic injury from grey seals. We therefore conclude that these porpoises survived a grey seal predatory attack,

An extracellular Leptospira interrogans leucine-rich repeat protein binds human E- and VE-cadherins.

Pathogenic Leptospira bacteria are the causative agents of leptospirosis, a zoonotic disease affecting animals and humans worldwide. These pathogenic species have the ability to rapidly cross host tissue barriers by a yet unknown mechanism. A comparative analysis of pathogens and saprophytes revealed a higher abundance of genes encoding proteins with leucine-rich repeat (LRR) domains in the genomes of pathogens. In other bacterial pathogens, proteins with LRR domains have been shown to be involved in mediating host cell attachment and invasion. One protein from the pathogenic species Leptospir

Transcriptomic validation of a 7,12 Dimethylbenz(a)anthracene (DMBA)-induced leukemia rat model: Parallels with human leukemogenesis.

BACKGROUND: Experimental animal models are essential for understanding the molecular mechanisms of human leukemia and testing potential therapies. Chemical induction using 7,12-dimethylbenz[a]anthracene (DMBA) has shown promise in recapitulating features of leukemogenesis in rodents, but its molecular fidelity to human disease remains underexplored. The aim of this study was to evaluate the transcriptomic landscape of a DMBA-induced leukemia rat model and assess its alignment with known gene expression signatures and oncogenic pathways in human leukemia. METHODS: Male Wistar rats were treated

Role of growth factors in cutaneous wound healing: a review.

The well-orchestrated, complex series of events resulting in the repair of cutaneous wounds are, at least in part, regulated by polypeptide growth factors. This review provides a detailed overview of the known functions, interactions, and mechanisms of action of growth factors in the context of the overall repair process in cutaneous wounds. An overview of the cellular and molecular events involved in soft tissue repair is initially presented, followed by a review of widely studied growth factors and a discussion of commonly utilized preclinical animal models. The article concludes with a summ

Pharmacological Interactions of Paraoxonase 1 (PON1): A HDL-Bound Antiatherogenic Enzyme.

A growing interest exists in documenting the role of paraoxonase 1 (PON1) in different human diseases including, cardiovascular disease, obesity, diabetes mellitus, cancers, aging, and several neurological disorders. Three aspects of PON1 have attracted the attention of researchers: (1) hydrolyzing and detoxifying of toxic organophosphorous compounds such as nerve gases; (2) antioxidative activity in hydrolyzing oxidized phospholipids in high-density lipoprotein (HDL) and low-density lipoprotein (LDL); (3) interaction with various drugs. Drugs and nutrients which can increase the activity of p

Systematic Review of Important Bacterial Zoonoses in Africa in the Last Decade in Light of the 'One Health' Concept.

Zoonoses present a major public health threat and are estimated to account for a substantial part of the infectious disease burden in low-income countries. The severity of zoonotic diseases is compounded by factors such as poverty, living in close contact with livestock and wildlife, immunosuppression as well as coinfection with other diseases. The interconnections between humans, animals and the environment are essential to understand the spread and subsequent containment of zoonoses. We searched three scientific databases for articles relevant to the epidemiology of bacterial zoonoses/zoonot

Long-term outcomes of developmental exposure to fluoxetine: a review of the animal literature.

During and following pregnancy, women are at high risk of experiencing depression, for which fluoxetine (FLX; brand names Prozac, Sarafem, Rapiflux) is the most commonly prescribed treatment. An estimated 1.4-2.1% of pregnant women use this medication, which inhibits the reuptake of serotonin and thereby increases serotonergic activity at the synapse. Serotonin acts as a cue guiding numerous neurodevelopmental processes, and changes in the concentration of serotonin can disrupt normal in utero brain development and organization in humans and other animals, thus providing a mechanism by which m

Reduced mobility but unaffected startle response in female rats exposed to prenatal dexamethasone: different sides to a phenotype.

An adverse fetal environment is strongly associated with behavioral and emotional development in later life, and environmental interactions with the genome are essential in the development of pathophysiology. This implicates that a genetic vulnerability or other predisposition may interact with the environment and stressful life events to trigger mental disease. The startle reflex is highly sensitive to fear and anxiety in humans and animals. Elevated startle magnitude has been proposed as a marker for neurodevelopmental disorders. We have recently established an animal model for possible deve

Retinal organoids in disease modeling and drug discovery: Opportunities and challenges.

Diseases leading to retinal cell loss can cause severe visual impairment and blindness. The lack of effective therapies to address retinal cell loss and the absence of intrinsic regeneration in the human retina leads to an irreversible pathological condition. Progress in recent years in the generation of human three-dimensional retinal organoids from pluripotent stem cells makes it possible to recreate the cytoarchitecture and associated cell-cell interactions of the human retina in remarkable detail. These human three-dimensional retinal organoid systems made of distinct retinal cell types an

Rift valley fever outbreak in Sembabule District, Uganda, December 2020.

