Evogliptin Directly Inhibits Inflammatory and Fibrotic Signaling in Isolated Liver Cells.

Chronic liver inflammation can lead to fibrosis, cirrhosis, and hepatocellular carcinoma. Kupffer cells (KC) secrete proinflammatory and fibrogenic cytokines in response to lipopolysaccharide (LPS), and so play an important role in liver inflammation, where they induce hepatocellular damage. LPS also activates hepatic stellate cells and induces extracellular matrix deposition. In this study, we used isolated primary KC, primary hepatocytes, and primary hepatic stellate cells (HSC) to investigate whether evogliptin directly inhibits inflammatory and fibrotic signaling. We found that evogliptin …

MicroRNA Interference in Hepatic Host-Pathogen Interactions.

The liver is well recognized as a non-immunological visceral organ that is involved in various metabolic activities, nutrient storage, and detoxification. Recently, many studies have demonstrated that resident immune cells in the liver drive various immunological reactions by means of several molecular modulators. Understanding the mechanistic details of interactions between hepatic host immune cells, including Kupffer cells and lymphocytes, and various hepatic pathogens, especially viruses, bacteria, and parasites, is necessary. MicroRNAs (miRNAs), over 2600 of which have been discovered, are…

Expression of pleiotrophin in hepatic nonparenchymal cells and preneoplastic nodules in carbon tetrachloride-induced fibrotic rat liver.

Pleiotrophin (PTN) is a heparin-binding protein, which induces growth, angiogenesis, differentiation, and transformation of cells. The aim o … ochemistry localized PTN protein on hepatic nonparenchymal cells, mostly stellate cells and some of Kupffer cells, and the preneoplastic nodules in fibrotic liver tissues. PTN mRNA expression is significantly upregulated in the CCl4-induced chronic rat fibrotic liver tissues. We suggest that PTN might be involved in fibrogenesis and preneoplastic changes of liv …

Copper toxicosis in 3 Japanese Black cattle with chronic cholangiohepatitis.

Three Japanese Black cattle raised in different farms died suddenly with severe jaundice and hemoglobinuria. Gross examination revealed diffuse hepatic discoloration and highly viscous bile in the gallbladder. Histologically, the livers showed chronic cholangiohepatitis with hepatocellular vacuolation, bile plugs, interlobular bile duct hyperplasia, and periportal fibrosis. Alcian blue-periodic acid-Schiff staining showed acidic mucin in the bile ducts. Rhodanine staining and metallothionein immunohistochemistry demonstrated copper accumulation mildly in hepatocytes and mainly in Kupffer cells…

Human fatal yellow fever. Immunohistochemical detection of viral antigens in the liver, kidney and heart.

An immunohistochemical method to detect yellow fever antigen was developed using immune sera from rabbits and hamsters and hyperimmune ascitic fluid from mice. A search for the antigen was carried out in liver, kidney and heart in three fatal cases of yellow fever. In the liver it was present in the cytoplasm of hepatocytes, Councilman bodies and Kupffer cells. Yellow fever antigen was also detected in renal tubular epithelium and in groups of myocardial fibers. These findings suggest that viral replication occurs at sites other than the liver. Since yellow fever shares many features with othe…

Hermansky-Pudlak syndrome with special reference to lysosomal dysfunction. A case report and review of the literature.

In addition to the triad in the Hermansky-Pudlak syndrome (tyrosine-positive oculocutaneous albinism, mild bleeding tendency with a normal platelet-count and widespread accumulation of ceroid-like pigment in various organs), we document severe pulmonary fibrosis, pseudomelanosis coli and deeply pigmented renal cortex. In the liver, innumerable number of pigment-laden Kupffer cells and macrophages in the Glisson capsule were seen. Interestingly, many intralysosomal accumulations of the pigment within the hepatocytes were found by electron microscopy, suggesting that these configurations possibl…

Gadolinium chloride-induced hepatocyte proliferation is prevented by antibodies to tumor necrosis factor alpha.

