Developmental exposure to the organochlorine insecticide endosulfan damages the nigrostriatal dopamine system in male offspring.

The contribution of environmental toxicants to the etiology and risk of Parkinson's disease (PD) has been clearly established, with organochlorine insecticides routinely shown to damage the nigrostriatal dopamine pathway. Although PD is generally considered an adult onset disease, it has been postulated that exposure to environmental contaminants or other factors early in life during critical periods of neurodevelopment could alter the dopaminergic circuit and predispose individuals to developing PD. Recent epidemiological evidence has found exposure to the organochlorine insecticide endosulfa…

Synthesis of the insecticide prothrin and its analogues from biomass-derived 5-(chloromethyl)furfural.

Prothrin, a synthetic pyrethroid insecticide, was synthesized from the biomass-derived platform chemical 5-(chloromethyl)furfural in six steps and overall 65% yield. Two structural analogues of prothrin were also prepared following the same synthetic approach. Preliminary testing of these furan-based pyrethroids against the yellow fever mosquito Aedes aegypti indicates promising insecticidal activities.

Insight into dihalogenation of E-ring of podophyllotoxins, and their acyloxyation derivatives at the C4 position as insecticidal agents.

Unexpected sequential E-ring dihalogenation of podophyllotoxin analogues is reported. It demonstrated that a chlorine/bromine atom was prior introduced at the C2' position of podophyllotoxin, and the corresponding free rotation of E-ring around the C1-C1' bond of 2'-chloro or 2'-bromopodophyllotoxin was restricted. When 2'-chloro or 2'-bromopodophyllotoxin reacted with N-chlorosuccinimide (NCS), the chlorine atom was regioselectively introduced at their C6' position on the E-ring. Whereas 2'-chloro or 2'-bromopodophyllotoxin reacted with NBS, the bromine atom was regioselectively introduced at

Insecticidal and repellent activity of Hiptage benghalensis L. Kruz (Malpighiaceae) against mosquito vectors.

Plant-based insecticides for vector control are urgently needed for Anopheles barbirostris, Culex quinquefasciatus, and Aedes albopictus which are the primary vectors of malaria, lymphatic filariasis, and dengue, respectively, in India and other South East Asian countries. In the present study, larvicidal, adulticidal, and repellent activities of acetone root bark extract of Hiptage benghalensis were tested against the larvae and adults of the three mosquito vectors. The acetone root bark extracts of H. benghalensis was more effective as larvicides with low LC(50) (11.15-16.78 ppm) and LT50 (1…

Insecticidal and repellent effects of tea tree (Melaleuca alternifolia) oil against Lucilia cuprina.

Laboratory studies were conducted to assess the effect of tea tree oil (TTO) from Melaleuca alternifolia (terpinen-4-ol chemotype) against different stages of the Australian sheep blowfly Lucilia cuprina. When applied to wool, 3% TTO formulation repelled gravid female L. cuprina and prevented oviposition for six weeks. Formulations containing 1% TTO caused 100% mortality of L. cuprina eggs and 1st instar larvae and 2.5% TTO caused mortality of most second and third instar larvae in agar feeding assays. In experiments where third instar larvae were dipped in TTO formulations for 60s, concentrat

Insecticidal peptides from the theraposid spider Brachypelma albiceps: an NMR-based model of Ba2.

Soluble venom and purified fractions of the theraposid spider Brachypelma albiceps were screened for insecticidal peptides based on toxicity to crickets. Two insecticidal peptides, named Ba1 and Ba2, were obtained after the soluble venom was separated by high performance liquid chromatography and cation exchange chromatography. The two insecticidal peptides contain 39 amino acid residues and three disulfide bonds, and based on their amino acid sequence, they are highly identical to the insecticidal peptides from the theraposid spiders Aphonopelma sp. from the USA and Haplopelma huwenum from Ch

Ivermectin as a rodent feed-through insecticide for control of immature sand flies (Diptera: Psychodidae).

Ivermectin was evaluated as a potential rodent feed-through for the control of immature stages of Phlebotomus papatasi. The survival of sand fly larvae fed feces of Syrian hamsters (Mesocricetus auratus) that had been fed a diet containing 0, 2, 6, 10, 20, 60, or 100 ppm ivermectin was measured. Sand fly larvae fed the feces of ivermectin-treated hamsters had significantly reduced survival, with 100% mortality of larvae fed feces of hamsters fed a diet containing 20, 60, and 100 ppm ivermectin. The results of this study suggest that a control strategy using rodent baits containing ivermectin t

Community cooperatives and insecticide-treated materials for malaria control: a new experience in Latin America.

BACKGROUND AND OBJECTIVES: Insecticide-treated materials (ITMs) are effective in substantially reducing the burden of malaria and other vector-borne diseases; but how can high coverage rates of ITMs be achieved and maintained? In south Mexico and on the Pacific and Atlantic coasts of Colombia 14 community-based cooperatives offering three different kinds of ITM services (sale of impregnation services; sale of impregnated nets; production of nets and sale of impregnated nets) were formed and supervised by a national health service (IMSS-SOLIDARIDAD, Mexico) and by an academic institution (the C

Molecular cloning of cDNAs encoding insecticidal neurotoxic peptides from the spider Phoneutria nigriventer.

