The potential role of nitric oxide synthase in survival and regeneration of magnocellular neurons of hypothalamo-neurohypophyseal system.

Previous investigations from this laboratory have demonstrated that hypophysectomy induces up-regulation of neuronal nitric oxide synthase (nNOS) in magnocellular neurons of the mammalian hypothalamo-neurohypophyseal system (HNS). Accompanied by this upregulation of nNOS, both neuronal regeneration and degeneration are also observed in this system following hypophysectomy. The specific aim of this study was to determine the potential role of nNOS upregulation in neuronal survival and regeneration after hypophysectomy in the adult Sprague-Dawley (SD) rat by using a competitive nitric oxide synt

[Changes in plasticity of rat hypothalamic neurons and astrocytes in humid and hot environment].

OBJECTIVE: To study the changes in the plasticity of the neurons and astrocytes in the paraventricular nucleus (PVN) and supraoptic nucleus (SON) of the hypothalamus of rats exposed to a humid and hot environment. METHODS: The rats were subjected to stimulation with a humid and hot environment for 120 min in a climate chamber (dry bulb temperature of 40.0-/+0.5 degrees C with relative humidity of 60-/+5%). During the exposure, the behavioral responses of the rats were observed, and the changes in the expressions of Fos and GFAP in the PVN and SON in response to the exposure evaluated using im

Neural regeneration and neuronal migration following injury. I. The endocrine hypothalamus and neurohypophyseal system.

Central to this investigation are several basic hypotheses that are designed to test the role of nitric oxide (NO) in the complex process of central regeneration and plasticity in a well established model system of the mammalian brain. We have employed histochemical techniques at the light and ultrastructural level coupled with correlative scanning electron microscopy, immunoelectron microscopy, and in situ hybridization in order to determine the functional significance of the increased expression of nitric oxide synthase (NOS) in neurons of the supraoptic (SON) and paraventricular (PVN) nucle

Aerobic Exercise Activates Fibroblast Growth Factor 21 and Alleviates Cardiac Ischemia/Reperfusion-induced Neuronal Oxidative Stress and Ferroptosis in Paraventricular Nucleus.

Cardiac ischemia/reperfusion (I/R) induces systemic oxidative stress, which in turn gives rise to the development of multiple organ abnormalities, including brain injury. The paraventricular nucleus (PVN) of the hypothalamus is a cardiovascular regulatory center. Aerobic exercise is an effective intervention to protect the heart against I/R injury. However, the effect of aerobic exercise on cardiac I/R-induced neuronal injury in the PVN has not been fully elucidated. The aim of this study is to investigate whether aerobic exercise can up-regulate fibroblast growth factor 21 (FGF21) and allevia

Linking oxytocin to nicotine dependence: An experimental study of the brain, behavior, and gut microbiota in rats.

Oxytocin (OXT) is a hypothalamic neuropeptide, and numerous studies have indicated that exposure to addictive substances, such as opioids, cocaine, etc., can result in decreased function of the OXT system. The study also found that OXT can reduce addictive behavior for certain drugs, including methamphetamine, alcohol, and cocaine, suggesting a close relationship between the OXT system and drug abuse. Although nicotine is the main addictive substance in tobacco, its interaction with the OXT system is unknown and requires further study. Therefore, OXT levels in plasma and brain regions associat

Sonogenetic neuromodulation prevents post-myocardial infarction malignant arrhythmia and cardiac remodeling through the brain-heart circuit.

Sympathetic hyperactivation within the paraventricular nucleus (PVN) exacerbates post-myocardial infarction (MI) malignant ventricular arrhythmias (VAs) and cardiac remodeling. Sonogenetics is an emerging reversible neuromodulation, which might achieve precise spatio-temporal controllability over targeted neurons. However, the current sonogenetic neuromodulation (SGN) strategies are mainly designed to facilitate neuronal activation, and experimental evidence supporting neuronal inhibition remains elusive. In the current study, we conducted the first inhibitory sonogenetic neuromodulation syste

Inhibition of cGAS in Paraventricular Nucleus Attenuates Hypertensive Heart Injury Via Regulating Microglial Autophagy.

