Chemical Contaminants: Their Metabolism and their Residues.

Chemical contaminants which food animals may be exposed to include agricultural chemicals (e.g. insecticides, herbicides, fungicides, fumigants), industrial chemicals, metals and natural toxins (e.g. mycotoxins, phytotoxins, bacterial toxins). In the past, most intoxications of food animals resulted from natural toxicants. However, rapid development and usage of synthetic chemicals, while greatly benefitting society, have also provided new sources of potential chemical contamination. Various sources of contamination exist, but generally at least 80% of all residues in food animals are estimate…

The effects of a clay mineral-based feed additive on metabolism and inflammation in heat-stressed lactating dairy cows.

Study objectives were to determine the effects of a dietary clay mineral-based feed additive on performance, metabolism, inflammation, and liver and gastrointestinal health in acutely heat-stressed lactating dairy cows. Primiparous Holstein cows (148 ± 25 DIM; 600 ± 50 kg BW; n = 12) were balanced by DIM and milk yield and assigned to 1 of 2 ad libitum fed treatments: 1) control (CON; 500 g ground corn/d; n = 6), or 2) clay mineral-based mycotoxin binder (CM; 500 g ground corn and 50 g of Biofix®/d; n = 6). Respective treatments were top-dressed into the individually fed TMR and the mycotoxin

Edeine B1 produced by Brevibacillus brevis reduces the virulence of a plant pathogenic fungus by inhibiting mitochondrial respiration.

Plant pathogenic fungi cause serious diseases, which result in the loss of crop yields and reduce the quality of crops worldwide. To counteract the escalating risks of chemical fungicides, interest in biological control agents to manage plant diseases has significantly increased. In this study, we comprehensively screened microbial culture filtrates using a yeast screening system to find microbes exhibiting respiratory inhibition activity. Consequently, we found a soil-borne microbe Brevibacillus brevis HK544 strain exhibiting a respiration inhibitory activity and identified edeine B1 (EB1) fr

Improved and adopted murine models to combat pulmonary aspergillosis.

The insufficient basic and clinical knowledge about invasive mold infections necessitated to review aspergillosis rodent models. The scope of this review has two major aspects. (1) It briefly summarizes Aspergillus toxicoses, the adverse effects of Aspergillus mycotoxins, the virulence factors of Aspergillus fumigatus, and how mild Aspergillus infections can turn to immunosuppressive diseases, ultimately to lethal invasive pulmonary aspergillosis. (2) The second major aspect of the review deals with earlier and recent murine models of pulmonary aspergillosis. Particular attention will be paid …

Two Babinski signs in seropositive (HAM) and seronegative tropical spastic paraparesis.

Tropical spastic paraparesis (TSP) may or may not be associated to HTLV-I antibodies and is usually characterized by clinical and pathologic … er than the spinal cord abnormalities usually considered. Possible non-infectious co-factors (e.g., mycotoxins) may be involved in disease pathogenesis in a multistep process of viruses, toxins and environment which may account for serological differences found in this group of patients.

Critical evaluation of environmental exposure agents suspected in the etiology of Balkan endemic nephropathy.

Balkan endemic nephropathy (BEN), a kidney disease that occurs in rural villages in Bosnia, Bulgaria, Croatia, Romania, and Serbia, is thoug … r epidemiologic evidence related to BEN and toxicologic or exposure evidence related to the agents. Mycotoxins and aristolochic acid are the primary targets of current toxicologic investigations, and while the evidence on exposures for both is potentially consistent, it is insufficient.

Fungal air-borne spores as health risk factors among workers in alimentary industries.

A survey to evaluate the occurrence of air-borne fungal spores in two different food industries, dairies and bakeries, was conducted. Our data revealed considerable fungal pollution in the environments of both industries, as well as some differences in the distribution of the genera of fungi recovered. Noteworthy was the frequent finding of numerous fungi frequently responsible for allergic rhinitis, asthma and other diseases, or well-known for their production of mycotoxins in foods or characterized by their degradative activity against various substances. Aspergillus, Candida, Fusarium, Geot…

Molecular docking analysis of Omt-A protein model from Aspergillus flavus with synthetic compounds.

Aflatoxin is a potent mycotoxin of Aspergillus flavus that has been classified as a Group I carcinogen. O-methyltransferase A (Omt-A) is a critical enzyme in the formation of aflatoxin. It catalyzes the methylation of norsalic acid to form the highly toxic intermediate averantin. The ligand-protein interaction of Omt-A was performed with piperlonguminin and blasticidins. The maximum affinity of -10.6 was found for the 5ICC_A piperlonguminine at site1 (X,Y,Z: -15.282, 21.785, 5.672). Compounds such as Blasticidin S, Neoeriocitrin, Blasticidin S - hydrochloric acid, 6,6''-Bigenkwanin, Pipernomal

Aspergillus flavus and aflatoxins B1, B2, and M1 in corn associated with equine death.

Corn from an Arkansas farm, where three horses died and others became sick, was investigated for causative principles. Necropsy of the three horses revealed what appeared to be severe hepatic necrosis. Histopathological examination indicated a pattern of hepatic lesions that was suggestive of aflatoxin contamination of the feed. Mycological examination of the corn by dilution plating revealed 95% of the colonies as Aspergillus flavus. Chemical analysis of the corn for mycotoxins was positive for aflatoxin B1, B2, and M1 at concentrations of 114, 10, and 6 micrograms/Kg, respectively. Cyclopiaz…

Yellow rain: chemical warfare in Southeast Asia and Afghanistan.

