Adrenomedullary Organoids Derived from Neural Crest and Schwann Cell Precursors.

The adrenal gland, essential for survival, contains the medulla and cortex. Chromaffin cells (ChrCs) in the adrenal medulla control the fight-or-flight response by releasing catecholamines like epinephrine and norepinephrine. Recent findings reveal that ChrCs originate from two progenitors: 20% from developmentally early, neural crest-derived sympathoadrenal progenitors (SAPs) and 80% from the later Schwann cell precursors (SCPs). We generated functional adrenomedullary organoids from human pluripotent stem cells (hPSCs) via both pathways and in mixed (20% SAP, 80% SCP-derived) cultures. When

Mechanistic insights into the efficient degradation and detoxification of chloramphenicol by Clostridium pasteurianum DSM 525.

Chloramphenicol (CAP), a chlorinated nitroaromatic antibiotic, posed significant ecological and human health risks due to its persistence and toxicity. Although microbial degradation represented a promising remediation strategy, its practical application was often constrained by slow degradation kinetics and incomplete biotransformation. In this study, rapid and efficient CAP degradation by the Gram-positive anaerobe Clostridium pasteurianum DSM 525 was demonstrated. The strain achieved 83.6 % removal of 400 mg L-1 CAP within 8 h, and the degradation kinetics followed a first-order model (R2 ≥

Characterization of atmospheric cadmium deposition and its simulated combined toxicological effects on wheat (Triticum aestivum L.) seedlings with sulfamethoxazole in soil.

Cadmium (Cd) accumulation caused by atmospheric deposition and combined Cd-sulfonamides stress via different input pathways in agricultural ecosystems warrants more attention. Based on a 2-year (2019-2020) observation of atmospheric Cd deposition, the effects of varying simulated Cd fluxes, different soil sulfamethoxazole (SMX) contents and their joint exposure on wheat (Triticum aestivum L.) seedlings were systematically investigated. Atmospheric Cd deposition was prominent in the region during the winter-to-spring and autumn-to-winter transitions (5 months, accounting for nearly 60 % of the

Multifunctional nano-platform for precision treatment of acute lung injury via synergistic anti-inflammation and oxidative stress modulation.

Acute lung injury (ALI) remains life-threatening conditions lacking effective disease-modifying therapies. Dysregulated inflammation, oxidative stress, and apoptosis form a self-amplifying pathogenic loop that drives alveolar-capillary barrier disruption, highlighting the urgent need for multifunctional therapeutic strategies. Here, we report a pH-responsive biomimetic nanoplatform, pDA/Esc@ZIF-8, for precision treatment of ALI. Zeolitic imidazolate framework-8 (ZIF-8) was employed to encapsulate the natural anti-inflammatory and antioxidant agent esculin (Esc), while a polydopamine (pDA) coat

Neuroendocrine interactions of the stress and reproductive axes.

Reproduction is controlled by a sequential regulation of the hypothalamo-pituitary-gonadal (HPG) axis. The HPG axis integrates multiple inputs to maintain proper reproductive functions. It has long been demonstrated that stress alters fertility. Nonetheless, the central mechanisms of how stress interacts with the reproductive system are not fully understood. One of the major pathways that is activated during the stress response is the hypothalamo-pituitary-adrenal (HPA) axis. In this review, we discuss several aspects of the interactions between these two neuroendocrine systems to offer insigh

Sympatric European white oaks species display contrasting epigenetic response to soil water availability.

In the context of climate change plants have to cope with adverse conditions, among which water scarcity is a major threat for their survival. They have developed diverse regulatory processes to face drought that may differ depending on their ecological niche. The two sympatric oaks species (Quercus robur L. and Quercus petraea (Matt.). Liebl) have different levels of drought tolerance. We have investigated their strategies to face drought stress by analysing the transcriptome, small RNAome and methylome dynamics of young plants grown under control and drought stress conditions. Data indicate

Crosstalk between Melatonin and Reactive Oxygen Species in Plant Abiotic Stress Responses: An Update.

