Persistent human parvovirus B19 infection in children under maintenance chemotherapy for acute lymphocytic leukemia.

OBJECTIVE: To report on B19 infection management and chemotherapy schedule consequences in five children treated for acute lymphocytic leukemia (ALL). PATIENTS AND METHODS: Between May 2001 and February 2002, five patients between 4 and 12 years of age, receiving maintenance chemotherapy for ALL, presented with symptoms suggesting B19 infection (pallor, fatigue, petechiae and pancytopenia in four patients; generalized rash in two patients; acute hepatitis in one patient). Qualitative polymerase chain reaction (PCR) on peripheral blood was used for diagnosis and follow-up of infection; quantit

Parvovirus B19 infection reminiscent of myelodysplastic syndrome in three children with chronic hemolytic anemia.

The authors have seen transient pancytopenia with erythroid hypoplasia and striking trilineage myelodysplasia reminiscent of true myelodysplastic syndrome (MDS) in 3 children, 1 with thalassemia intermedia and the other 2 with previously undiagnosed hereditary spherocytosis. In these 3 children transient pancytopenia and myelodysplasia coincided with serological evidence of acute parvovirus-B19 (PV-B19) infection, strongly suggesting their relevance. It is of interest that these 3 cases were encountered within a period of 6 months. This might be an incidental event, but it might also be conclu…

Parvovirus B19-induced anemia as the presenting manifestation of X-linked hyper-IgM syndrome.

Parvovirus B19 (B19) can cause chronic anemia due to persistent infection in immunocompromised hosts who cannot produce neutralizing antibody necessary for clearing B19. Three patients with X-linked hyper-IgM syndrome (XHIM), who were all asymptomatic until they developed B19-induced chronic anemia at the ages of 8, 14, and 17 years, respectively, were found to have mutations of the CD40L gene, including a missense mutation (T254M), a nonsense mutation resulting in a new initiation codon and loss of the intracellular domain (R11X), and a splice site mutation (nt 309+2t-->a). Antibody responses…

[Aplastic crisis due to human parvovirus B19 infection in glucose-6-phosphate dehydrogenase deficiency].

Human parvovirus B19 is known to cause aplastic crisis in patients with hemolytic anemias due to cytotoxic effect of the infection to erythroid progenitor cells. We report here the first case of aplastic crisis by B19 in a patient with glucose-6-phosphate dehydrogenase deficiency. A five-year-old boy was admitted to the hospital because of severe anemia, fever and jaundice. Four weeks after admission, he developed erythema infectiosum. B19 infection was confirmed using countercurrent immunoelectrophoresis, Southern blotting and hybridization method, and radioimmunoassay for B19 specific IgM. B…

Electron microscopic identification of parvovirus virions in erythroid-line cells in fatal hydrops fetalis.

Intrauterine infection with parvovirus B19 may lead to fatal hydrops fetalis. Intranuclear particles, consistent with parvovirus virions, within erythroid cells were readily identified on transmission electron microscopy of formalin-preserved material obtained at necropsy from a neonate and two fetuses in whom clinical and light microscopic criteria for parvovirus B19 infection were met. No such particles were seen in similar material from a neonate and two fetuses with erythroblastosis fetalis due to alpha-thalassemia, maternofetal Rh incompatibility, and an erythrocyte membrane protein defec…

Experimental infection of 35, 50 and 60 day old pig foetuses with porcine parvovirus.

Foetuses of six seronegative gilts, two of which each respectively 35, 50 and 60 days pregnant, were inoculated intrauterinely with porcine parvovirus (PPV) and examined 7 and 11 days after inoculation. HI antibody was not detected in any of the foetuses although all but one gilt developed low levels of antibody. All but one of the foetuses inoculated with PPV died in utero prior to examination at 11 days after inoculation. Infection also spread to non-inoculated litter mates. Histological changes were mild in the gilts but there was widespread tissue necrosis in infected foetuses, and intranu…

Safety of and serological response to a new canine distemper-parvovirus combination vaccine in domestic ferrets.

BACKGROUND: Pet ferrets are typically vaccinated against distemper using a canine multivalent vaccine despite the lack of relevant safety or efficacy data. Here, we investigate the safety of a novel live canine parvovirus type 2c (CPV-2c) vaccine strain and the efficacy of the live Onderstepoort canine distemper virus (CDV) vaccine strain used in a new bivalent vaccine. METHODS: Following an overdose of the CPV-2c component to 16 ferrets, blood, faeces and various tissues were tested to investigate viral dissemination and shedding. Serum antibody concentrations were also measured. Long-term s…

The effects of leukocyte's RNA-binding proteins on the immune responses of dogs infected with canine parvovirus.

