Rapid and Robust Generation of Human Cortical Interneurons from Induced Neural Stem Cells.

Current protocols for generating cortical interneurons from human pluripotent stem cells are hindered by slow differentiation kinetics and poor reproducibility across cell lines. Here, we present a defined small-molecule-based strategy that efficiently directs human-induced neural stem cells (hiNSCs) toward cortical GABAergic interneurons within 14-18 days, which is substantially faster than conventional methods. Short-term dual-SMAD and WNT inhibition rapidly commits hiNSCs to an interneuron progenitor fate, reaching transcriptional states equivalent to those obtained with prolonged protocols

The Amygdalostriatal Transition Area Exhibits Lateral Amygdala-Like Spiking Activity and Tone-Shock Pairing-Induced Plasticity.

During Pavlovian fear conditioning, presentation of a conditioned stimulus, such as a tone, together with an unconditioned stimulus, such as an electrical shock, excites neurons in the lateral amygdala (LA). Prevailing models propose that sensory stimulus-evoked activity in the LA is subsequently propagated to its downstream nuclei: the basal amygdala (BA) and central amygdala (CeA). To test this assumption, we performed in vivo electrophysiological recordings in awake, head-fixed male and female mice. We found that tone presentation did not elicit a significant increase in firing in BA or CeA

Stress and medial prefrontal cortex: Sex-dependent effects in rodent models.

Chronic stress significantly alters rodent medial prefrontal cortical (mPFC) structure and function, contributing to cognitive and affective dysfunction associated with stress-related psychiatric disorders. Although the effects of stress on the male mPFC have been extensively characterized, growing evidence demonstrates that females exhibit distinct behavioral, structural, and molecular responses to chronic stress. This review synthesizes current findings regarding sex differences in stress-induced plasticity within the adult mPFC, discussing the behavioral outcomes, activation, and neural pla

Arid1b haploinsufficiency in excitatory neurons alters neocortical circuits but not social behavior or seizure phenotypes.

Arid1b is a high confidence risk gene for autism spectrum disorder that encodes a subunit of a chromatin remodeling complex initially expressed in neuronal progenitors. Haploinsufficiency causes a broad range of social, behavioral, and intellectual disability phenotypes, including Coffin-Siris syndrome. Recent work suggests pathology is due to deficits in proliferation, survival, and synaptic development of cortical neurons. Here, we used transgenic mice to investigate how Arid1b dysfunction in cortical excitatory neurons impacts their intrinsic membrane properties, synaptic connectivity and p

GABAergic interneurons in local circuits of primate prefrontal cortex.

Cortical GABAergic interneurons exhibit remarkable diversity of morphological, electrophysiological and molecular features, which underlies their specialized roles in circuit functions. Molecular characteristics have proven to be a reasonable starting point in the past for classification, grouping them into parvalbumin (PV), somatostatin (SST), vasoactive intestinal polypeptide (VIP) and "others" (e.g. LAMP5). While Patch-seq studies provide valuable insight into human cortical interneurons, similar results from non-human primates are lagging behind. This is problematic because the studying of

Pairing extinction training with vagus nerve stimulation reduces drug-seeking by altering activity in afferents to the medial prefrontal cortex.

INTRODUCTION: Relapse triggered by drug-associated cues remains a major challenge in treating substance use disorders, as extinction learning is often weak and context dependent. Vagus nerve stimulation (VNS) enhances learning-related plasticity and, when paired with extinction training, reduces cue-induced reinstatement of cocaine seeking; however, the underlying circuit mechanisms remain unclear. METHODS: We examined how VNS paired with extinction reshapes medial prefrontal cortex (mPFC) networks that regulate drug seeking, focusing on afferent inputs from the anterior (aPVT) and posterior

Circuit dynamics of binocular conflict in mouse primary visual cortex.

Binocular vision requires that the brain integrate input from both eyes to form a unified percept. Small interocular differences support depth perception (stereopsis), while larger disparities can cause diplopia or binocular rivalry. The neural mechanisms by which early visual circuits process concordant versus conflicting binocular signals remain incompletely understood. Here, we used visually evoked potentials (VEPs), unit recordings, and 2-photon calcium imaging in the binocular region of mouse primary visual cortex (bV1) to examine how distinct forms of binocular disparity engage local cir

Focal ischemic stroke impairs REM sleep and inhibits c-Fos expression of SST neuron in EP of male rats.