Neuroinflammation in the cardiovascular center plays a critical role in the progression of hypertensive heart disease. And microglial autophagy is involved in the regulation of neuroinflammation. Cyclic GMP-AMP synthase (cGAS), a cytosolic DNA sensor, senses mitochondrial DNA (mtDNA) and regulates autophagy. The detailed mechanisms of central cGAS affects neuroinflammatory response in hypertensive heart disease via regulating autophagy remain unknown. Angiotensin II (Ang II, 1.5 mg·kg-1·12 h-1, 2 weeks) was intraperitoneally injected to induce hypertension in mice. The cGAS-STING pathway was a

Shifts in hormonal stress reactivity during adolescence are not associated with changes in glucocorticoid receptor levels in the brain and pituitary of male rats.

Preadolescent animals display protracted hormonal stress responses mediated by the hypothalamic-pituitary-adrenal (HPA) axis compared to adults. Though the mechanisms that underlie this shift in stress reactivity are unknown, reduced glucocorticoid-dependent negative feedback on the HPA axis has been posited to contribute to this differential responsiveness. As the glucocorticoid receptors (GRs) are integral to this feedback response, we hypothesize that prior to puberty there will be fewer GRs in the neural-pituitary network that mediate negative feedback. To test this hypothesis we measured

Deficiency of antinociception and excessive grooming induced by acute immobilization stress in Per1 mutant mice.

Acute stressors induce changes in numerous behavioral parameters through activation of the hypothalamic-pituitary-adrenal (HPA) axis. Several important hormones in paraventricular nucleus of the hypothalamus (PVN) play the roles in these stress-induced reactions. Corticotropin-releasing hormone (CRH), arginine-vasopressin (AVP) and corticosterone are considered as molecular markers for stress-induced grooming behavior. Oxytocin in PVN is an essential modulator for stress-induced antinociception. The clock gene, Per1, has been identified as an effecter response to the acute stresses, but its fu

Urocortin I inhibits the effects of ghrelin and neuropeptide Y on feeding and energy substrate utilization.

The corticotropin releasing hormone-related ligand, urocortin-I (UcnI), suppresses food intake when injected into multiple hypothalamic and extrahypothalamic areas. UcnI also alters energy substrate utilization, specifically via enhanced fat oxidation as reflected in reductions in respiratory quotient (RQ). In the present study we compared the feeding and metabolic effects of ghrelin and NPY following pretreatment with UcnI. Direct PVN injections of NPY (50 pmol) and ghrelin (50 pmol) were orexigenic while UcnI (10-40 pmol) reliably suppressed food intake. Both ghrelin and NPY increased RQ, in

Satietin: Fos mapping of putative brain sites of action.

A purified extract of a blood-borne satiety factor, called satietin, was injected into the cerebral ventricles of rats that were either fed ad libitum or were food deprived. The animals were killed 2 h after injection and their brains subsequently sectioned and stained for Fos-like immunoreactivity (Fos-IR) to determine the putative sites of action for satietin in the brain. Fos-IR was induced in only a few locations, the most prominent sites being the bed nucleus of the stria terminalis, the central nucleus of the amygdala, and the parvocellular division of the paraventricular nucleus of the

[Morphometric and histologic studies of the significance of the paraventricular nucleus for the thyroid gland axis after ganglionectomy and cold exposure with special consideration of the pineal gland].

To analyse the role of the peripheral sympathetic fibres for the nucleus paraventricularis, the nuclear size and the neurosecretory material were studied 30 d after bilateral cervical ganglionectomy (GX). In order to examine the influence of GX at normal temperature (24 degrees C) and exposure to cold (10 degrees C), rats were subjected to a 72 h exposure to cold before killing. Additionally thyroxine (T4), serum thyrotropin (TSH), and pituitary TSH were investigated. 4 times a day (light-dark cycle: 14L:10D, light on 05h00), the rats were sacrificed: middle light, middle darkness, 1 h after "

Central and peripheral distribution of corticotropin-releasing factor.

