This study investigates the interactions between ivermectin (IVM) and lipid membranes with varying cholesterol contents by using a combined molecular dynamics (MD) and experimental approach. DOPC bilayers containing 0, 10, 20, or 30% cholesterol were simulated, and SPC liposomes were employed for experimental validation. Mass density profiles indicated that the membrane thickness increased from 4.16 nm (0% cholesterol) to 4.60 nm (30% cholesterol), while ivermectin was most deeply embedded in membranes with 10% cholesterol with an average distance of 1.09 nm from the bilayer center. van der Wa

Lemon-derived extracellular vesicles (LDEVs) have attracted increasing attention as promising bioactive nanovesicles for cosmetic and skin engineering applications owing to their biocompatibility, bioactive cargo, and skin-friendly lipid bilayer structure. However, the lack of standardized purification methods and limited scalability remain critical bottlenecks for the practical application and industrial translation of LDEVs. In this study, we developed an electrophoretic oscillation-assisted tangential flow filtration (EPOTF) platform using a SiNx membrane nanofilter for the scalable and hig

Current treatments for inflammatory bowel disease often fail due to systemic side effects, but bovine milk-derived extracellular vesicles (EVs) show promise for targeted delivery to inflamed gut tissue via the leaky gut effect. This study assessed the stability of EVs as drug carriers in simulated gastrointestinal (GI) fluids and their efficacy in a colitis mouse model. EVs were characterised after incubation in PBS at various pH levels, and their lipid bilayer stability in biorelevant GI fluids was evaluated using the polar probe laurdan. Two small molecules, acridine orange (lipophilic) and

The clinical application of antimicrobial peptides (AMPs) is frequently hindered by the inherent limitations of linear peptides. Previous studies have primarily focused on the physicochemical properties of AMPs, and there is a scarcity of information regarding the transmembrane structure and interactions of AMPs with cell membranes and their antimicrobial activity. The present study is the first to propose that the backbone cyclization of linear R3W4V (l(R3W4V)) into the cyclic R3W4V (c[R3W4V]) form can enhance the stability of its transmembrane structure and consequently improve its antibacte

Liposomal formulations remain mostly suited for amphiphilic drugs of mild potency. This high selectivity challenges the formulation of potent chemotherapeutics that are preponderantly hydrophobic. Spontaneous accumulation of certain hydrophobic drugs within the lipid bilayer of liposomes leads to aggregation, destabilization, and inadequate drug retention. Here, we depict an alternative approach to load such hydrophobic drugs using SN38, the active form of CPT-11 (Irinotecan, Camptosar), through SN38 glucuronide (SN38-G), its hydrophilic glucuronide prodrug derivative. SN38-G was esterified in

Enveloped viruses, such as flaviviruses and coronaviruses, are pathogens of significant medical concern that cause severe infections in humans. Some photosensitizers are known to possess virucidal activity against enveloped viruses, targeting their lipid bilayer. Here we report a series of halogenated difluoroboron-dipyrromethene (BODIPYs) photosensitizers with strong virus-inactivating activity. Our structure-activity relationship analysis revealed that BODIPY scaffolds with a heavy halogen atom demonstrate significant efficacy against both tick-borne encephalitis virus (TBEV; Flaviviridae fa

An unacceptable bitter taste limits the application of luteolin in healthier food systems. In this study, a bitterness-masking assessment was performed on whey protein isolate-coated liposomes loaded with luteolin (WPI-coated liposomes) using an electronic tongue and human sensory test. The physical properties of the WPI-coated colloidal nanocarrier were characterized by zeta potential, average diameter, distribution, and morphology analyses. The results indicated that WPI-coated nanocarrier systems exhibited a uniformly dispersed distribution and spherical morphology. After the comparison of

Glycosaminoglycans (GAGs) are long and unbranched anionic heteropolysaccharides that have been associated with virtually all amyloid deposits. Soluble sulfated GAGs are known for their propensity to promote the self-assembly of numerous amyloidogenic proteins and to modulate their cytotoxicity. Nonetheless, although GAGs are prevalent on the outer leaflet of eukaryotic cell plasma membrane as part of proteoglycans, their contributions in the perturbation of lipid bilayer induced by amyloid polypeptides remain unknown. Herein, we investigate the roles of GAGs in the cytotoxicity and plasma memb

Due to their tunable optical properties and their well-defined nanometric size, core/shell nanocrystals (quantum dots, QDs) are extensively used for the design of biomarkers as well as for the preparation of nanostructured hybrid materials. It is thus of great interest to understand their interaction with soft lipidic membranes. Here we present the synthesis of water-soluble peptide CdSe/ZnS QDs and their interaction with the fluid lipidic membrane of vesicles. The use of short peptides results in the formation of small QDs presenting both high fluorescence quantum yield and high colloidal sta