PURPOSE: The basal ganglia contribute to sleep-wake regulation and are vulnerable to ischemic stroke. Specifically, the striatum, as an upstream input region, appears critically involved, since rapid eye movement (REM) sleep disruption occurs in both extensive (cortex and striatum) and focal striatal infarcts. Nonetheless, the post-stroke dynamics of REM sleep disturbances and their neural targets remain unclear, largely due to the technical limitations in animal models and complexity of stroke outcomes. METHODS: We modified the transient middle cerebral artery occlusion model in rats to indu

A Novel Mouse Model for Developmental and Epileptic Encephalopathy by Purkinje Cell-Specific Deletion of Scn1b.

Loss of function variants of SCN1B are associated with a range of developmental and epileptic encephalopathies (DEEs), including Dravet syndrome. These DEEs feature a wide range of severe neurological disabilities, including changes to social, motor, mood, sleep, and cognitive function which are notoriously difficult to treat, and high rates of early mortality. While the symptomology of SCN1B-associated DEEs indicates broad changes in neural function, most research has focused on epilepsy-related brain structures and function. Mechanistic studies of SCN1B/Scn1b have delineated diverse roles in

Disruption of theta-timescale spiking impairs learning but spares hippocampal replay.

The ability to rapidly learn and retrieve salient information about new environments is critical for survival. In mammals, the hippocampus plays a crucial role in that learning. Specialized features of hippocampal population coding, including network-level theta oscillatory activity, location-specific firing of principal cells, and reactivation of experience during immobility (replay), have been implicated in rapid storage and retrieval of spatial information. Disruptions of theta and replay jointly, or replay alone, are sufficient to impair learning; however, the specific contribution of thet

Microglia-mediated Perineuronal nets loss contributes to social memory deficit in male mice after repeated neonatal sevoflurane exposure.

BACKGROUND: Repeated early-life exposure to general anesthetics might affect social behavior. Perineuronal nets (PNNs), which enwrap around parvalbumin (PV) interneurons and support their function, are crucial for social memory. Given that microglia contribute to PNN remodeling and are responsive to anesthetic exposure, we hypothesized that repeated neonatal sevoflurane exposure impairs social memory via microglia-mediated PNN degradation. METHODS: Mice were exposed to 2.5 % sevoflurane for 2 h daily during postnatal days 7-9. At postnatal day 28, we evaluated social behavior, PNN integrity,

Human milk oligosaccharides improve white matter and interneuron development in a double-hit rat model for preterm brain injury.

Mother's own milk is the preferred source of nutrition for preterm infants due to its beneficial compounds, including human milk oligosaccharides (HMOs). HMOs support microbiota and immune development, but their effect on the preterm brain remains unstudied. Here, we examined the therapeutic potential of HMOs and short-chain galacto- and long-chain fructo-oligosaccharides (scGOS/lcFOS) in a preclinical model for encephalopathy of prematurity. Pregnant Wistar rats were injected with 10 μg/kg lipopolysaccharides at embryonic day 20, and pups were exposed to hypoxia (8% O2, 140 min) at postnatal

Interneuron loss and microglia activation by transcriptome analyses in the basal ganglia of Tourette syndrome.

Tourette syndrome (TS) is a disorder of high-order integration of sensory, motor, and cognitive functions afflicting as many as 1 in 150 children and characterized by motor hyperactivity and tics. Despite high familial recurrence rates, a few risk genes and no biomarkers have emerged as causative or predisposing factors. The syndrome is believed to originate in basal ganglia, where patterns of motor programs are encoded. Postmortem immunocytochemical analyses of brains with severe TS revealed decreases in cholinergic, fast-spiking parvalbumin, and somatostatin interneurons within the striatum

1-Methyltryptophan treatment ameliorates high-fat diet-induced depression in mice through reversing changes in perineuronal nets.