Melatonin acts as a multifunctional molecule that takes part in various physiological processes, especially in the protection against abiotic stresses, such as salinity, drought, heat, cold, heavy metals, etc. These stresses typically elicit reactive oxygen species (ROS) accumulation. Excessive ROS induce oxidative stress and decrease crop growth and productivity. Significant advances in melatonin initiate a complex antioxidant system that modulates ROS homeostasis in plants. Numerous evidences further reveal that melatonin often cooperates with other signaling molecules, such as ROS, nitric o

Multi-omics analyses to assess the effects of multi-layer cultivation with supplemental LED lighting on potato yield and quality.

Integrating a vertical layout into greenhouse allows space-efficient cultivation and energy saving; however, shading of the lower canopy makes supplemental artificial lighting necessary. Here, we investigated the effect of red-blue LED supplementation in a vertical greenhouse system for potato cultivation on plant growth, tuber yield, and mineral and metabolite composition, compared to full natural sunlight as a control. Cultivation units were designed to simulate a multiple-layer growing system, in which the uppermost level received only natural light, while LEDs integrated reduced light inte

Shifts in hormonal stress reactivity during adolescence are not associated with changes in glucocorticoid receptor levels in the brain and pituitary of male rats.

Preadolescent animals display protracted hormonal stress responses mediated by the hypothalamic-pituitary-adrenal (HPA) axis compared to adults. Though the mechanisms that underlie this shift in stress reactivity are unknown, reduced glucocorticoid-dependent negative feedback on the HPA axis has been posited to contribute to this differential responsiveness. As the glucocorticoid receptors (GRs) are integral to this feedback response, we hypothesize that prior to puberty there will be fewer GRs in the neural-pituitary network that mediate negative feedback. To test this hypothesis we measured

In Silico Perturbome Analysis Reveals Conserved Genes and Drug-Target Interactions in Pseudomonas aeruginosa, Escherichia coli, and Staphylococcus aureus in the Response to Stress.

BACKGROUND: Bacterial adaptation to environmental and chemical stress involves coordinated, system-level responses collectively described as perturbome. Understanding conserved elements within core perturbomes may reveal strategic vulnerabilities for antimicrobial development. METHODS: In this study, we implemented an integrative framework combining functional and comparative genomics, drug-target interactions and molecular docking to prioritize conserved stress-response targets in Escherichia coli, Pseudomonas aeruginosa, and Staphylococcus aureus. RESULTS: A total of 147 genes from previou

Chemical stress drives opposing bacterial biomass responses at the single-cell level.

Microbial biomass is a central functional trait governing nutrient turnover and trophic transfer in aquatic ecosystems, yet how chemical stress reshapes biomass at the level of individual cells remains poorly resolved. Conventional toxicity assays primarily detect population-level growth inhibition and overlook sublethal physiological responses and heterogeneity within microbial populations. Here, we quantify single-cell dry-mass, a label-free proxy for cellular biomass and biosynthetic state, in the fast-growing marine bacterium Vibrio natriegens. Across thousands of cells, we assessed how ex

Integrating Senescence and Oxidative Stress in Cardiac Disease.

Cellular senescence and oxidative stress constitute an interdependent axis that underlies cardiac pathophysiology. Cellular senescence, defined as durable proliferative arrest, is initiated and sustained by redox imbalance, whereas mitochondrial reactive oxygen species function as signaling molecules or mediators of injury. In the heart, cellular senescence and oxidative stress influence remodeling and dysfunction across diseases, including ischemia-reperfusion injury, heart failure with preserved ejection fraction, dilated cardiomyopathy, and cardiac hypertrophy. Accordingly, delineating stre

Physiological Function during Exercise and Environmental Stress in Humans-An Integrative View of Body Systems and Homeostasis.

