Solid-State NMR Captures Time-Resolved Conformational Remodeling of Amelogenin Assemblies in Guiding Hydroxyapatite Morphogenesis.

Amelogenin is a major component in developing enamel protein matrix and plays a key role in regulating enamel mineral morphology. However, intrinsic disorder encoded in amelogenin's low-complexity regions leads to structural heterogeneity, thereby evading high-resolution characterization under physiological conditions. We hypothesize that amelogenin undergoes domain-specific rather than uniform structural and dynamic remodeling in regulating enamel apatite formation. Solid-state NMR (SSNMR), coupled with complementary techniques including scanning electron microscopy (SEM), AFM-based nano-scal

Quantitative live imaging reveals PRICKLE1 controls junctional neural tube morphogenesis independent of Planar Cell Polarity.

Neurulation, the process that forms the neural tube - the precursor to the brain and spinal cord - is frequently disrupted in congenital malformations. Primary and secondary neurulation are integrated at a junctional zone, yet the cellular dynamics linking these programs remain unknown. Using high-resolution quantitative live imaging in transgenic quail embryos, we show that the junctional neural tube forms through two coordinated processes: mediolateral convergence and EMT-driven ingression of medial neuroepithelial cells. We demonstrate that PRICKLE1, a core PCP protein, orchestrates these b

Protein Tyrosine Phosphatase Regulates Sporulation, Trap Morphogenesis, Stress Responses, and Secondary Metabolism in Arthrobotrys oligospora.

Arthrobotrys oligospora is a widely distributed nematode-trapping (NT) fungus that captures nematodes by developing flexible traps. In fungi, protein tyrosine phosphatases (PTPs) are crucial for intracellular signaling, governing processes such as cell growth, proliferation, and differentiation in filamentous species. In this study, we characterized the functions of AoPtp (an orthologous PTP) through gene knockout, phenotypic, multi-omics, and yeast two-hybrid (Y2H) analyses. Inactivation of Aoptp caused a marked increase in trap number and nematode predation ability. The ΔAoptp mutants exhibi

Juglone suppresses morphogenesis and early biofilm formation in Candida albicans through disruption of oxidative homeostasis.

Candida albicans is an important opportunistic pathogen causing superficial to life-threatening infections, especially in immunocompromised individuals. Its pathogenicity is largely driven by biofilm formation and phenotypic switching, especially the yeast-to-hypha (germ tube) transition, which plays a crucial role in antifungal tolerance and clinical progression. Despite growing interest in natural antifungals, the role of juglone (5-hydroxy-1,4-naphthoquinone, a natural naphthoquinone derived from Juglans species) in germ tube formation and redox-dependent virulence remains underexplored. Mitotically driven cytoskeletal reorganization governs zebrafish left-right organizer detachment from EVL and lumen morphogenesis.

Left-right asymmetry in vertebrate embryos is established by the left-right organizer (LRO), with the zebrafish Kupffer's vesicle (KV) providing a tractable model for studying de novo epithelial morphogenesis. During KV formation, dorsal forerunner cells (DFCs) initially form polarized attachments to the enveloping layer (EVL) before reorganizing into multicellular rosettes that precede lumen formation. Here, we show that while DFC-EVL junctions form independently of mitosis, early cytokinetic events are required for remodeling these contacts. Live imaging and targeted laser ablation reveal th…

Pink1 represses apoptosis and allows proper morphogenesis after exposure to ionizing radiation in Drosophila melanogaster.

Autophagy has emerged as a potential drug target for enhancing the efficacy of radiotherapy, but its precise role in cellular responses to ionizing radiation (IR) remains incompletely understood. To address this, we conducted a focused RNA interference (RNAi) screen in Drosophila melanogaster targeting autophagy-related genes. We found that knocking down Pink1 or ref(2)P (Drosophila p62/SQSTM1) in larval eye discs resulted in adult eye defects when the larvae were irradiated during the 3rd instar stage. Cell biological analyses revealed that Pink1RNAi expression did not affect cell proliferati

Heterochronic limb patterning in marsupials reveals flexibility in the processes underlying lateral plate mesoderm morphogenesis.

Marsupial embryos develop under intense functional constraints: neonates are born after a short gestation but must immediately crawl to the teat using precociously developed forelimbs. To meet this demand, marsupials have evolved extreme acceleration of limb morphogenesis, yet the cellular and molecular mechanisms underlying this shift remain unresolved. Using two distantly related marsupials, the fat-tailed dunnart (Sminthopsis crassicaudata) and the gray short-tailed opossum (Monodelphis domestica), we show that this acceleration extends upstream to the earliest stages of lateral plate mesod…

Adh1-Programmed SNF1 Phosphogradients Decrypt Morphogenesis in Candida albicans: Chemical Interrogation Unveils Hyphal Transition Thresholds.

