Protein-lipid interactions in food matrices and their roles in gastrointestinal lipid digestion.

Dietary proteins and lipids coexist in foods in diverse structural forms, and their interactions influence lipid digestion and the subsequent intestinal absorption of lipolysis products, including free fatty acids and monoacylglycerols. This review examines the role of protein-lipid interactions (PLIs) in lipid digestion, with emphasis on bile salt displacement, lipolysis, mixed micelle formation, and the transport of lipid-soluble nutrients. PLIs occur mainly at interfaces within food matrices and are further modulated by matrix organisation, processing-induced structural changes, and physiol…

Lipid Profiles and Lipid-Lowering Therapy at the Time of Acute Myocardial Infarction.

BACKGROUND: Low-density lipoprotein cholesterol (LDL-C) lowering with lipid-lowering therapy (LLT) is foundational for atherosclerotic cardiovascular disease (ASCVD) prevention. OBJECTIVES: The purpose of this study was to evaluate LDL-C levels and LLT at the time of acute myocardial infarction (AMI). METHODS: We retrospectively assessed LDL-C levels and LLT use at presentation and discharge among patients hospitalized with AMI from March 2018 to August 2022. RESULTS: Among AMI patients with no prior ASCVD diagnosis (n = 1,159), 94.7% had LDL-C ≥55 mg/dL, 86.7% had LDL-C ≥70 mg/dL, and 33.6

Tumour-driven lipid accumulation in oenocytes reflects systemic lipid alterations.

Cancer cachexia is a multifactorial syndrome characterized by systemic metabolic dysfunction, including liver steatosis. In this study, we e … id not significantly improve muscle integrity. Together, our data suggests that dynamic exchange of lipids occur between the fat body, oenocytes and the muscle during cancer cachexia. While the fat body and muscle lipid pools are key regulators of muscle integrity, oenocytes - despite their metabolic responsiveness, do not appear to play an active role i …

Selective targeting of kinesin on lipid droplets in the liver reduces plasma lipids.

The liver controls plasma lipids by secreting lipid-rich very low density lipoproteins (VLDL) into blood. Inside hepatocytes in the liver, Lipid Droplets (LDs) are transported to the Smooth Endoplasmic Reticulum by kinesin-1 motors, where they are catabolized to supply lipids for VLDL assembly. Here we find that kinesin-1 uses its tail domain to bind the monolayer phospholipid membrane of LDs, but alternative mechanisms to bind cellular organelles with bilayer membranes. A peptide corresponding to the tail domain of kinesin-1 therefore competes with and removes kinesin-1 selectively from LDs w…

Influence of ionisable lipid and sterol variations on lipid nanoparticle properties and performance.

Lipid nanoparticles (LNPs) have emerged as a vital delivery system for nucleic acids, particularly in the context of mRNA vaccines and therapeutics. This study evaluates the impact of various proprietary ionisable lipids on the physicochemical characteristics, encapsulation efficiency, and expression of mRNA within LNP formulations. We compared 11 novel ionisable lipids against the clinically used ALC-0315 in both in vitro and in vivo models. Alongside comprehensive physicochemical characterisation (including size, zeta potential, and polydispersity index), we uncovered significant variations …

Lipid profiling reveals unsaturated lipid reduction in women with Alzheimer's disease.

INTRODUCTION: Alzheimer's disease (AD) is a devastating neurological disease that disproportionately affects women. This study aimed to investigate sex-specific single lipids associated with AD. METHODS: Plasma samples from 841 participants, comprising 306 individuals with AD, 165 with mild cognitive impairment (MCI), and 370 cognitively healthy controls were curated from the AddNeuroMed cohort. Lipidomics identified 268 single lipids for each sample. We investigated sex-specific associations from lipid modules and single lipids to AD and probed for causality with mediation analyses. RESULTS…

Tailoring the adjuvanticity of lipid nanoparticles by PEG lipid ratio and phospholipid modifications.

Lipid nanoparticles (LNPs) represent the leading delivery platform for mRNA vaccines with advantageous biocompatibility, scalability, adjuvant activity and often an acceptable safety profile. Here we investigate the physicochemical characteristics and adjuvanticity of four-component LNPs. Previous vaccine studies have demonstrated that altering the ionizable lipid influences the adjuvanticity of an LNP; however, the impact of the polyethylene glycol lipid and phospholipid has received less attention. Our mRNA-LNP vaccine formulations utilized different phospholipids and varying ratios of polye…

Lipid droplet-associated lncRNA LIPTER preserves cardiac lipid metabolism.

