NMDA preconditioning prevents object recognition memory impairment and increases brain viability in mice exposed to traumatic brain injury.

Recent studies have focused on the role of N-methyl-d-aspartate (NMDA) in brain injury. The present study is aimed at verifying memory, anxiety/depression parameters, and cellular viability in the brain of mice preconditioned with NMDA and subjected to the model of mild traumatic brain injury. For this purpose, male albino CF-1 mice were pre-treated with NMDA (75 mg/kg) and subjected to brain trauma, and after 24h submitted to memory tasks and anxiety and depression-like behavioral tests. The memory tests were evaluated at 1.5h, 24h, and 7 days after the training. In addition, the cellular via

Differential functional immune recall across homologous and heterologous COVID-19 vaccination regimens.

INTRODUCTION: Understanding how different COVID-19 vaccine combinations shape long-term immunity is essential for improving durability and guiding booster strategies. Despite extensive characterization of neutralizing antibodies, long-term memory B and T cell responses after homologous and heterologous vaccination regimens remain poorly understood. METHODS: In this study, peripheral blood mononuclear cells (PBMCs) from 50 individuals were analyzed 12 months after completion of a homologous two-dose primary COVID-19 vaccination series and had received a single booster dose (12-month follow-up)

Perineuronal Net Receptor PTPσ Regulates Retention of Memories.

Perineuronal nets (PNNs) have an important physiological role in the retention of learning by restricting cognitive flexibility. Their deposition peaks after developmental periods of intensive learning, usually in late childhood, and they help in long-term preservation of newly acquired skills and information. Modulation of PNN function by various techniques enhances plasticity and regulates the retention of memories, which may be beneficial when memory persistence entails negative symptoms such as post-traumatic stress disorder (PTSD). In this study, we investigated the role of PTPσ [receptor

The many faces of amnesia.

Results from studies of retrograde amnesia provide much of the evidence for theories of memory consolidation. Retrograde amnesia gradients are often interpreted as revealing the time needed for the formation of long-term memories. The rapid forgetting observed after many amnestic treatments, including protein synthesis inhibitors, and the parallel decay seen in long-term potentiation experiments are presumed to reveal the duration of short-term memory processing. However, there is clear and consistent evidence that the time courses obtained in these amnesia experiments are highly variable with

Dorsal hippocampal CA3 to CA1 circuit maintains contextual cocaine memories in a rat model of drug relapse.

Exposure to a cocaine-associated environment triggers drug craving and relapse by prompting the retrieval of drug-related long-term memories. Upon retrieval, memory traces enter a labile state during which they can be modified until their reconsolidation into long-term memory stores. Disrupting labile memory traces is a promising therapeutic approach for treating neuropsychiatric disorders characterized by maladaptive memories, including substance use disorder. The dorsal hippocampus cornu ammonis 3 (dCA3) is a critical regulator of cocaine-memory strength, and the dorsal CA1 (dCA1) has been i

The differential effects of ecstasy/polydrug use on executive components: shifting, inhibition, updating and access to semantic memory.

RATIONALE/OBJECTIVES: Recent theoretical models suggest that the central executive may not be a unified structure. The present study explored the nature of central executive deficits in ecstasy users. METHODS: In study 1, 27 ecstasy users and 34 non-users were assessed using tasks to tap memory updating (computation span; letter updating) and access to long-term memory (a semantic fluency test and the Chicago Word Fluency Test). In study 2, 51 ecstasy users and 42 non-users completed tasks that assess mental set switching (number/letter and plus/minus) and inhibition (random letter generation

DNA methylation inhibition participates in the anterograde amnesia key mechanism through the suppression of the transcription of genes involved in memory formation in grape snails.

The study of the amnesia mechanisms is of both theoretical and practical importance. The mechanisms of anterograde amnesia are the least studied, due to the lack of an experimental model that allows studying this amnesia type molecular and cellular mechanisms. Previously, we found that conditional food aversion memory reconsolidation impairment in snails by NMDA glutamate receptor antagonists led to the amnesia induction, in the late stages of which (>10 days) repeated training did not cause long-term memory formation. In the same animals, long-term memory aversion to a new food type was forme

Selective expression of IL-7 receptor on memory T cells identifies early CD40L-dependent generation of distinct CD8+ memory T cell subsets.

Several recent studies have demonstrated that T-helper cell-dependent events during the initial priming period are required for the generation of CD8(+) T cell-mediated protective immunity. The underlying mechanisms of this phenomenon have not yet been determined, mostly because of difficulties in studying memory T cells or their precursor populations at early stages during immune responses. We identified IL-7 receptor (CD127) surface expression as a marker for long-living memory T cells, most importantly allowing the distinction between memory and effector T cells early after in vivo priming.

Memory and executive functions in adolescents with posttreatment Lyme disease.

