High-resolution cytogenetic analysis was performed on peripheral blood lymphocytes of 11 patients (seven women and four men, range 24-56 years) with proven autosomal dominant polycystic kidney disease (ADPKD). We describe two patients with myelodysplastic syndrome (MDS) and the Philadelphia chromosome (Ph). The patients were 64- and 69-year-old men who were diagnosed as having refractory anaemia with excess of blasts. During the terminal phase, the MDS evolved to myeloblastic leukaemia. A dose-effect curve obtained by analysis of dicentric chromosomes after irradiation of peripheral blood samples, from one donor, at 11 different doses of gamma-rays is presented. In this report we present the cytogenetic findings of low frequency (X or Y)-mosaicism (1.0%-3.6%) in six patients aged between 28 and 36 years (average age 32.17) among sixteen individuals (analysis of 5402 mitoses) referred for genetic counseling after unsuccessful assisted reproductive treatment. The absence of folic acid results in an increase in spontaneous chromosome breakage in the presence of, but not in the absence of, methionine. This is similar to the methionine effect seen in studies of the fragile X chromosome.