While Candida albicans alcohol dehydrogenase I (Adh1) conventionally functions as an alcohol dehydrogenase, this study builds upon previous work to redefine its novel role in regulating hyphal morphogenesis and elucidates the underlying mechanisms. Adh1 knockout strains exhibited hyperfilamentation, suggesting that Adh1 directly regulates true hyphal development, independent of its canonical metabolic activity. Leveraging this phenotype, a 'reverse screening strategy' identified 2-hydroxyanthraquinone (HAQ) through high-throughput screening as a potent inhibitor of biofilm formation and hyphal…

Transcriptomic and biochemical analyses revealed the dynamic regulatory mechanisms underlying leaf morphogenesis in Camellia sinensis.

Tea leaf development is a biological process marked by sequential morphogenesis that directly affects tea yield and quality. Despite their importance, the dynamic regulatory mechanisms governing these developmental stages remain poorly understood. In this study, we utilized tea buds, first leaves, second leaves, and third leaves from tea plants as experimental materials. By integrating transcriptomic sequencing with biochemical analysis, we systematically elucidated the transcriptional-metabolic co-regulatory network underlying leaf morphogenesis. The findings revealed significant differences …

Cell and tissue mechanics in 3-D organ morphogenesis: Insights from Drosophila pupal retina development.

Morphogenesis requires dynamic changes in cell and tissue mechanics. Because organs are complex multi-cellular structures that develop slowly, understanding how they acquire their unique 3-D forms requires in vivo experimental systems that can bridge the discrete processes that occur at different physical-scales, from cell, to tissue, to multi-organ, and at different time-scales, from minutes, to hours, to days. The challenge of establishing causal relationships across scales has limited exploration of the cell biological processes that collectively produce the changes in tissue material prope…

Parallel regulation of tissue morphogenesis and cell fate specification by RAR:RXR signalling during pancreas formation.

During embryogenesis, morphologically diverse epithelial organs bud from different regions of the embryonic gut. The unique features of each nascent organ result from distinct cell fate specification and morphogenetic events elicited by organ-inducing cues. Retinoic acid is a powerful inducer of the pancreas, yet, how it regulates pancreas specification and budding from the foregut endoderm remains poorly understood. By modeling these processes using a new hESC organoid system, we demonstrate that both pancreas budding and specification are induced by RAR:RXR signalling. On the molecular level

Ume6 protein complexes connect morphogenesis, adherence and hypoxic genes to shape Candida albicans biofilm architecture.

Biofilms of the fungal pathogen Candida albicans can form on implanted medical devices and contribute to fungal virulence and are recalcitra … ion to drive Ume6-target gene interaction. Ume6 therefore acts as a bridge that connects the hyphal morphogenesis and adherence genes that shape biofilm architecture and the hypoxic response genes required for growth in the low-oxygen biofilm environment. These findings are vital for our understanding of the pathobiology of C. albicans and could open the way to …

A conserved regulation of cell expansion underlies notochord mechanics, spine morphogenesis, and endochondral bone lengthening.

Cell size is a key contributor to tissue morphogenesis 1 . As a notable example, growth plate hypertrophic chondrocytes use cellular biogenesis and disproportionate fluid uptake to expand 10-20 times in size to drive lengthening of endochondral bone 2,3 . Similarly, notochordal cells expand to one of the largest cell types in the developing embryo to drive axial extension 4-6 . In zebrafish, the notochord vacuolated cells undergo vacuole fusion to form a single large, fluid-filled vacuole that fills the cytoplasmic space and contributes to vacuolated cell expansion 7 . When this process goes a…

A subpellicular microtubule dynein transport machinery regulates ookinete morphogenesis for mosquito transmission of Plasmodium yoelii.

The cortical cytoskeleton of subpellicular microtubules (SPMTs) supports the Plasmodium ookinete morphogenesis during mosquito transmission of malaria. SPMTs are hypothesized to function as the cytoskeletal tracks in motor-driven cargo transport for apical organelle and structure assembly in ookinetes. However, the SPMT-based transport motor has not been identified in the Plasmodium. The cytoplasmic dynein is the motor moving towards the minus end of microtubules (MTs) and likely be responsible for cargo transport to the apical part in ookinetes. Here we screen 7 putative dynein heavy chain (D…

MAML3 Contributes to Induction of Malignant Phenotype of Gallbladder Cancer Through Morphogenesis Signalling Under Hypoxia.

BACKGROUND/AIM: Hedgehog (HH) signalling is a potential therapeutic target for gallbladder cancer (GBC), and Mastermind-like 3 (MAML3) is involved in the transcription of Smoothened (SMO), which is a key protein of HH signalling during hypoxia in the cancer microenvironment. MAML3 is a NOTCH signalling activator, and HH and NOTCH are involved in morphogenesis signalling. However, the association between MAML3-NOTCH and HH signalling and its role in regulating GBC cells remain unknown. This study aimed to determine whether NOTCH signalling affects tumour aggressiveness in GBC under hypoxic cond…
