The kynurenine pathway in RNA virus infection: immunometabolic control of antiviral defense and viral persistence.

RNA virus infections are shaped by the interplay among viral replication, innate immune sensing, and host metabolism, which together determine the magnitude, duration, and quality of antiviral responses. The tryptophan-kynurenine (Trp-Kyn) pathway has emerged as an important immunometabolic axis linking interferon-driven inflammation, amino acid availability, immune-cell function, tissue homeostasis, and viral persistence. This review examines the roles of indoleamine 2,3-dioxygenase 1 (IDO1), IDO2, tryptophan 2,3-dioxygenase (TDO), Trp depletion, general control nonderepressible 2 (GCN2) sign

Bifurcation and optimal control analysis for a fractional-order model of drug-resistant HBV infection.

Hepatitis B Virus (HBV) remains a significant global health challenge, with control efforts complicated by the emergence of drug resistance and limitations in healthcare capacity. This study develops and analyzes a more realistic epidemiological model to investigate these complex dynamics and identify optimal intervention strategies. We formulate a novel six-compartment fractional-order model using the Caputo derivative, which incorporates memory effects, drug resistance, vertical transmission, and a crucial non-linear saturated treatment function to reflect healthcare system limitations. The

Natural Killer Cells Are Dispensable for Virus Control in Rag2-/- Mice During Primary RSV Infection.

Respiratory syncytial virus (RSV) is one of the major causes of severe lower respiratory tract infections, especially in children, the elderly, and immunocompromised individuals. Complications arising from viral infections in these age groups can present therapeutic challenges, as most of these individuals have impaired adaptive immunity. Using the T- and B cell-deficient Rag2-/- mice, the mechanisms that mediate protection in immunocompromised hosts during RSV infection can be investigated. RSV-infected Rag2-/- mice showed no symptoms of disease or chronic inflammation in the lungs and airway

Optimal control analysis of a mathematical model of malaria and COVID-19 co-infection dynamics.

In this paper, we analyze a deterministic model of malaria and Corona Virus Disease 2019 co-infection within a homogeneous population. We first studied the single infection model of each disease and then the co-infection dynamics. We calculate the basic reproduction number of each model and study the existence and stability of the steady states. Then, we show that under some suitable conditions, both the malaria single infection model and co-infection model exhibit backward bifurcation. Furthermore, we analyze the conditions of extinction, competitive exclusion and co-existence of these two di

Dual GSK-3β/HDAC Inhibitors Enhance the Efficacy of Macrophages to Control Mycobacterium tuberculosis Infection.

Multitarget drug discovery, including host-directed therapy, is particularly promising for tuberculosis (TB) due to the resilience of Mycobacterium tuberculosis (Mtb) as well as the complexity of the host's immune response. In this proof-of-concept study, we used high-content imaging to test a novel panel of dual glycogen synthase kinase 3 beta (GSK-3β) and histone deacetylase (HDAC) 1 and 6 inhibitor candidates for their efficacy in reducing the growth of green fluorescent protein (GFP)-expressing mycobacteria in human primary macrophages. We demonstrate that all ten test compounds, also incl

Infection prevention and control in 2030: a first qualitative survey by the Crystal Ball Initiative.

BACKGROUND: Healthcare delivery is undergoing radical changes that influence effective infection prevention and control (IPC). Futures research (short: Futures), the science of deliberating on multiple potential future states, is increasingly employed in many core societal fields. Futures might also be helpful in IPC to facilitate current education and organisational decisions. Hence, we conducted an initial survey as part of the IPC Crystal Ball Initiative. METHODS: In 2019, international IPC experts were invited to answer a 10-item online questionnaire, including demographics, housekeeping,
