alpha-Latrotoxin, from black widow spider venom, stimulates exocytosis of small synaptic vesicles at central and peripheral synapses. However, it is widely accepted that neuropeptide-containing large dense-core vesicles are insensitive to the toxin. In the present study, we investigated whether alpha-latrotoxin releases neuropeptides from primary afferent and enteric neurons. The guinea-pig renal pelvis is innervated by primary sensory neurons containing tachykinins and calcitonin gene-related peptide, but has no functional cholinergic or noradrenergic motor innervation. alpha-Latrotoxin incre…

The olfactory bulbs of young and adult normal rats and of colchicine-treated rats and of some other species were analysed for the presence of neuropeptide Y and neuropeptide Y messenger RNA, using immunohistochemistry at the light- and electron-microscopic levels and with in situ hybridization. In the rat and mouse, but not in monkey and guinea-pig, neuropeptide Y-like immunoreactivity and neuropeptide Y messenger RNA were observed in ensheathing cells in the olfactory nerve layer of the olfactory bulb and within nerve bundles in the olfactory mucosa. Double staining experiments revealed that

The ciliary processes (CPs) in the rat were investigated for the presence of neuropeptide Y (NPY)-like immunoreactivity and for histofluorescence indicating catecholamine (CAM)-ergic innervation in the rat, rabbit and cynomolgus monkey. Special attention was paid to those CP vessel segments which in previous studies showed distinct reactions on application of CAM or NPY, and on the respective sections of the CP epithelium. In the rat and cynomolgus monkey most CAM-ergic nerve fibers concentrated along the terminal arterioles and the epithelium of the anteriormost portion of the major CPs. In c

Neuropeptide Y (NPY) administration into the third brain ventricle of normal and castrated male rats decreases prolactin and LH secretion almost not affecting FSH. Simultaneous augmentation of hypophyseal response to LH-RH is observed. NPY effect on LH release is more pronounced in castrated animals as compared to normal ones. It is concluded that in the CNS inhibitory action of NPY prevails as regards to neuroendocrine control of gonadotropin and prolactin secretion. The obtained results allow NPY participation in feedback regulation of LH secretion in males.

The biosynthesis of the hormone melatonin (MEL) by the mammalian pineal gland has been thought to be regulated strictly by stimulatory factors, most predominantly norepinephrine (NE), released from the sympathetic nerve fibers which heavily innervate the gland. Evidence from many investigators suggests that sympathetic fibers may colocalize other neuroactive factors in addition to NE. One of these factors is neuropeptide Y (NPY), which has been found in the nerve fibers of the pineal gland. The present study sought to explore potential interactions between NE and NPY in the regulation of pinea

Immunohistochemical studies in several mammalian species and in red-eared turtles have shown that somatostatin (SS) and neuropeptide Y (NPY) … r structures) neurons labeled only for NPY or only SS were more abundant than those containing both neuropeptides. Outside the telencephalon, the only forebrain cell group containing neurons in which SS and NPY were co-localized was in the lateral hypothalamus. A series of double- and triple-label immunohistochemical studies was undertaken to determine the exte …

The related central nervous system peptides neuropeptide Y and peptide YY have been found to be among the most potent endogenous stimulants of feeding behavior. We measured these neuropeptides in cerebrospinal fluid to determine whether they contributed to the pathophysiologic characteristics of anorexia and bulimia nervosa. Cerebrospinal fluid neuropeptide Y concentrations were significantly elevated in underweight anorectic patients and in many of the anorectic patients studied at intervals after weight restoration. These levels normalized in long-term weight-restored anorectic patients who …

We investigated the possible relation between neuropeptides and cerebral vasoconstriction in samples of ventricular or cisternal cerebrospinal fluid from 14 patients with subarachnoid hemorrhage. Neuropeptide Y, calcitonin gene-related peptide, atrial natriuretic peptide, and pituitary polypeptide 7B2 were present in the cerebrospinal fluid of these patients. Concentrations of calcitonin gene-related peptide and 7B2 were not significantly different from those in control subjects, but that of atrial natriuretic peptide was significantly lower. Although the mean concentration of neuropeptide Y w…

Neuropeptide Y (NPY, 0.47 nmol) and epinephrine (28.9 nmol) evoked robust, and quantitatively similar, increments in food intake and local eating rate following administration into the third cerebral ventricle (IIIV). Whereas IIIV pretreatment with phentolamine (71 nmol), a nonselective alpha-adrenoceptor antagonist, or prazosin (9.5 nmol), a selective alpha 1-adrenoceptor antagonist, was without effect on NPY-induced feeding behavior, pretreatment with the alpha 2-adrenoceptor antagonist yohimbine (15 nmol) dramatically attenuated the stimulatory effects of NPY or epinephrine on both food int

