Cellular and molecular contractile function in aged human skeletal muscle is altered by phosphate and acidosis and partially reversed with an ATP analog.

Skeletal muscle fatigue occurs, in part, from the accumulation of hydrogen (H+) and phosphate (Pi); however, the molecular basis through which these ions inhibit function is not fully understood. Therefore, we examined the effects of these metabolites on myosin-actin cross-bridge kinetics and mechanical properties in skeletal muscle fibers from older (65-75 yr) adults. Slow-contracting myosin heavy chain (MHC) I and fast-contracting MHC IIA fibers were examined under control (5 mM Pi, pH 7.0) and fatigue (30 mM Pi, pH 6.2) conditions at maximal calcium-activation [5 mM adenosine triphosphate (…

Muscular causes of exercise intolerance in horses.

The muscular system of the horse is remarkable in its athletic scope and capacity to adapt to the demands placed on it. Muscular fatigue often causes exercise intolerance in horses as a primary muscular dysfunction or secondary to abnormalities in other integrated systems such as the cardiovascular, respiratory, or skeletal system. This article reviews basic muscular physiology leading to a discussion of the physiologic causes of muscular fatigue. In addition, a review of pathologic muscle disorders such as muscle strains and exertional myopathies that lead to poor performance is provided.

Enhanced dimethylarginine degradation improves coronary flow reserve and exercise tolerance in Duchenne muscular dystrophy carrier mice.

Duchenne muscular dystrophy (DMD) is an X-linked disease caused by null mutations in dystrophin and characterized by muscle degeneration. Cardiomyopathy is common and often prevalent at similar frequency in female DMD carriers irrespective of whether they manifest skeletal muscle disease. Impaired muscle nitric oxide (NO) production in DMD disrupts muscle blood flow regulation and exaggerates postexercise fatigue. We show that circulating levels of endogenous methylated arginines including asymmetric dimethylarginine (ADMA), which act as NO synthase inhibitors, are elevated by acute necrotic m

Tension-type headache: pain, fatigue, tension, and EMG responses to mental activation.

Twenty patients with tension-type headache (14 chronic and 6 episodic) and 20 group-matched controls were selected for this study. They participated in a 1-hour, complex, two-choice, reaction-time test, as well as 5-minute pretest and 20-minute posttest periods. Subjects reported any pain in the forehead, temples, neck, and shoulders, as well as any feelings of fatigue and tension during the pretest, and every 10 minutes during the test and posttest by visual analog scales. Superficial electromyography was recorded simultaneously from positions representing the frontal and temporal muscles, ne

Impaired abdominal muscle contractility after high-intensity exhaustive exercise assessed by magnetic stimulation.

High-intensity exercise can induce diaphragm fatigue which can, in turn, limit exercise performance. We investigated whether expiratory musc … e can impair abdominal muscle contractility similar to diaphragmatic contractility. Thus, abdominal muscle fatigue may also contribute to exercise limitation, especially when expiratory resistance is increased as in patients with chronic obstructive pulmonary disease.

Acute Effects of Different Inspiratory Muscle Training Intensities on Intercostal Muscle Oxygenation in Healthy Adults: A Randomized Crossover Study.

BACKGROUND: Increased inspiratory loading increases oxygen consumption and metabolic demand in the respiratory muscles, affecting muscle oxygenation. However, evidence regarding different inspiratory muscle training (IMT) intensities on intercostal muscle oxygenation remains limited. This study compared acute effects of different IMT intensities on intercostal muscle oxygenation and cardiorespiratory responses. METHODS: This randomized, single-blind, crossover study included 30 healthy individuals. Participants performed IMT at 0% (sham), 30% maximal inspiratory pressure (endurance-based) (MI

Does maturation influence neuromuscular performance and muscle damage after competitive match-play in youth male soccer players?

Poor neuromuscular control and fatigue have been proposed as a risk factor for non-contact injuries especially around peak height velocity (PHV). This study explored the effects of competitive soccer match-play on neuromuscular performance and muscle damage in male youth soccer players. 24 youth players aged 13-16y were split into a PHV group (-0.5 to 0.5y) and post PHV group (1.0-2.5y) based on maturity off-set. Leg stiffness, reactive strength index (RSI), muscle activation, creatine kinase (CK), and muscle soreness were determined pre and post a competitive soccer match. Paired t-tests were

Ligands for FKBP12 increase Ca2+ influx and protein synthesis to improve skeletal muscle function.

Rapamycin at high doses (2-10 mg/kg body weight) inhibits mammalian target of rapamycin complex 1 (mTORC1) and protein synthesis in mice. In contrast, low doses of rapamycin (10 μg/kg) increase mTORC1 activity and protein synthesis in skeletal muscle. Similar changes are found with SLF (synthetic ligand for FKBP12, which does not inhibit mTORC1) and in mice with a skeletal muscle-specific FKBP12 deficiency. These interventions also increase Ca(2+) influx to enhance refilling of sarcoplasmic reticulum Ca(2+) stores, slow muscle fatigue, and increase running endurance without negatively impactin…

Application of electromyography in ergonomics: new vistas.