Corticotropin-releasing factor (CRF) is widely distributed in the central nervous system and is associated with nuclei and pathways that control the release of adrenocorticotropin and beta-endorphin from the pituitary, and others that mediate central and peripheral autonomic responses to stress. Major concentrations of CRF immunoreactive neurons have been described in the hypothalamus, parts of the limbic system, several nuclei of the basal forebrain and brain stem, and the cerebral cortex. Recent data on the distribution of CRF immunoreactive perikarya and fibers in the preoptic-septal area,

From Cellular Stress to Systemic Adaptation: The Circadian Clock and Stress Response at Cellular and Systemic Levels.

The stress response is essential for cellular and organismal survival as it acts as a protective and adaptive mechanism to maintain homeostasis. At the organismal level, physical stressors induce responses in mammals that are mediated primarily by the hypothalamic-pituitary-adrenal (HPA) axis, which regulates glucocorticoid secretion. The HPA axis functions as a circadian-regulated, multi-oscillator system, in which the paraventricular nucleus, the pituitary, and the adrenal gland exhibit intrinsic rhythmicity while remaining coordinated by the output from the suprachiasmatic nucleus. Astringent flavanol fires the locus-noradrenergic system, regulating neurobehavior and autonomic nerves.

Astringency is a characteristic exhibited by only a limited number of polyphenolic compounds which show high electrochemical activity and are susceptible to oxidation at neutral pH conditions like the oral cavity and small intestine. Large-scale intervention studies have demonstrated that the astringent flavanol (FLs) can restore hippocampal-dependent memory. However, due to the low bioavailability of FLs, the mechanism of action remains unclear. In this study, we aimed to elucidate the mechanism by which FLs acts on the nervous system through the gastrointestinal tract. Following a single gav

Zinc deficiency enhances salt preference through altered peripheral and central taste processing mechanisms.

OBJECTIVES: Zinc deficiency alters salt preference, however, the underlying mechanisms remain unclear. We hypothesized that altered salt preference, during zinc deficiency, is associated with brainstem and hypothalamic nuclei modified activity which is involved in the gustatory processing and fluid balance regulation. METHODS: This study elucidated the abnormal intake of high-concentrated sodium chloride solution caused by zinc deficiency in taste reception, taste nerve, and brain. Initially, we performed taste behavioral tests including the chorda tympani (CT) nerve transection (CTx) and the

Sex-Specific Effects of Stress on Respiratory Control: Plasticity, Adaptation, and Dysfunction.

As our understanding of respiratory control evolves, we appreciate how the basic neurobiological principles of plasticity discovered in other systems shape the development and function of the respiratory control system. While breathing is a robust homeostatic function, there is growing evidence that stress disrupts respiratory control in ways that predispose to disease. Neonatal stress (in the form of maternal separation) affects "classical" respiratory control structures such as the peripheral O2 sensors (carotid bodies) and the medulla (e.g., nucleus of the solitary tract). Furthermore, earl

Cholecystokinin system is involved in the anorexigenic effect of peripherally applied palmitoylated prolactin-releasing peptide in fasted mice.

Prolactin-releasing peptide (PrRP) has been proposed to mediate the central satiating effects of cholecystokinin (CCK) through the vagal CCK1 receptor. PrRP acts as an endogenous ligand of G protein-coupled receptor 10 (GPR10), which is expressed at the highest levels in brain areas related to food intake regulation, e.g., the paraventricular hypothalamic nucleus (PVN) and nucleus of the solitary tract (NTS). The NTS and PVN are also significantly activated after peripheral CCK administration. The aim of this study was to determine whether the endogenous PrRP neuronal system in the brain is in…

Selective stimulation of central GABAAα2,3,5 receptors increases intake and motivation to consume sucrose solution in rats.

Benzodiazepines are one of the most commonly prescribed anxiolytic drugs in America, and between 2006 and 2015 prescription rates increased by an estimated 27.1%. Weight gain is a common side effect of these drugs and it may result from increased feeding caused by drug-enhanced food palatability. We investigated the role of specific GABAA receptor subtypes involved with benzodiazepine-induced food consumption through third ventricle injections of L-838,417, a partial agonist of GABAA α2, α3, and α5 subunits, and a full antagonist of the α1 receptor subunit.
