COVID-19 is known to impair red blood cell (RBC) function, which may affect oxygen transport and disease progression. However, the metabolic consequences of SARS-CoV-2 infection on RBCs and how these changes relate to disease severity and sex differences, have not been systematically explored. Here, we compared RBC metabolomic profiles from healthy controls and individuals with COVID-19 using nuclear magnetic resonance (NMR) to gain insight into disease mechanisms and potential biomarkers. Multivariate and univariate analyses were performed within the men and women cohorts, and clinical factor

Chlorination is a cornerstone of drinking water disinfection; however, the persistent oxidative stress it imposes may inadvertently select for metabolically resilient microorganisms within distribution systems. In this study, we investigated the universally conserved metabolic mechanisms underlying chlorine tolerance in a representative environmental bacterial isolate recovered from a full-scale drinking water distribution network. By integrating transcriptomic profiling, targeted metabolomics, and constraint-based flux balance analysis (FBA) within a proof-of-concept framework, we identified

Recurrent vulvovaginal candidiasis (RVVC) is a multifactorial condition in which vaginal microbiota dysbiosis plays a key role. This study aimed to characterize the vaginal microbiome of patients with RVVC using metagenomic sequencing. Vaginal scraping samples were collected from 34 women aged 20-47 years and classified into three groups: (1) 14 women with RVVC who had experienced 3-7 episodes of VVC in the previous year; (2) 9 women with severe RVVC, defined as ≥8 episodes in the last year; and (3) 11 healthy women as controls. The results revealed an increased relative abundance of bacteria

To improve hepatoprotective delivery after hepatectomy, a ROS- and pH-responsive silymarin nanoplatform was constructed using phenylboronic acid-grafted chitosan (Sil@PBACS Nps). The nanoparticles displayed a uniform size of 250-300 nm, high encapsulation efficiency, and favorable colloidal stability. Their release behavior was accelerated under acidic and ROS-enriched conditions, supporting lesion-responsive silymarin delivery. Compared with free silymarin, Sil@PBACS Nps (Nps) exhibited stronger antioxidant activity, with DPPH and ABTS scavenging rates of 92.31% and 73.78%, respectively, toge

Cisplatin resistance remains a primary challenge in the clinical management of non-small-cell lung cancer (NSCLC), yet the regulatory targets underlying this resistance remain largely unknown. It is well established that cisplatin kills tumor cells through the induction of DNA damage and the accumulation of reactive oxygen species, which exacerbate DNA damage. Here, we identify the glycolytic metabolic enzyme PGAM1, and specifically its elevated activity in cisplatin-resistant tumors, as a pivotal metabolic driver of this resistance. Y119 phosphorylation, which reflects increased PGAM1 activit

INTRODUCTION: In physoclist fish, that is, in fish with a closed swimbladder, oxygen has been shown to be the main gas in the swimbladder. Gas gland cells, crucial for the filling of the swimbladder, produce and secrete lactic acid. The resulting acidification of the blood releases oxygen from the hemoglobin via the Root effect, generating high oxygen partial pressures required for the diffusion of oxygen into the swimbladder. CO2 in swimbladder gas was believed mainly to result from a wash out of HCO3 - from the plasma due to acidification, but this reaction would diminish blood acidification

While nitrogen fertilizers are widely used in agricultural production, their application incurs significant environmental and energetic costs. In contrast, some crops are less dependent on these fertilizers because they engage in symbioses with rhizobia, nitrogen-fixing bacteria that provide ammonium to the plant in exchange for carbon. However, the carbon cost associated with nitrogen fixation can negatively impact crop yields. Improving the efficiency of this metabolic process could alleviate this impact on crop productivity. Mathematical models can help us quantitatively explore metabolic b

Furfural is an important biomass-derived furanic platform chemical that has been produced on an industrial scale. Conventional batch processes suffer from low yield, numerous by-products, and high energy consumption. Herein, a continuous countercurrent blowing strategy using superheated water-ethanol mixed steam is proposed to directly convert the acid hydrolysis mixture of corncob (containing pentose and hydrolysis residue) into furfural. By utilizing superheated steam for synchronous dehydration and stripping, a furfural yield of 61.8% with selectivity > 70% is achieved at 180 °C, a relative

Diatoms play a crucial role in aquatic ecosystems, yet the mechanisms underlying their long-term dominance remain poorly understood. This study investigated the relationship between diatom ecological persistence and their phycosphere bacterial communities by comparing the long-term dominant species Cyclotella atomus with the short-term dominant species Ulnaria ulna. 16S rRNA gene sequencing combined with predictive functional profiling revealed that the bacterial community associated with C. atomus was more diverse, stable, and interconnected than that associated with U. ulna. Taxonomic analys

