Oral Rehabilitation of Ectodermal Dysplasia Using modified Groper's Appliance - A Case Report.

RATIONALE: Ectodermal dysplasia is a genetic condition characterised by atypical development of organs derived from the embryonic ectoderm, such as teeth, nails, hair and sweat glands. CLINICAL SIGNS: The most prevalent oral manifestation is hypodontia, which involves the absence of some or all primary and permanent teeth. Consequently, individuals with this condition often require dental prosthetic interventions during their formative years. PATIENT CONCERNS: This report discusses the oral rehabilitation of a 2-year-old child affected by ectodermal dysplasia with oligodontia using a modifie

How the mouse got its spots.

One's ultimate phenotype is the result of a combination of genotype and environment, and includes a poorly understood component termed "developmental noise". This "developmental noise", also known as "intangible variation", is rarely discussed even though it appears to make a significant contribution to the variance of quantitative traits within a species. The molecular basis of developmental noise remains unknown, but it appears to be established in embryonic development and to be retained for the life of the organism. We propose that the molecular basis of developmental noise is, at least in

Bicoid-independent formation of thoracic segments in Drosophila.

The maternal determinant Bicoid (Bcd) represents the paradigm of a morphogen that provides positional information for pattern formation. However, as bicoid seems to be a recently acquired gene in flies, the question was raised as to how embryonic patterning is achieved in organisms with more ancestral modes of development. Because the phylogenetically conserved Hunchback (Hb) protein had previously been shown to act as a morphogen in abdominal patterning, we asked which functions of Bcd could be performed by Hb. By reestablishing a proposed ancient regulatory circuitry in which maternal Hb con

The Beckwith-Wiedemann syndrome phenotype and the risk of cancer.

Beckwith-Wiedemann syndrome (BWS) comprises of a number of childhood abnormalities, often associated with one or more tumors. Thirty-eight patients were investigated to determine clinical and/or biological signs associated with a tumor presence. Our patients exhibited a higher incidence of tumor development (21%) than that previously reported, underlying the care with which such patients should be followed, when particular clinical features are observed: visceromegaly affecting three organs (liver, kidney, spleen), and also family history with sign of BWS such as macroglossia, omphalocele, hem

Targeted disruption of the GATA3 gene causes severe abnormalities in the nervous system and in fetal liver haematopoiesis.

GATA-3 is one member of a growing family of related transcription factors which share a strongly conserved expression pattern in all vertebrate organisms. In order to elucidate GATA-3 function using a direct genetic approach, we have disrupted the murine gene by homologous recombination in embryonic stem cells. Mice heterozygous for the GATA3 mutation are fertile and appear in all respects to be normal, whereas homozygous mutant embryos die between days 11 and 12 postcoitum (p.c.) and display massive internal bleeding, marked growth retardation, severe deformities of the brain and spinal cord,

[The metabolism of fluorogenic monoamines during the differentiation of embryonic pancreatic endocrine cells of birds].

The FIF technique applied to embryonic pancreas of the chick shows that the islte cells of the splenic lobe are able to take up L-DOPA and to synthesize high amounts of catecholamines from the 4th day of incubation. In the ventral pancreatic lobes fluorescent cells are not detectable before day 12. Later on, islet cells of the whole organ spontaneously accumulate fluorogenic amines and L-DOPA injection greatly increases the intensity of the fluorescence. It appears that in endocrine pancreas, as in various other polypeptide hormone producing cells, an aminoacid decarboxylase activity is detect

Development of pre-implantation mouse embryos under the influence of pineal indoles.

The developmental toxicity of pineal hormones on mouse embryos was examined both in vitro and in vivo. Pregnant ICR mice were divided into groups which received at 1.5 days post-coitum (p.c.) and again at 2.5 days p.c. a subcutaneous injection of one of the following pineal indoles: hydroxyindoleacetic acid (HIAA), melatonin (MEL), methoxytryptophol (MTP) or methoxytryptamine (MTA). Mice treated with the injection vehicle served as the control. The animals were sacrificed at 17.5 days p.c. The pineal indole treatment did not cause changes in the gravid uterine weight, numbers of implants, earl

Transcriptomic and spatial organization of telencephalic GABAergic neurons.