Assembling structures to divide space controllably and spontaneously into subunits at the nanometer scale is a significant challenge, although one that biology has solved in two distinct ways: prokaryotes and eukaryotes. Prokaryotes have a single compartment delimited by one or more lipid-protein membranes. Eukaryotes have nested-membrane structures that provide internal compartments--such as the cell nucleus and cell organelles in which specialized functions are carried out. We have developed a simple method of creating nested bilayer compartments in vitro via the "interdigitated" bilayer pha

Rational development of new selective paramagnetic contrast agents (PCAs) requires a detailed understanding of their interactions with biolo … o addition of Gd3+ complexes. This work demonstrates that interaction of Gd3+ complexes with phospholipid bilayers can be observed directly from changes in their continuous wave (CW) EPR spectra obtained at frequencies higher than X-band (9.5 GHz), where signals arising from aqueous and lipid-bound Gd3+ complexes become resolved. Analysis of frequency dependence …

Mechanisms of proton transport were investigated in planar phospholipid bilayer membranes exposed to aspirin (acetylsalicylic acid), acetaminophen (4-acetamidophenol), benzoic acid and three aspirin metabolites (salicylic acid, gentisic acid and salicyluric acid). The objectives were to characterize the conductances and permeabilities of these weak acids in lipid bilayer membranes and then predict their effects on mitochondrial membranes. Of the compounds tested only aspirin, benzoate and salicylate caused significant increases in membrane conductance. The conductance was due mainly to proton

The separation of two photoreactive derivatives of the alpha-scorpion toxin from Leiurus quinquestriatus is described. When the two photoreactive derivatives were photolyzed separately in the presence of brain membranes containing voltage-sensitive sodium channels, one labeled the alpha subunit preferentially while the other labeled beta 1 more intensely than alpha. Batrachotoxin enhanced the efficiency of covalent labeling by the photoreactive derivatives of scorpion toxin. In all the labeling experiments, the specific incorporation of radioactive scorpion toxin was eliminated by an excess of

Chlorpromazine (CP), an amphipathic, antipsychotic agent, causes concave membrane bending in red blood cells with formation of stomatocytic shapes by modulation of the phospholipid bilayer. This study was designed to investigate the effects of CP on the shape of bovine aortic endothelial cells (BAEC) and their membranes in confluent monolayers with phase-contrast and transmission electron microscopy. Exposure of BAECs to nanomolar levels of CP leads to membrane curvature changes. With increasing CP concentrations, the membrane assumed a shape with enhanced numbers of intracellular caveolae and

Linear peptide antibiotics have been isolated from amphibians, insects and humans and used as templates to design cheaper and more potent analogues for medical applications. Peptides such as cecropins or magainins are < or = 40 amino acids in length. Many of them have been prepared by solid-phase peptide synthesis with isotopic labels incorporated at selected sites. Structural analysis by solid-state NMR spectroscopy and other biophysical techniques indicates that these peptide antibiotics strongly interact with lipid membranes. In bilayer environments they exhibit amphipathic alpha-helical co

Mycobacteria are responsible for causing severe illnesses like tuberculosis and leprosy in humans. Studying the mycobacteria cell envelope presents a significant challenge due to its intricate lipid compositions and structural variations and also its harmful nature in a typical experiment setting. In this study, we use all-atom molecular dynamics simulation to study mycobacterial inner membranes (MIMs). By incorporating different types of phosphatidyl-myo-inositol-mannosides (PIMs) and their glycoconjugates such as lipomannans (LM) and lipoarabinomannans (LAM) lipoglycans, we have constructed

The topology of membrane phospholipids in the red cells of patients with hereditary spherocytosis has been studied with the non-penetrating probe 2,3,5-trinitrobenzenesulphonate. There was no significant difference in the assymetric distribution of the aminophospholipids phosphatidylserine and phosphatidylethanolamine across the two halves of the membrane bilayer in red cells of five patients with hereditary spherocytosis as compared to seven normal controls. These studies indicated that the complex processes responsible for membrane lipid assymetry are intact in hereditary spherocytosis.

Chronic wound infections, particularly those complicated by antibiotic-resistant bacteria and biofilm formation, present persistent clinical challenges due to limited drug penetration and unresolved inflammation. Here, we report a lipid-droplet-engineered, gallium-loaded sonodynamic therapeutic platform (LDTG) incorporated into a bilayer microneedle (MN) patch co-delivering all-trans retinoic acid (ATRA), termed LDTG A-MNs, for synergistic antibacterial therapy and accelerated wound repair. The engineered LDTG exhibit strong bacteria-targeting capability by mimicking host lipid droplets and ef