Although adults with late stage posttreatment Lyme disease often experience difficulties in memory, little is known about the relationship between cognition and Lyme disease in children and adolescents. Twenty-five adolescents with late stage posttreatment Lyme disease (symptoms > 6 months) and 25 participants without Lyme disease (matched on gender, IQ, age, socioeconomic status) were assessed for neuropsychological functioning, depression, school functioning, and predisease academic achievement. The Lyme group had significant deficits in cognition (short-term visual memory, short-term and de

Memory persistence enhancement by post-learning moderate exercise requires de novo protein synthesis in the dorsal hippocampus.

Acute moderate-intensity exercise (AME) after learning has been reported to exogenously boost consolidation of hippocampus-dependent memory, resulting in improved long-term persistence. However, the neuronal mechanism remains poorly understood. Short-term, hippocampus-dependent memory produced by weak encoding can be transformed into long-term memory through an immediate, strong behavioral event, which causes overlapping activation of the hippocampus. Hippocampal de novo protein synthesis is essential for achieving memory consolidation in this way. As AME activates the hippocampus, enhanced me

Dissociable memory modulation mechanisms facilitate fear amnesia at different timescales.

Memory reactivation renders consolidated memory fragile and thereby opens the window for memory updates, such as memory reconsolidation. However, whether memory retrieval facilitates update mechanisms other than memory reconsolidation remains unclear. We tested this hypothesis in three experiments with healthy human participants. First, we demonstrate that memory retrieval-extinction protocol prevents the return of fear expression shortly after extinction training, and this short-term effect is memory reactivation dependent (Study 1, N=57 adults). Furthermore, across different timescales, the

Similarity in activity and laterality patterns in the angular gyrus during autobiographical memory retrieval and self-referential processing.

Long-term memory is arguably one of the key cognitive functions. At the neural level, the lateral parietal cortex and the angular gyrus, particularly in the left hemisphere, exhibit strong activations during autobiographical and episodic memory retrieval. In a separate sub-field, left-lateralized activations of the angular gyrus are also found during self-referential processing, defined as higher activity when a trait term is judged by participants as being related to them vs. related to someone else. The question is whether episodic/autobiographical memory retrieval and self-referential proce

Long-term voluntary running improves cognitive ability in developing mice by modulating the cholinergic system, antioxidant ability, and BDNF/PI3K/Akt/CREB pathway.

Moderate physical exercise has positive effects on memory. The present study aimed to investigate the impact of long-term exercise on spatial memory in developing mice, as well as its association with the cholinergic system, antioxidant activities, apoptosis factor, and BDNF/PI3K/Akt/CREB pathway in the brain. In this study, Y maze and Novel object recognition (NOR) tests were employed to assess the impact of long-term voluntary exercise on memory. The cholinergic system, antioxidant activities, and apoptosis factors in the brain were quantified using Elisa. Additionally, western blot analysis

A revised clinical assessment of motor and memory disturbances.

For clinical assessment of motor disturbances, the motor system is better classified into the voluntary versus automatic motor systems than into the pyramidal versus extrapyramidal motor systems. The voluntary motor system is related to externally guided movements initiated by the premotor area while the automatic motor system is related to memory guided automatic movements initiated by the supplementary motor area and supported by an appropriate posture and associated movements. Among the pyramidal tract signs, muscle weakness alone is related to involvement of the corticospinal fibers of Bet

17DD Yellow Fever Revaccination and Heightened Long-Term Immunity in Populations of Disease-Endemic Areas, Brazil.

We evaluated the duration of neutralizing antibodies and the status of 17DD vaccine-specific T- and B-cell memory following primary and revaccination regimens for yellow fever (YF) in Brazil. We observed progressive decline of plaque-reduction neutralization test (PRNT) seropositivity and of the levels of effector memory CD4+ and CD8+ T cells, as well as interferon-γ+CD8+ T cells, 10 years after primary vaccination. Revaccination restored PRNT seropositivity as well as the levels of effector memory CD4+, CD8+, and interferon-γ+CD8+ T cells. Moreover, secondary or multiple vaccinations guarante

Accelerated long-term forgetting in focal epilepsy: Do interictal spikes during sleep matter?

Accelerated long-term forgetting (ALF) is a particular form of amnesia mostly encountered in focal epilepsy, particularly in temporal lobe epilepsy. This type of memory loss is characterized by an impairment of long-term consolidation of declarative memory, and its mechanisms remain poorly understood. In particular, the respective contribution of lesion, seizures, interictal epileptic discharges, and sleep is still debated. Here, we provide an overview of the relationships intertwining epilepsy, sleep, and memory consolidation and, based on recent findings from intracranial electroencephalogra

Photostimulation of locus coeruleus CA1 catecholaminergic terminals reversed Spatial memory impairment in an alzheimer's disease mouse model.

RATIONALE: One of the earliest changes associated with Alzheimer's disease (AD) is the loss of catecholaminergic terminals in the cortex and hippocampus originating from the Locus Coeruleus (LC). This decline leads to reduced catecholaminergic neurotransmitters in the hippocampus, affecting synaptic plasticity and spatial memory. However, it is unclear whether restoring catecholaminergic transmission in the terminals from the LC may alleviate the spatial memory deficits associated with AD. OBJECTIVES: This study aims to investigate the effects of optogenetic stimulation of LC catecholaminergi
