Impairment of pH gradient and membrane potential mediates redox dysfunction in the mitochondria of the post-ischemic heart.

The mitochondrial electrochemical gradient (Δp), which comprises the pH gradient (ΔpH) and the membrane potential (ΔΨ), is crucial in controlling energy transduction. During myocardial ischemia and reperfusion (IR), mitochondrial dysfunction mediates superoxide (·O2-) and H2O2 overproduction leading to oxidative injury. However, the role of ΔpH and ΔΨ in post-ischemic injury is not fully established. Here we studied mitochondria from the risk region of rat hearts subjected to 30 min of coronary ligation and 24 h of reperfusion in vivo. In the presence of glutamate, malate and ADP, normal mitoc

One-electron oxidation of acetohydroxamic acid: the intermediacy of nitroxyl and peroxynitrite.

The pharmacological effects of hydroxamate derivatives have been attributed not only to metal chelation or enzyme inhibition but also to their ability to serve as nitroxyl (HNO/NO(-)) and nitric oxide (NO) donors. However, the mechanism underlying the formation of these reactive nitrogen species is not clear and requires further elucidation. In the present study, one-electron oxidation of acetohydroxamic acid (aceto-HX) by (•)OH, (•)N(3), (•)NO(2), CO(3)(•-), and O(2)(•-) radicals was investigated using pulse radiolysis. It is demonstrated that only (•)OH, (•)N(3), and CO(3)(•-) radicals attac

The role of 18-methyleicosanoic acid in the structure and formation of mammalian hair fibres.

Although branched chain fatty acids perform many functions in biological systems, the importance of the anteiso 18 methyleicosanoic acid (MEA) has only recently been recognized. In this first review on MEA its role and distribution is explored. MEA has been found in minor amounts in the fatty acid components of a wide range of biological materials, but the current interest results from it being the major covalently bound fatty acid in mammalian hair fibres, a finding which is unusual because protein-bound fatty acids are typically straight-chain, even-numbered acids (C14-C18). MEA is released

Host-targeted repurposed diltiazem enhances the antiviral activity of direct acting antivirals against Influenza A virus and SARS-CoV-2.

Viral respiratory infections remain a major and recurrent public health threat. Among them, influenza viruses are responsible for ⁓500,000 deaths worldwide and a high economic burden. The recurrent threat of emerging zoonotic or pandemic viruses worsens this scenario, being SARS-CoV-2 and the millions of COVID-19 deaths the most recent example. The rapid evolution of circulating influenza and SARS-CoV-2 viruses allows the emergence and dissemination of variant strains carrying mutations resulting in suboptimal vaccine protection and/or reduced efficacy of current limited therapeutic arsenal. I

Deciphering the Enzymatic Function of the Bovine Enteric Infection-Related Protein YfeJ from Salmonella enterica Serotype Typhimurium.

Non-typhoidal Salmonella can colonize the gastrointestinal system of cattle and can also cause significant food-borne disease in humans. The use of a library of single-gene deletions in Salmonella enterica serotype Typhimurium allowed identification of several proteins that are under selection in the intestine of cattle. STM2437 ( yfeJ) encodes one of these proteins, and it is currently annotated as a type I glutamine amidotransferase. STM2437 was purified to homogeneity, and its catalytic properties with a wide range of γ-glutamyl derivatives were determined. The catalytic efficiency toward t

Sorption-spectrophotometric determination of Ni(II) in natural waters using nitrazine yellow immobilised on a polyamine-modified polyacrylonitrile fibre.

A solid-phase sorption-spectrophotometric procedure was developed for the determination of Ni(II) in natural waters using nitrazine yellow (NY) immobilised on PPA-1, an anion-exchange fibre obtained by treating hydroxylamine-activated polyacrylonitrile with polyethylene polyamine. Reagent immobilisation and Ni(II) binding were characterised by absorption and diffuse-reflectance spectroscopy, Raman spectroscopy, conductometry and scanning electron microscopy with energy-dispersive X-ray analysis (SEM-EDS). In aqueous solution, the absorption maximum of NY shifts from 465 to 583 nm on complexati

Rhobot-Screen: an integrated robotic platform for functional screening of rhodopsin variants.

BACKGROUND: Rhodopsins are photoreceptive membrane proteins widely used as optogenetic tools in basic research and medical applications, and extensive mutational studies have been performed to improve or modify their functional properties. Recently, in the broader field of protein engineering, data-driven strategies based on machine learning have attracted increasing attention, as they enable efficient exploration of vast mutational spaces with a reduced number of experiments. Such approaches require large, consistent datasets that link predefined mutations to quantitative functional propertie

Detection of NADH/NAD+ dysregulation in MELAS via diazo-carboxyl click derivatization mass spectrometry.

OBJECTIVES: Mitochondrial encephalomyopathy, lactic acidosis, and stroke-like episodes (MELAS) is a mitochondrial disorder driven by mutations in mitochondrial or nuclear DNA, involving an altered NADH/NAD+-associated redox metabolism as a key pathological mechanism. The traditional metabolomic analyses in MELAS face sensitivity and sample volume limitations, particularly for carboxylic acid metabolites. This study employed a recently established diazo-carboxyl/hydroxylamine-ketone double-click derivatization (DQmB-HA) mass spectrometry method to overcome these barriers, enabling highly sensit

Synthesis and PASS-assisted evaluation of new heterocyclic compounds containing hydroquinoline scaffolds.

Currently, there is a growing interest in the synthesis of heterocyclic compounds containing hydroquinoline fragments. This surge can be attributed to the broad range of pharmaceutical and industrial applications that these compounds possess. In this study, the synthesis of both linear and fused heterocyclic systems that incorporate hydroquinoline fragments was described. Furthermore, the pharmacological activity spectra of the synthesized compounds were predicted using the in silico method, employing the Prediction of Activity Spectra of Substances (PASS) program. Hydroquinolines containing t