The application of electromyography in analysis of muscle strain in working situations is far from straight-forward. Muscle force, for instance, is not directly accessible using electromyography. New insight in electrophysiological phenomena of human muscle provides however a firm basis for analysis of localized muscle fatigue at work. Such applications entail information at the muscle fiber membrane level.

Fatigue is a multidimensional concept covering both physiological and psychological aspects. Chronic fatigue is a typical symptom of disease … Except for several rare myopathies with specific metabolic derangements leading to exercise-induced muscle fatigue, most studies have not identified a prominent peripheral cause for the fatigue in this population. In contrast, the central activation of the diseased neuromuscular system is generally found to be suboptimal. The reliability of the psychological and …

Potential effects of racket grip size on lateral epicondilalgy risks.

The effects of tennis racket grip size on the forces exerted by muscles affecting lateral epicondylalgia (LE) were assessed in this study. Grip forces and joint moments applied on the wrist were quantified under three different handle size conditions, with and without induced muscle fatigue for intermediate and advanced players. The obtained experimental results were then used as input data of a biomechanical model of the hand. This simulation aimed to quantify the impact of grip strength modulation obtained in the experiment on the wrist extensor muscle forces. Our results show that there is …

A video game based hand grip system for measuring muscle force in children.

BACKGROUND: While new therapies are continuously introduced to treat muscular dystrophy, current assessment tests are challenging to quantify, cannot be used in non-ambulatory patients, or can de-motivate pediatric patients. We developed a simple, engaging, upper-limb assessment tool that measures muscle strength and fatigue in children, including children with muscular dystrophy. The device is a bio-feedback grip sensor that motivates children to complete maximal and fatiguing grip protocols through a game-based interface. METHODS: To determine if the new system provided the same maximum gri

Effects of tape and Kinesiotape on ankle range of motion, Single Leg Drop Jump and balance after running-induced fatigue: a cross-over clinical trial.

The prevention and management of injuries in runners is a key area of research in sports medicine. Fatigue during running can lead to biomechanical imbalances and inhibition of stabilizing muscles, increasing the risk of injury. With the ankle being the most commonly injured joint in runners, rigid tape (RT) and Kinesiotape (KT) have been proposed as effective methods to improve joint stability and reduce injury risk. The objective of this study was to compare the effects of a KT and RT and no tape (control group) on lower limb balance, ankle dorsiflexion ROM, and electromyographic (EMG) activ

Can grip strength be used as a surrogate marker to monitor recovery from shoulder fatigue?

Muscular fatigue impacts on normal shoulder function, which is particularly pertinent to throwing athletes. This study aimed to investigate the relationship between grip strength and shoulder muscle fatigue to evaluate the role of grip strength as a surrogate measure for upper limb performance. Twenty healthy participants were recruited. EMG was recorded from 15 shoulder muscles during different fatiguing contractions: an initial baseline recording (Fat-Baseline); after a shoulder exhausting exercise regime (Fat-Exhaustion); and after a 10 min rest period (Fat-Recovery). Grip strength was simi…

The Relationship between Vitamin D Levels, Injury and Muscle Function in Adolescent Dancers.

Vitamin D has been shown to benefit a diverse range of health functions including muscle function. The aim of the present study was to identify serum 25(OH)D3 levels in a sample of adolescent dancers and compare them to muscle function and injury incidence. We incorporated a cross-sectional design to study 49 pre-professional male and female dancers (17±4.44 yrs, 52.1±6.72 kg, 1.63±0.07 m) in full-time training in Bazil. Serum 25[OH]D3 was analyzed by Enzyme-linked Immunosorbent Assay; quadriceps and hamstring peak torque and muscle fatigue were measured by isokinetic dynamometer at 60 and 300…

Shoulder abduction fatiguability.

Shoulder abduction fatiguability has been measured using a hand held myometer in normal subjects and patients with peripheral neuromuscular diseases. An index of fatiguability was based on the decline in force over a series of ten maximum voluntary contractions performed in under a minute. The technique was repeatable and well tolerated. Patients with myasthenia, mitochondrial myopathy and motor neuron disease tended to show excess fatiguability independent of muscle strength. Serial measurements demonstrated alterations in fatiguability but not necessarily strength, associated with changes in

Upper-Body Neuromechanical Coordination Strategies During Fatiguing Sustained Dead Hangs in Climbers.

This study investigated how upper-body joint and muscle coordination strategies are related and adapted under fatigue during sustained dead hangs, a task relevant to climbing performance. Eleven climbers performed a dead hang until failure. Joint coordination variability (rCV) was assessed using a modified vector coding method in kinematic data. Muscle coordination was analyzed through EMG linear and non-linear dependencies between signals with coherence (β/γCOH) and mutual information (β/γMI) at beta and gamma bands. Fatigue induced joint-specific changes in coordination variability, with wri

Effect of fatigability on sprint time performance and force-velocity profile according to maturity status in young rugby players.

Little is known about the influence of fatigue in repeated overground sprinting on force-velocity properties in children and adolescents, while this ability to repeat sprints is important for future progress in rugby union. Sprint time decline is commonly used to assess fatigability. However, it does not provide data on biomechanical aspects of sprint performance such as maximal power, force, and velocity production. As sprint time performance and force-velocity properties do not linearly change during adolescence, considering maturity status is important. This study aimed to assess the effect