Circumstantial and laboratory evidence has accumulated supporting the use in Southeast Asia and Afghanistan of Russian-made lethal chemical agents. In this paper we will attempt to answer the question, "What is Yellow Rain?", summarize some of the massive data on microtoxins and mycotoxicoses, discuss the toxicology of the trichothecenes, give a brief historical perspective on chemical warfare, and touch on some of the political implications of these developments. The ubiquitous and insidious mycotoxins have been more or less causally linked to several human diseases, from ergotism to Reye's s…

Toxic factors of mould origin.

The chemistry and effects of mycotoxins associated with human and animal foodstuffs are reviewed. The aflatoxins, metabolites of Aspergillus flavus, have been implicated in fatal diseases of farm stock fed on infected peanut cake. Muscarine and the phalloidins are the causative agents in mushroom poisoning. Lysergic acid alkaloids are involved in ergotism. "Yellow rice" toxicity arises from infection with Penicillium islandicum, the active principles being islanditoxin and luteoskyrin. Various species of Penicillium, Aspergillus and Fusarium have been linked with other mycotoxicoses and their …

Chlorella sorokiniana attenuates deoxynivalenol-exacerbated allergic dermatitis and type 2 immune responses in mice.

Chronic dietary exposure to low concentrations of deoxynivalenol (DON), a trichothecene mycotoxin found in cereal products, disrupts immune homeostasis and exacerbates allergic inflammation. Chlorella sorokiniana (CS) can adsorb DON and reduce its systemic absorption. However, whether this DON-binding activity protects against DON-induced immune dysregulation remains unclear. Here, we investigated the effects of CS on DON-induced toxicity and DON-exacerbated allergic dermatitis. In vitro, murine DC2.4 dendritic cells were exposed to DON in the presence or absence of CS-treated DON preparations

Understanding the working of magnetic COFs in the extraction and determination of target contaminants for food safety.

Rising food safety concerns necessitate sensitive, rapid, and reliable analytical methods for contaminant detection. Covalent organic frameworks (COFs), especially magnetic COF composites, have emerged as powerful sorbents for sample preparation due to their tunable porosity, functional diversity, and magnetic recoverability. This review summarizes recent advances in COF-based magnetic solid-phase extraction (MSPE) applied to food matrices for the enrichment and detection of diverse contaminants, including mycotoxins, sulfonamides, aromatic hydrocarbons, phenolic compounds, pesticides, and bis…

Animal performance and biochemical parameters are sex-dependent in peripubertal rats exposed to deoxynivalenol.

Deoxynivalenol (DON), a mycotoxin produced mainly by Fusarium graminearum and F. culmorum commonly contaminates food commodities across the globe. Due to this, exposure to DON might pose potential health hazards to humans and animals. Biological factors like sex and age can influence the toxicity of DON. However, in toxicological studies involving DON, the sex and age-dependent response has been often overlooked. Thereby, the objective of this study was to evaluate if sex differences are evident in DON's systemic effects in peripubertal rats. Juvenile animals (n = 24) with 28 days postnatal da

Biodegradation of Fumonisins by the Consecutive Action of a Fusion Enzyme.

Fumonisins (FBs) are toxic mycotoxins that commonly exist in food and feed. FBs can induce many aspects of toxicity, leading to adverse effects on human and animal health; therefore, investigating methods to reduce fumonisin contamination is necessary. In our study, we generated a recombinant fusion enzyme called FUMDI by linking the carboxylesterase gene (fumD) and the aminotransferase gene (fumI) by overlapping polymerase chain reaction (PCR). The fusion enzyme FUMDI was successfully, secretively expressed in the host Pichia pastoris (P. pastoris) GS115, and its expression was optimized. Our…

Candidalysin: discovery and function in Candida albicans infections.

Candidalysin is a cytolytic peptide toxin secreted by the invasive form of the human pathogenic fungus, Candida albicans. Candidalysin is critical for mucosal and systemic infections and is a key driver of host cell activation, neutrophil recruitment and Type 17 immunity. Candidalysin is regarded as the first true classical virulence factor of C. albicans but also triggers protective immune responses. This review will discuss how candidalysin was discovered, the mechanisms by which this peptide toxin contributes to C. albicans infections, and how its discovery has advanced our understanding of

Myotoxic activity of an acidic phospholipase A2 isolated from Lachesis muta (Bushmaster) snake venom.

An acidic phospholipase A2 isolated from Lachesis muta snake venom denoted LM-PLA2, showed neither toxic nor anticoagulant activities in contrast to a potent inhibitory effect of collagen-induced platelet aggregation [Fuly, A.L., Machado. O.L.T., Alves, E.W. and Carlini, C.R., 1997. Thromb. Haemost 78, 1372-1380.]. Now, the myotoxicity induced by LM-PLA2 was investigated by using both in vivo and in vitro experiments. LM-PLA2 induced in vitro a dose- and time-dependent release of creatine-kinase (CK) from mouse Extensor Digitorium Longus (EDL) muscles and also increased the plasma CK activity

Toxicity of echinulin from Aspergillus chevalieri in rabbits.

Rabbits were injected intraperitoneally with purified echinulin, a product of Aspergillus chevalieri. After 2 h, the rabbits were bled and enzyme analyses were carried out on the supernates of liver homogenates and citrated plasma. Elevated levels of total plasma lactate dehydrogenase, cardiac derived isozyme, glutamic-oxaloacetic and glutamic-pyruvic transaminase activities in animals receiving toxin were observed. These levels were statistically significant compared to the vehicle control. A significant increase in liver lactate dehydrogenase of toxin-treated rabbits was also observed. Light