From a Phoneutria nigriventer venom gland cDNA library several clones coding for the insect specific neurotoxin Tx4(6-1) were isolated. cDNA analysis showed that the encoded protein contained three distinct segments, comprising a signal sequence of 16 amino acids, followed by a glutamate-rich sequence of 18 amino acids and, finally, the coding region for the mature toxin. The deduced amino acid sequence for the mature polypeptide was identical to the protein sequence determined chemically. In addition, two new putative toxins called Pn4A and Pn4B were characterized and their predicted complete

Increased transcription of CYP6D1 causes cytochrome P450-mediated insecticide resistance in house fly.

Insecticide resistance is a major problem that continues to plague efforts to control pests of animals and crops. An important mechanism by which insects become resistant to insecticides is via increased detoxification mediated by the cytochrome P450 microsomal monooxygenases (monooxygenases). One of the fundamental gaps in our knowledge about this resistance mechanism is an understanding of how insects express high levels of the specific cytochrome P450(s) responsible for resistance. One such P450, CYP6D1, causes resistance to pyrethroid in the house fly and is expressed at 9-fold higher leve…

Solution structures of a highly insecticidal recombinant scorpion alpha-toxin and a mutant with increased activity.

The solution structure of a recombinant active alpha-neurotoxin from Leiurus quinquestriatus hebraeus, Lqh(alpha)IT, was determined by proton two-dimensional nuclear magnetic resonance spectroscopy (2D NMR). This toxin is the most insecticidal among scorpion alpha-neurotoxins and, therefore, serves as a model for clarifying the structural basis for their biological activity and selective toxicity. A set of 29 structures was generated without constraint violations exceeding 0.4 A. These structures had root mean square deviations of 0.49 and 1.00 A with respect to the average structure for backb

Prevalence of and potential risk factors for symptoms associated with insecticide use among animal groomers.

Pet groomers make numerous insecticide applications during the flea season, but few studies have examined their health complaints. The Pesticide Control Program of the New Jersey Department of Environmental Protection conducted a health and safety survey of this population. All licensed pet applicators in New Jersey were contacted, as were New Jersey veterinarians listed as pet-animal practitioners by the American Veterinary Medical Association. Approximately 36% of the respondents indicated that during the 1994 flea season, they had experience at least one of the 17 symptoms associated with i

Indoor spraying with the pyrethroid insecticide lambda-cyhalothrin: effects on spraymen and inhabitants of sprayed houses.

In March 1990 a study was carried out in the village of Kicheba, United Republic of Tanzania, in which the pyrethroid insecticide lambda-cyhalothrin was sprayed on all the internal surfaces of houses and other shelters at a coverage of about 25 mg of active ingredient per m2. Every day for 6 days, 12 spraymen and 3 squad-leaders were interviewed about symptoms of overexposure to the insecticide. Each sprayman used up to 62 g of lambda-cyhalothrin over 2.7-5.1 hours every day. All the spraymen complained at least once of symptoms that were related to exposure to lambda-cyhalothrin, the commones

Effect of subacute insecticide exposure on body weight, drug responses & electrical convulsions in mice.

Effect of subacute insecticide exposure was studied in male albino mice treated with phosphamidon, propoxur or aldrin at 1/40 LD50 dose intr … eased. The observed changes may be due to the induction of hepatic drug metabolising enzymes by the insecticides. The study suggests that certain dose adjustment of drugs may be necessary in those exposed to insecticides over long periods.

Gluconeogenic compensation renders glycogen phosphorylase non-essential as an insecticidal target in Plutella xylostella.

Glycogen phosphorylase (GP) catalyzes the rate-limiting step of glycogenolysis and supplies precursors for chitin biosynthesis in insects. Acyl urea compounds structurally related to benzoylphenylurea (BPU) insecticides are potent inhibitors of mammalian GP, raising the question of whether insect GP could serve as an independent insecticidal target. Here, we show that a mammalian GP inhibitor (GPI) potently inhibits recombinant Plutella xylostella GP (PxGP; IC50=2.96 nM), while the BPU insecticide diflubenzuron does not. Molecular docking and MM/GBSA analysis predict that this selectivity refl…

Design, synthesis and insecticidal activities of novel meta-diamide compounds containing ethyl group.

To develop structurally novel and efficient insecticides, a series of ethylpyridine-containing meta-diamide derivatives were designed and synthesized using cyproflanilide as the lead compound. The core bioactive scaffold was retained while integrating active substructure combination and pharmacophore fusion strategies. All compounds were characterized by ¹H NMR, ¹³C NMR, 19F NMR and HRMS. Preliminary bioassay data showed that some target compounds exhibited good insecticidal activity against Plutella xylostella, Mythimna separata, and Tetranychus cinnabarinus. Particularly, compound 7 g exhibi…

Eco-friendly-synthesized chitosan-zinc oxide-selenium nanocomposites: characterization and insecticidal activity.

In this work, the chitosan-zinc oxide-selenium nanocomposites (Ch-ZnO-Se NCs) were prepared in an affordable and eco-friendly way. Throughout the synthesis stage, chitosan served as reducing and stabilising agent. Different analytical techniques were employed to characterize the resulting nanocomposites. The findings demonstrated that irregular spherical-nanoparticles with a mean diameter of 73.95 nm were the predominant feature of the generated Ch-ZnO-Se NCs. The present study demonstrates that the mosquito vector Culex pipiens exhibit larvicidal and adulticidal effects upon exposure to the g