BACKGROUND: Rift Valley Fever (RVF) is a viral zoonosis that can cause severe haemorrhagic fevers in humans and high mortality rates and abortions in livestock. On 10 December 2020, the Uganda Ministry of Health was notified of the death of a 25-year-old male who tested RVF-positive by reverse-transcription polymerase chain reaction (RT-PCR) at the Uganda Virus Research Institute. We investigated to determine the scope of the outbreak, identify exposure factors, and institute control measures. METHODS: A suspected case was acute-onset fever (or axillary temperature > 37.5 °C) and ≥ 2 of: head

Understanding consumption of animal- and plant-based protein sources in the Netherlands: A stakeholder-driven causal loop diagram.

Current consumption of animal-based protein sources is environmentally unsustainable and poses risks to human health, animal welfare, and food security. Policymakers in many countries seek to reduce the consumption of animal-based protein sources. However, this transition is affected by many factors, and it remains unclear how they interact and what their potential is for stimulating systemic change. This study synthesized stakeholders' perspectives on factors driving the consumption of animal- and plant-based protein sources into a Causal Loop Diagram (CLD), visualiing factors and their cause

Multi-season survey of ixodid tick species collected from domestic dogs in Chad, Africa.

Ticks are medically important vectors of pathogens, many of which are zoonotic or impact domestic animal and/or wildlife health. Climate change, landuse modifications, and increasing interactions between domestic animals, wildlife, and humans have resulted in changes in tick-host dynamics and the emergence of novel pathogens worldwide. Therefore, describing the host and geographic ranges of vector species is essential in assessing disease risk, especially in understudied areas, and should be conducted in a One Health context. In sub-Saharan Africa, previous work on ticks has focused primarily

Are ticks venomous animals?

INTRODUCTION: As an ecological adaptation venoms have evolved independently in several species of Metazoa. As haematophagous arthropods ticks are mainly considered as ectoparasites due to directly feeding on the skin of animal hosts. Ticks are of major importance since they serve as vectors for several diseases affecting humans and livestock animals. Ticks are rarely considered as venomous animals despite that tick saliva contains several protein families present in venomous taxa and that many Ixodida genera can induce paralysis and other types of toxicoses. Tick saliva was previously proposed

Cryptosporidium parvum-induced ileo-caecal adenocarcinoma and Wnt signaling in a mouse model.

Cryptosporidium species are apicomplexan protozoans that are found worldwide. These parasites constitute a large risk to human and animal health. They cause self-limited diarrhea in immunocompetent hosts and a life-threatening disease in immunocompromised hosts. Interestingly, Cryptosporidium parvum has been related to digestive carcinogenesis in humans. Consistent with a potential tumorigenic role of this parasite, in an original reproducible animal model of chronic cryptosporidiosis based on dexamethasone-treated or untreated adult SCID mice, we formerly reported that C. parvum (strains of a

[The gene of human prion: reassessment of its features and pathogenic role].

After a detailed examination of some features of the human prion gene, we propose a reassessment of its role in the pathogenicity of the derived neurodegenerative diseases e.g. Creutzfeldt-Jakob's disease. The particular resistance of this gene towards physico-chemical agents is related to the relative abundance of the guanine and cytosine bases. Other features such as the paucity of CpG sequences, GC box, polyadenylic tail, direct and inverted repeats, all evoke those of retroviral LTRs. Finally, the presence of polypurinic and polyadenylic sequences, as well as that of a functional AP2 eleme

Knockout of key receptors (PDGFRA and NRP2) in the guinea pig model blocks direct and endocytic pathways of CMV cell entry.

The guinea pig with guinea pig cytomegalovirus (GPCMV) is the only small-animal model for congenital cytomegalovirus, a leading cause of cognitive impairment and hearing loss in newborns. GPCMV encodes human cytomegalovirus (HCMV) homologues of viral entry glycoprotein complexes, which are neutralizing-antibody vaccine targets. As with HCMV, GPCMV has two pathways of cell entry (direct and endocytic). Specific viral gH/gL-based complexes are necessary for receptor interaction and cell entry: gH/gL/gO trimer (direct) and pentamer complex (PC) (endocytic). Both pathways also require gB as the fu

Protease-activated receptor 2 and interleukin-13 receptor α1 activation is linked to eosinophilic chronic rhinosinusitis.

BACKGROUND: Protease-activated receptor 2 (PAR-2) and interleukin (IL)-13 receptor α1 (Rα1) play major roles in type 2 inflammation. However, most of the literature was limited to allergic asthma. OBJECTIVE: To evaluate ways these receptors contribute to upper respiratory tract inflammation and to explore potential therapeutic targets in patients with eosinophilic chronic rhinosinusitis. METHODS: Using protein interaction analysis, animal experiments, and human tissue samples, we assessed the effects of exposure to house dust mite allergen on PAR-2 and IL-13Rα1 activation and inflammatory ma

Associations between maternal folate status and choline intake during pregnancy and neurodevelopment at 3-4 years of age in the Alberta Pregnancy Outcomes and Nutrition (APrON) study.

Folate and choline are methyl donor nutrients that may play a role in fetal brain development. Animal studies have reported that prenatal folate and choline supplementation are associated with better cognitive outcomes in offspring and that these nutrients may interact and affect brain development. Human studies that have investigated associations between maternal prenatal folate or choline levels and neurodevelopmental outcomes have reported contradictory findings and no human studies have examined the potential interactive effect of folate and choline on children's neurodevelopment. During t