The telencephalon of the mammalian brain contains multiple regions and circuits that have adaptive and integrative roles in a variety of brain functions. GABAergic neurons in the telencephalon are diverse; they have many circuit functions, and dysfunction of these neurons has been implicated in various brain disorders1-3. Here we conducted a systematic and in-depth analysis of the transcriptomic and spatial organization of GABAergic neuronal types in all regions of the mouse telencephalon and their developmental origins. This was accomplished using 611,423 young adult single-cell transcriptome

Knockdown of zebrafish YY1a can downregulate the phosphatidylserine (PS) receptor expression, leading to induce the abnormal brain and heart development.

BACKGROUND: Yin Yang 1 (YY1) is a ubiquitously expressed GLI-Kruppel zinc finger-containing transcriptional regulator. YY1 plays a fundamental role in normal biologic processes such as embryogenesis, differentiation, and cellular proliferation. YY1 effects on the genes involved in these processes are mediated via initiation, activation, or repression of transcription depending upon the context in which it binds. The role of the multifunctional transcription factor Yin Yang 1 (YY1) in tissue development is poorly understood. In the present, we investigated YY1a role in developing zebrafish on P

Comprehensive In Vitro Toxicity Testing of a Panel of Representative Oxide Nanomaterials: First Steps towards an Intelligent Testing Strategy.

Nanomaterials (NMs) display many unique and useful physico-chemical properties. However, reliable approaches are needed for risk assessment of NMs. The present study was performed in the FP7-MARINA project, with the objective to identify and evaluate in vitro test methods for toxicity assessment in order to facilitate the development of an intelligent testing strategy (ITS). Six representative oxide NMs provided by the EC-JRC Nanomaterials Repository were tested in nine laboratories. The in vitro toxicity of NMs was evaluated in 12 cellular models representing 6 different target organs/systems

New advances in human embryology: morphofunctional relationship between the embryo and the yolk sac.

Organogenesis occurs during the first 8 weeks of human embryonic development; in consequence, early human growth and development take place before and in the absence of fully developed internal organs. During this period, normal development depends on several factors, but two are imperative: nutrition and a functional transport system for the distribution of nutrients and for waste disposal. The yolk sac (YS), a highly differentiated adnexal organ, is known to accomplish this fundamental task during early pregnancy. In this review, we summarize our contribution to the understanding of early hu

Phenotypic variations among paternal centrosomes expressed within the zygote as disparate microtubule lengths and sperm aster organization: correlations between centrosome activity and developmental success.

This study describes a paternal effect on sperm aster size and microtubule organization during bovine fertilization. Immunocytochemistry using tubulin antibodies quantitated with confocal microscopy was used to measure the diameter of the sperm aster and assign a score (0-3) based on the degree of radial organization (0, least organized; 3, most organized). Three bulls (A-C) were chosen based on varying fertility (A, lowest fertility; C, highest fertility) as assessed by nonreturn to estrus after artificial insemination and in vitro embryonic development to the blastocyst stage. The results in

Charting Endocrine Progenitors Across Species and Organs.

Hormone-producing cells in the pancreas and intestine regulate whole-body metabolism and are dysregulated in obesity and diabetes. A deeper understanding of endocrine lineage formation (endocrinogenesis) in these organs is essential to elucidate the healthy state and disease trajectories. Here, we performed cross-species (mouse and human) and cross-system (in vivo and in vitro) comparisons of pancreatic endocrine progenitors (EPs), complemented by cross-organ (pancreas and intestine) analyses, using single-cell transcriptomics and epigenomics, together with mouse bulk proteomics. We uncovered

The membrane receptor for plasma retinol-binding protein, a new type of cell-surface receptor.