Claude Bernard's milieu intérieur (internal environment) and the associated concept of homeostasis are fundamental to the understanding of the physiological responses to exercise and environmental stress. Maintenance of cellular homeostasis is thought to happen during exercise through the precise matching of cellular energetic demand and supply, and the production and clearance of metabolic by-products. The mind-boggling number of molecular and cellular pathways and the host of tissues and organ systems involved in the processes sustaining locomotion, however, necessitate an integrative examin

A Genome-Wide Search for Ionizing-Radiation-Responsive Elements in Deinococcus radiodurans Reveals a Regulatory Role for the DNA Gyrase Subunit A Gene's 5' Untranslated Region in the Radiation and Desiccation Response.

Tight regulation of gene expression is important for the survival of Deinococcus radiodurans, a model bacterium of extreme stress resistance. Few studies have examined the use of regulatory RNAs as a possible contributing mechanism to ionizing radiation (IR) resistance, despite their proffered efficient and dynamic gene expression regulation under IR stress. This work presents a transcriptome-based approach for the identification of stress-responsive regulatory 5' untranslated region (5'-UTR) elements in D. radiodurans R1 that can be broadly applied to other bacteria. Using this platform and a

Lysosomal TMEM165 remodels calcium signaling to drive hypoxia adaptation and tumor progression.

Hypoxia is a common stress encountered by animal tissues during development, physiology, and disease. To cope with hypoxic stress, cells remodel metabolic and signaling networks to preserve viability and function. Lysosomes serve as central hubs for metabolic control and intracellular signaling, yet their role in hypoxic adaptation remains unclear. Here, we identify the lysosomal calcium transporter TMEM165 as a hypoxia-responsive regulator of cellular homeostasis. Under hypoxic conditions, TMEM165 expression increases, promoting calcium redistribution from the endoplasmic reticulum to lysosom

A role for glucocorticoids in stress-impaired reproduction: beyond the hypothalamus and pituitary.

In addition to the well-characterized role of the sex steroid receptors in regulating fertility and reproduction, reproductive events are also mediated by the hypothalamic-pituitary-adrenal axis in response to an individual's environment. Glucocorticoid secretion in response to stress contributes to the well-characterized suppression of the hypothalamic-pituitary-gonadal axis through central actions in the hypothalamus and pituitary. However, both animal and in vitro studies indicate that other components of the reproductive system are also regulated by glucocorticoids. Furthermore, in the abs

Role of Esterase BmeFE4 in Mediating the Alleviating Effects of Resveratrol on NaF-Induced Fluorosis in Bombyx mori.

Fluoride exposure can induce tissue injury and oxidative stress dysregulation, ultimately leading to organismal dysfunction and death, whereas resveratrol (RSV) has been reported to counteract diverse oxidative injuries. Despite its antioxidant potential, the application of RSV is limited by its low bioavailability. Here, using a silkworm model, we established NaF exposure, RSV treatment, and combined RSV+NaF groups to investigate the mechanism by which RSV attenuates fluorosis. Comparative analyses of body weight, histopathological damage, and oxidative stress indices showed that RSV markedly

Long-term imoxin treatment does not attenuate disease severity in dystrophic diaphragm of male mice.

Duchenne muscular dystrophy (DMD) is characterized by chronic skeletal muscle injury and degeneration. We discovered activation of protein kinase R (PKR; EIF2AK2), a regulator of the integrated stress response, in dystrophic muscle; however, its role in DMD remains unknown. We hypothesized that the PKR inhibitor imoxin (IMX) would improve muscle force and attenuate fibrosis, inflammatory signaling, and ER stress in diaphragms from mdx mice. C57 and mdx mice were treated with vehicle or IMX for ~24 wks (0.5 mg/kg, subcutaneous, 5 times/wk). Specific force was decreased (p < 0.001), and dynamic …

Beyond the Leash: Exploring the Public Health Dimension of Facility Dog Handler Experiences in Military Graduate Education.

Facility dogs are increasingly incorporated throughout higher-education institutions as integrative conduits for supporting students by reducing stress, anxiety, and emotional exhaustion. While these programs are designed to enhance student well-being, little is known about the experiences of the handlers who train, care for, and work with these dogs-specifically within military-affiliated programs. The handlers' role necessitates a unique dynamic of caregiving, emotional labor, and professional responsibility and management that can create role conflict and additional stress among those gradu