Depression and obesity are prevalent disorders with significant public health implications. In this study, we used a high-fat diet (HFD)-induced obese mouse model to investigate the mechanism underlying HFD-induced depression-like behaviors. HFD-induced obese mice exhibited depression-like behaviors and a reduction in hippocampus volume, which were reversed by treatment with an indoleamine 2,3-dioxygenase (IDO) inhibitor 1-methyltryptophan (1-MT). Interestingly, no changes in IDO levels were observed post-1-MT treatment, suggesting that other mechanisms may be involved in the anti-depressive e

Multiplexed volumetric CLEM enabled by antibody derivatives provides new insights into the cytology of the mouse cerebellar cortex.

Mapping neuronal networks that underlie behavior has become a central focus in neuroscience. While serial section electron microscopy (ssEM) can reveal the fine structure of neuronal networks (connectomics), it does not provide the molecular information that helps identify cell types or their functional properties. Volumetric correlated light and electron microscopy (vCLEM) combines ssEM and volumetric fluorescence microscopy to incorporate molecular labeling into ssEM datasets. We developed an approach that uses small fluorescent single-chain variable fragment (scFv) immuno-probes to perform

Integrated multimodal cell atlas of Alzheimer's disease.

Alzheimer's disease (AD) is the most common cause of dementia in older adults. Neuropathological and imaging studies have demonstrated a progressive and stereotyped accumulation of protein aggregates, but the underlying molecular and cellular mechanisms driving AD progression and vulnerable cell populations affected by disease remain coarsely understood. The current study harnesses single cell and spatial genomics tools and knowledge from the BRAIN Initiative Cell Census Network to understand the impact of disease progression on middle temporal gyrus cell types. We used image-based quantitativ

Nuclei and tracts in the epithalamus of crocodiles.

The epithalamus, an area of the dorsal diencephalon found in all vertebrates, consists of the habenula, the subhabenular nuclei, and associated tracts. The habenula is itself divisible into two parts-a medial and a lateral nucleus differing in their inputs, outputs, and cellular morphology. The medial component is related to the limbic system and serotonergic raphe, while the lateral nucleus is more interconnected with the basal ganglia and midbrain dopamine systems. These findings, which come from experiments mainly done on mammals, serve as a basis for comparison with other vertebrates. Howe

Distinct histological alterations of cortical interneuron types in mouse models of Huntington's disease.

Huntington's disease (HD) is a debilitating hereditary motor disorder caused by an expansion of the CAG triplet repeat in the Huntingtin gene. HD causes neurodegeneration particularly in the basal ganglia and neocortex. In the cortex, glutamatergic pyramidal neurons are known to be severely affected by the disease, but the involvement of GABAergic interneurons remains unclear. Here, we use a combination of immunostaining and genetic tracing to investigate histological changes in three major cortical interneuron types - parvalbumin (PV), somatostatin (SST), and vasoactive intestinal peptide (VI

Propranolol Administration Modulates Neural Activity in the Hippocampal Hilus During Fear Retrieval.

Altered fear learning is a strong behavioral component of anxiety disorders such as post-traumatic stress disorder (PTSD). Recent efforts have attempted to combine exposure therapies with drugs that target fear memory retrieval and memory reconsolidation, in order to improve treatment efficacy. The noradrenergic (NA) signaling system is of particular interest, due to its role in regulating the stress response and its involvement in fear and learning processes. Importantly, propranolol (P), a non-selective β-adrenergic antagonist, has shown the potential in decreasing exaggerated fear in both h

Stimulus-Selective Response Plasticity in Primary Visual Cortex: Progress and Puzzles.

Stimulus-selective response plasticity (SRP) is a robust and lasting modification of primary visual cortex (V1) that occurs in response to exposure to novel visual stimuli. It is readily observed as a pronounced increase in the magnitude of visual evoked potentials (VEPs) recorded in response to phase-reversing grating stimuli in neocortical layer 4. The expression of SRP at the individual neuron level is equally robust, but the qualities vary depending on the neuronal type and how activity is measured. This form of plasticity is highly selective for stimulus features such as stimulus orientat