Vitamin A is essential for diverse aspects of life ranging from embryogenesis to the proper functioning of most adult organs. Its derivatives (retinoids) have potent biological activities such as regulating cell growth and differentiation. Plasma retinol-binding protein (RBP) is the specific vitamin A carrier protein in the blood that binds to vitamin A with high affinity and delivers it to target organs. A large amount of evidence has accumulated over the past decades supporting the existence of a cell-surface receptor for RBP that mediates cellular vitamin A uptake. Using an unbiased strateg

Aging and transplantation - a topic for biomedicine or bioethics?

The aged patient stands at the nexus of significant biomedical and bioethical issues in transplantation. This in itself can be seen as a microcosm of an imminent global tempest, stemming from expanding numbers and longer lives of the aged population. As a candidate for receiving organ and tissue transplants, the geriatric patient is challenging because they present unique physiology for medical management. As organ and tissue donors, the aged are perceived of as providing "marginal" organs, which drives the fear that the graft will fail before the recipient. Such difficulties lead inexorably t

Autophagy as an architect of embryogenesis: Understanding the new mechanisms, emerging paradigms, and future directions.

Autophagy is no longer considered just a generic degradation pathway but is rather acknowledged as a programmed developmental mechanism whose roles in coordination across embryogenesis are still not very well understood. Various mammalian and non-mammalian models present evidence, where autophagy is switched on at fertilization, maintains proteostasis during cleavage, allocates lineage during blastocyst formation, and enforces genomic as well as cellular quality control by the selective removal of abnormal cells or paternal organelles. Moreover, following gastrulation, which gives rise to the

Complex Organ Construction from Human Pluripotent Stem Cells for Biological Research and Disease Modeling with New Emerging Techniques.

Human pluripotent stem cells (hPSCs) are grouped into two cell types; embryonic stem cells (hESCs) and induced pluripotent stem cells (hiPSCs). hESCs have provided multiple powerful platforms to study human biology, including human development and diseases; however, there were difficulties in the establishment of hESCs from human embryo and concerns over its ethical issues. The discovery of hiPSCs has expanded to various applications in no time because hiPSCs had already overcome these problems. Many hPSC-based studies have been performed using two-dimensional monocellular culture methods at t

Tissue engineering: current strategies and future directions.

Novel therapies resulting from regenerative medicine and tissue engineering technology may offer new hope for patients with injuries, end-stage organ failure, or other clinical issues. Currently, patients with diseased and injured organs are often treated with transplanted organs. However, there is a shortage of donor organs that is worsening yearly as the population ages and as the number of new cases of organ failure increases. Scientists in the field of regenerative medicine and tissue engineering are now applying the principles of cell transplantation, material science, and bioengineering

GABAergic system of the pineal organ of an elasmobranch (Scyliorhinus canicula): a developmental immunocytochemical study.

The present immunocytochemical study provides evidence of a previously unrecognized, rich, gamma-aminobutyric acid (GABA)-ergic innervation of the pineal organ in the dogfish (Scyliorhinus canicula). In this elasmobranch, the pineal primordium is initially detected at embryonic stage 24 and grows to form a long pineal tube by stage 28. Glutamic acid decarboxylase (GAD)-immunoreactive (-ir) fibers were first observed at stage 26, and by stage 28, thin GAD-ir fibers were detectable at the base of the pineal neuroepithelium. In pre-hatchling embryos, most fibers gave rise to GAD-ir boutons that w

Parallel regulation of tissue morphogenesis and cell fate specification by RAR:RXR signalling during pancreas formation.

During embryogenesis, morphologically diverse epithelial organs bud from different regions of the embryonic gut. The unique features of each nascent organ result from distinct cell fate specification and morphogenetic events elicited by organ-inducing cues. Retinoic acid is a powerful inducer of the pancreas, yet, how it regulates pancreas specification and budding from the foregut endoderm remains poorly understood. By modeling these processes using a new hESC organoid system, we demonstrate that both pancreas budding and specification are induced by RAR:RXR signalling. On the molecular level