6-Phosphogluconate dehydrogenase (6PGDH) functions in the oxidative phase of the pentose phosphate pathway (oxiPPP), together with glucose-6-phosphate dehydrogenase (G6PDH). These two enzymes play central roles in cellular NADPH generation. In pepper fruit, two genes were identified in the transcriptome, designated Ca6PGDH1 and Ca6PGDH2, which appear to encode a peroxisomal and a plastidial putative isozyme, respectively. During ripening, Ca6PGDH1 exhibited only minor changes in its expression, whereas Ca6PGDH2 was markedly downregulated. The expression patterns of both genes diverged in respo

Metronidazole (MTZ) remains the first-line therapy for trichomoniasis; however, increasing resistance in Trichomonas vaginalis threatens treatment efficacy. Because iron availability enhances MTZ sensitivity, we investigated whether MTZ-induced parasite death involves ferroptosis-related mechanisms and explored alternative pathways contributing to drug resistance. Exposure of metronidazole-sensitive and -resistant isolates to ferroptosis inducers reduced parasite viability, yet neither ferroptosis inhibition nor iron chelation restored survival, and lipid peroxidation was not detected. These f

Metabolic syndrome (MetS), characterized by abdominal obesity, insulin resistance, dyslipidemia, and hypertension, affects a substantial proportion of the global population and increases the risk for cardiovascular disease, diabetes, and metabolic dysfunction-associated steatotic liver disease (MASLD). Despite its prevalence, there are currently no effective pharmacological therapies targeting MetS, highlighting the need to identify novel etiological mechanisms, particularly within the gastrointestinal (GI) tract. Using a mouse model of MetS and healthy lean controls, we assessed the colonic m

Halobenzoquinones (HBQs), emerging disinfection byproducts (DBPs) in drinking water, were prioritized as potential bladder carcinogens. This study aimed to elucidate the interaction between HBQs and DNA, which may represent a critical initial molecular event in the adverse outcome pathway toward carcinogenesis. By combining high- and low-resolution mass spectrometry (MS), we developed a highly selective and sensitive analytical strategy integrating pentose neutral loss and information-dependent acquisition to characterize trace-level HBQ-DNA adducts in complex biological matrices. Superoxide is a toxic byproduct of aerobic cellular respiration. The cellular regulation of bacterial responses to superoxide stress remains incompletely understood. The present work established an Escherichia coli cell model for superoxide stress by deleting superoxide dismutase (SOD) SodA and SodB. Proteomic analysis revealed that SOD deficiency not only induced high expression of the oxidative stress regulator SoxSR but also upregulated the catalase KatE and the organic peroxidases Tpx and BtuE, suggesting that SOD deficiency leads to the subsequent production of multiple reactive oxygen sp

Alzheimer's disease (AD) is increasingly recognized as a disorder marked not only by amyloid-β and tau pathology, but also by profound disturbances in brain energy metabolism that arise early in disease progression. Accumulating evidence indicates that impairments in glucose utilization, insulin signaling, and mitochondrial function precede neurodegeneration and contribute directly to synaptic failure and cognitive decline. This chapter presents a comprehensive overview of Alzheimer's disease through the lens of metabolic dysfunction, highlighting disrupted neuronal bioenergetics as a central

Streptococcus suis (S. suis) is an important zoonotic pathogen that often forms biofilm, leading to persistent clinical infections and exacerbation of antibiotic resistance. There is an urgent need to develop novel antibiofilm strategies. In this study, for the first time, lactate dehydrogenase (LDH) was targeted with the aim of reactivating the metabolic activity of bacteria embedded in biofilm using natural product molecules, thereby disrupting mature biofilm. Through virtual screening based on the TCMSP database, molecular docking, and molecular dynamics simulations, two potential molecules

Enzalutamide is a cornerstone therapy for castration-resistant prostate cancer (CRPC), yet acquired resistance remains a major clinical challenge. Although metabolic enzymes are increasingly recognized as modulators of therapeutic response, their specific roles-particularly their non-enzymatic functions-in sustaining enzalutamide resistance remain incompletely understood. In this study, we performed an in vivo screen using a custom metabolic CRISPR library in enzalutamide-treated xenografts and identified the pentose phosphate pathway enzyme ribulose-5-phosphate 3-epimerase (RPE) as a critical

Endometrial cancer (EC) is the most common gynecological malignancy in high-income countries. Extracellular vesicles (EVs) are key mediators of intercellular communication and metabolic reprogramming, but their molecular cargo in EC remains poorly characterized. EVs were isolated from four EC cell lines representing Type I and Type II subtypes (AN3CA, ISHIKAWA, HEC1A, and KLE). Untargeted metabolomics was performed by HILIC-LC-MS/MS, proteomics by data-independent acquisition (DIA) mass spectrometry, and multi-omics integration using MetaboAnalyst and OmicsNet. Metabolomic profiling identified