Lipid droplets (LDs) are cellular organelles critical for lipid homeostasis, with intramyocyte LD accumulation implicated in metabolic disorder-associated heart diseases. Here we identify a human long non-coding RNA, Lipid-Droplet Transporter (LIPTER), essential for LD transport in human cardiomyocytes. LIPTER binds phosphatidic acid and phosphatidylinositol 4-phosphate on LD surface membranes and the MYH10 protein, connecting LDs to the MYH10-ACTIN cytoskeleton and facilitating LD transport. LIPTER and MYH10 deficiencies impair LD trafficking, mitochondrial function and survival of human indu

Present-day uses of niacin: effects on lipid and non-lipid parameters.

Existing guidelines for the prevention and treatment of coronary artery disease focus on lowering low-density lipoprotein cholesterol (LDL-C) as the primary lipid target. However, there has been increasing interest in raising high-density lipoprotein cholesterol (HDL-C) due to strong evidence linking low HDL-C levels with an increased risk of atherosclerosis. Raising HDL-C levels with lifestyle changes and pharmacologic interventions appear to reduce the risk of coronary artery disease beyond that of lowering LDL-C alone. Niacin has a substantial HDL-C raising effect, and also may beneficially

Lipid peroxide formation in microsomes. Relationship of hydroxylation to lipid peroxide formation.

1. Aminopyrine strongly inhibits NADPH-induced lipid peroxide formation in rat liver microsomes, but ascorbate-induced peroxidation is inhibited to a smaller extent. 2. Aminopyrine oxidation is stimulated by Mg(2+) but inhibited by Ca(2+). Concentrated solutions (10mm) of iron-chelating agents inhibit aminopyrine oxidation, but the more dilute solutions (0.5mm) of chelators that block lipid peroxide formation do not inhibit aminopyrine oxidation. Microsomes prepared from sucrose-EDTA homogenates rapidly oxidize aminopyrine, but do not form lipid peroxide when incubated with ascorbate or NADPH.

Fluorination-Tuned Ionizable Lipids Regulate Nano-PROTAC Trafficking for Enhanced Targeted Protein Degradation.

Nanoscale proteolysis-targeting chimeras (nano-PROTACs) offer modular platforms for targeted protein degradcine-rich α-2-glycoprotein 1 (LRG1). i16αCF3-LipoTAC showed greater degradation potend endocytosis, and promoted endosomal escape. In 4T1 tumor models, i16αCF3-LipoTAC prolonged circulation, increased tumor accumulation and target depletion, and improved antitumor efficacy. These findings support fluorination tuning as a strategy to enhance nano-PROTAC trafficking and degradation efficiency.Nanoscale proteolysis-targeting chimeras (nano-PROTACs) offer modular platforms for targeted protei

Differences in lipids and antibacterial activities among three chemotypes of Cinnamomum camphora seeds.

Medium-chain triacylglycerols confer health benefits to humans by virtue of their medium-chain fatty acids (MCFAs). To promote the utilization of different chemotypes of Cinnamomum camphora seed lipids in food industry, we investigated the seed morphologies, lipid content, fatty acid compositions of three main chemotypes and antibacterial activities of the lipid extracts. Among camphor (Cam), linalool (Lin) and eucalyptol (Eul) chemotypes, Cam seeds showed large size and great mass, followed by Lin and then Eul seeds. Seeds of the three chemotypes contained storage lipids rich in MCFAs (mainly…

NTA-Ni lipids affect membrane phase behavior and fluorescence of lipid-like dyes.

The nitrilotriacetic acid group coordinated with nickel, NTA(Ni), is the standard chelator for purifying fusion proteins containing poly-histidine tags. The development of lipids with NTA(Ni) headgroups has enabled the specific attachment of His-tagged peptides and proteins to model membranes. Recently, these lipids have become essential for the study of surface-mediated protein phase separation and membrane interaction with biomolecular condensates. While this approach is gaining popularity, the intrinsic effects of NTA(Ni) lipids on the properties of the host membrane have been very little s…