Nerves in the uterine cervix of the rat were examined with regard to co-existence of markers for noradrenaline and neuropeptide Y, and differential tissue innervation by nerves containing different combinations of these markers. Immunohistochemical labeling of single and adjacent serial cryostat sections, and double labeling was employed. Some animals were treated with the noradrenergic neurotoxin, 6-hydroxydopamine. In control animals neuropeptide Y-immunoreactive fibers were numerous in the myometrium and around arteries; noradrenergic fibers were few in the myometrium and moderate in number

The effect of the intrathecal administration of neuropeptide Y (NPY) on blood pressure and heart rate of anesthetized normotensive and hypertensive rats was studied. Neuropeptide Y was observed to produce a decrease in the blood pressure of Sprague-Dawley, Wistar Kyoto (WKY), DOCA-salt, and DOCA-sham control rats. The maximum percent decrease in blood pressure of Sprague-Dawley rats was 12.8 and 15.2% in response to 0.1 and 1.0 nmol NPY, respectively. Similar changes in heart rate were observed. The depressor effect of intrathecal NPY was attenuated by prior treatment with yohimbine and propra

The aganglionic intestinal segment in Hirschsprung's disease is known to contain a reduced number of nerve fibers storing vasoactive intestinal peptide (VIP), substance P (SP), enkephalin, and gastrin-releasing peptide (GRP). In this study, nerves containing three newly described neuropeptides: neuropeptide Y (NPY), calcitonin gene-related peptide (CGRP), and galanin were examined using immunocytochemistry. Nerve fibers displaying NPY immunoreactivity were found to be more frequent in the aganglionic than in nonafflicted ganglionic intestine. Nerve fibers storing CGRP and galanin on the other …

The myometrial levels of noradrenaline (NA) and neuropeptide Y (NPY), a recently discovered neuropeptide that coexists with NA in many sympathetic nerves, have been measured by immunohistochemistry and biochemistry in normal and pre-eclamptic pregnancy and compared with non-pregnant controls. In the non-pregnant uterus there were high levels of NA and NPY and a rich presence of NA/NPY nerves in the myometrium and around blood vessels. In pregnancy both substances were significantly lower as compared with non-pregnant women. Five patients with pre-eclampsia had levels of NA/NPY that were less m

Neurons identified by their immunoreactivity with antisera against neuropeptide Y (NPY) were studied in three selected areas of the cerebral cortex in brains from controls and in senile dementia of the Alzheimer type (ATD). Changes were more profound in temporal cortex than in parietal cortex, and more severe in parietal cortex than in frontal cortex, paralleling the severity of neuritic plaque formation and incidence of neurofibrillary tangles in these regions. NPY-i neurons became distorted, with enlarged misshapen cell somata and reduced, thickened, and gnarled dendrites. There was a sharp

Neuropeptide Y (NPY) has been demonstrated in the human circle of Willis by specific radioimmunoassay. Concentrations were similar in cadaver and peroperative specimens. NPY concentrations were highest around the basilar bifurcation (2.9 +/- 1.0 pmol/g), anterior cerebral artery (2.7 +/- 0.5 pmol/g), and carotid trifurcation (2.4 +/- 0.6 pmol/g). Concentrations were lowest in the basilar artery (1.0 +/- 0.4 pmol/g). Intracarotid administration of NPY (50 pmol to 2 nmol) to rats reduced mean cerebral cortical blood flow by 40-98%. This vasoconstriction lasted for at least 2 h. These findings su

Alternative processing of the RNA transcribed from the calcitonin gene appears to result in the production of a messenger RNA in neural tissue distinct from that in thyroidal 'C' cells. The thyroid mRNA encodes a precursor to the hormone calcitonin whereas that in neural tissues generates a novel neuropeptide, referred to as calcitonin gene-related peptide (CGRP). The distribution of CGRP-producing cells and pathways in the brain and other tissues suggests functions for the peptide in nociception, ingestive behaviour and modulation of the autonomic and endocrine systems. The approach described

The micronutrients vitamins B9 and B12 act as methyl donors in the one-carbon metabolism involved in transmethylation reactions which critically influence epigenetic mechanisms and gene expression. Both vitamins are essential for proper development, and their deficiency during pregnancy has been associated with a wide range of disorders, including persisting growth retardation. Energy homeostasis and feeding are centrally regulated by the hypothalamus which integrates peripheral signals and acts through several orexigenic and anorexigenic mediators. We studied this regulating system in a rat m