Gadolinium chloride (GdCl(3)) destroys large Kupffer cells and has been used extensively in mechanistic studies in a number of disease and toxicity processes; however, it cannot be used to study hepatocyte turnover since it increases cell proliferation itself. The mechanism by which GdCl(3) activates cell turnover in liver is unknown, but several possibilities exist. Here it was demonstrated that a direct mitogenic action on hepatocytes is unlikely since GdCl(3) did not stimulate the growth of primary rat hepatocyte in vitro. Therefore, it was hypothesized that GdCl(3) acts indirectly through …

Cellular uptake studies indicate that LNPs prefer to reside in Kupffer cells, leading to prolonged and delayed hepatotoxicity even after the cessation of NPs administration, while SNPs have much less Kupffer cell uptake. Advances in the research of nanodrug delivery system for targeted treatment of liver fibrosis.

Liver fibrosis is one of the major threats to human health. At present, anti-liver fibrosis drugs exist some problems, such as, lack of targeting, limited treatment effect and more or less toxic side effects. In order to improve the efficacy of targeted therapy for liver fibrosis, the development of a large number of nanocarriers and strategies of targeted therapy for liver fibrosis have been widely studied. In this paper, the research progress of nano-delivery vectors targeting cells related to the process of liver fibrosis in recent years was reviewed in terms of targeting vectors and the typ

Single Cell Analysis Reveals the Presence of Novel Intermediate Cells in Both Mice and Patients With Severe MASLD.

The landscape of cellular abnormalities occurring during metabolic dysfunction-associated steatotic liver disease (MASLD) is not completely clarified. We investigated cell heterogeneity involved in progressive MASLD at single cell resolution in mice fed AMLN. Single cell RNA sequencing (Sc-RNAseq) and spatial proteomics were applied to decipher cell populations and genes/pathways guiding MASLD progression. We identified 32 clusters, including hepatocytes (HEPs), hepatic stellate cells (HSCs), endothelial cells (ENDOs), Kupffer cells (KCs), and immune cells. HEPs clusters changed across disease…

Early effects of high doses of retinol (vitamin A) on the in situ cellular metabolism in rat liver.

Understanding of the possible toxicity associated with hypervitaminosis A becomes increasingly important in view of the popularity of vitamin A supplementation. Hypervitaminosis A for many years may eventually lead to hepatocellular damage. In the present study, rats were treated for 7 days with high doses of retinol to study the early effects on the metabolism of different types of liver cells using (enzyme) histochemistry, immunohistochemistry and electron microscopy. Excessive intake of vitamin A activates Kupffer cells and induces accumulation of lipid droplets in fat-storing cells as well…

Hepatic changes during short-term haemodialysis.

Twenty five patients with end stage renal disease were studied to find out the biochemical and hepatic morphological changes during short term haemodialysis. Asymptomatic hepatomegaly was seen in 32% of cases, transaminase elevation in 28% and positive Australia antigen in 12% of cases. Histopathological changes were observed in all the 12 patients in whom liver biopsy was done. Ten patients (82%) exhibited multiple abnormalities. The commonest findings were fatty change (8 patients); hepatic congestion, focal necrosis, Kupffer cell hyperplasia (6 patients each); and portal triaditis (5 patien

Size-dependent plastic exposure disrupts macrophage function and tissue-specific metabolism.

Plastic pollution is an emerging yet understudied environmental risk to the immune system. Once ingested, microplastic and nanoplastic particles (MNPs) can translocate from the gut to internal organs1-3, with macrophages serving as primary targets4. Kupffer cells (KCs), the liver-resident macrophages, have a central role in immune surveillance and metabolism5, yet their response to MNPs remains unclear. Here we identify KCs as the primary hepatic reservoir for MNPs in young male mice. Chronic plastic exposure over 12 weeks alters their transcriptional profile, impairs phagocytic function and i…