BACKGROUND: Canine parvovirus (CPV) is a highly contagious virus that can lead to severe clinical complications in dogs. RNA-binding proteins (RBPs) play a crucial role in regulating translation and stabilizing RNA. Due to the failure of the immune system to respond effectively in dogs infected with CPV, this study aimed to investigate various immunological markers in CPV-infected dogs and to evaluate the impact of RBPs from these infected dogs on immune responses. Blood samples were collected from both healthy and parvovirus-infected dogs. Parvovirus infection was confirmed using immunochroma…

Insights into growth retardation and dwarfism caused by goose parvovirus in goslings: a transcriptomic profiling study.

Goose parvovirus (GPV) poses a significant threat to the waterfowl industry as it results in a high mortality rate and stunted growth in surviving goslings, leading to significant economic losses. We used 120 goslings and goose embryo fibroblasts inoculated with the GPV SYG61 strain to study the pathogenesis of GPV by pathological and gene expression profile changes. Fourteen days after infection with the GPV SYG61 strain, goslings showed a mortality rate of 63.33%, along with dwarfism, significant weight loss, and severe histopathological lesions in the liver and jejunum. Serum analysis revea…

Transient severe anemia triggered by parvovirus B19 infection in a patient with homozygous RhAG deficiency.

BACKGROUND: Deficiency of Rh antigens affects the red blood cell (RBC) membrane integrity, thus reducing erythrocyte survival, clinically manifesting as chronic hemolytic anemia. Various mutations cause such Rhnull/mod phenotypes. Parvovirus B19 infection causes transient red cell aplasia, leading to potentially severe anemia in patients with chronic hemolytic disorders. STUDY DESIGN AND METHODS: A patient with known, etiologically unclear mild chronic anemia presented with a syncopal event. Clinical and laboratory work-up, including targeted genetic analyses of RBC genes, was performed. RES…

Outbreak of paediatric myocarditis associated with parvovirus B19 infection in Italy, January to October 2024.

Acute myocarditis has risen among paediatric patients in Italy, with 65 clinically suspected cases reported by 12 centres in 2024, 32 linked to parvovirus B19 (B19V) infection. In 11 cases, B19V was not ruled out despite a concurrent European outbreak. Twenty-nine children required intensive care; eight fatalities occurred. While effective for both severe B19V infection and myocarditis, intravenous immunoglobulins were given in only one-third of cases. These findings highlight the need for timely diagnosis, stronger surveillance, and standardised treatment protocols.

Identification of AXL as a co-receptor for human parvovirus B19 infection of human erythroid progenitors.

Parvovirus B19 (B19V) infects human erythroid progenitor cells (EPCs) and causes several hematological disorders and fetal hydrops. Amino acid (aa) 5-68 of minor capsid protein VP1 (VP1u5-68aa) is the minimal receptor binding domain for B19V to enter EPCs. Here, we carried out a genome-wide CRISPR-Cas9 guide RNA screen and identified tyrosine protein kinase receptor UFO (AXL) as a proteinaceous receptor for B19V infection of EPCs. AXL gene silencing in ex vivo expanded EPCs remarkably decreased B19V internalization and replication. Additions of the recombinant AXL extracellular domain or a pol…

High prevalence of parvovirus B19 infection in patients with thyroid nodules: A case-control study.

BACKGROUND: The incidence of thyroid nodules has increased dramatically in recent decades. Although this increase has been attributed to improved imaging modalities, the question arises as to whether other environmental factors, such as infectious agents are influential. METHODS: Adult patients with newly diagnosed papillary thyroid carcinoma, benign thyroid nodules, and healthy euthyroid controls without nodules; were recruited. Various clinical and biochemical parameters including thyroid function tests and serum Parvovirus B19 Ab (IgG) were assessed and compared between groups. RESULTS: I…

Parvovirus Infection and Thrombotic Thrombocytopenic Purpura in an Adult Patient With Sickle Cell Beta-Thalassemia.

Parvovirus infection and thrombotic thrombocytopenic purpura (TTP) are rare manifestations in adults with sickle cell beta-thalassemia. Due to the lack of a clear demarcation between the complications related to sickle cell disease (SCD) and TTP, the diagnosis is often challenging. The treatment requirements for both these entities are divergent and complicated, thus necessitating a careful plan of action during atypical presentations. Here we present a case of a 22-year-old woman during the peripartum period with fever, generalized body aches, and large joint pains that soon evolved into labo…
