Biomolecular condensates in spatiotemporal regulation of the bacterial cell division cycle.

Bacteria lack membrane-bound organelles, yet they achieve chromosome replication, segregation, and cell division with remarkable spatial and temporal precision. Recent work shows that biomolecular condensates formed by liquid-liquid phase separation play a central role in this organization. In the crowded bacterial cytoplasm, multivalent proteins and nucleic acids form dynamic condensates that concentrate factors, tune reaction kinetics, and create distinct microenvironments. Here we focus on phase separation by cell division proteins, including the divisome scaffold FtsZ and regulators operat…

Improved separation of furocoumarins of essential oils by supercritical fluid chromatography.

Separation of furocoumarins has become of a great interest for cosmetic industry and human health, since the recent directive of the European Union. Furocoumarins are a class of compounds presenting varied substituents linked mainly in two positions to an identical skeleton made by a furan ring bonded to a coumarin nucleus (Psoralen). The substituents are mainly methoxy, or alkyl chains, which can contain double bonds, hydroxyl or epoxy groups. Due to the variety of compounds, and their subtle structure differences, their separation requires high-performance methods. Multi-gradient high-perfor

A novel pillar[3]trianglimine macrocycle with a deep cavity used as a chiral selector to prepare a chiral stationary phase by thiol-ene click reaction for enantioseparation in high-performance liquid chromatography.

A chiral pillar[3]trianglimine (C60 H72 N6 O6 ) with a deep cavity has been developed as a chiral selector and bonded to thiolated silica by thiol-ene click reaction to fabricate a novel chiral stationary phase for enantioseparation in high-performance liquid chromatography. The enantioseparation performance of the fabricated chiral stationary phase has been evaluated by separating various racemic compounds, including alcohols, esters, amines, ketones, amino acids, and epoxides, in both normal-phase and reversed-phase elution modes. In total, 14 and 17 racemates have been effectively separated

Cyano-modified molecular cage silica gel stationary phase: Multi-functional chromatographic performance by high-performance liquid chromatography.

This study successfully prepared different loading levels of cyano-functionalized RCC3 molecular cage silica gel stationary phase (RCC3-CN@SiO2) through aldehyde-amine condensation reaction and subsequent modification strategies. Fourier transform infrared spectroscopy, thermogravimetric analysis, nitrogen adsorption-desorption, and scanning electron microscopy confirmed the successful synthesis of RCC3-CN@SiO2 chromatographic stationary phase. The research demonstrates that due to hydrophobic/hydrophilic interactions, π-π interactions, hydrogen bonding, and size-selective porous structure, th

A unified intermediate-resolution framework for modeling biomolecular condensates at near-cellular complexity.

Biomolecular condensates, formed through phase separation of multivalent biomolecules, are central to cellular organization and function. Here, we present a unified, physics-based intermediate-resolution framework for efficient and quantitative simulation of biomolecular phase separation at near-cellular complexity. The framework integrates the HyRes protein and iConNA nucleic acid models to explicitly describe major driving forces and capture transient local and global structural features underlying phase transitions. With well-balanced hydrogen bonding, electrostatic, cation-π, and hydrophob…

Purification of two triterpenoids from Schisandra chinensis by macroporous resin combined with high-speed counter-current chromatography.

A method for preparative purification of corosolic acid and nigranoic acid from Schisandra chinensis (SC) was established using a combination of macroporous absorption resin column separation and high-speed counter-current chromatography (HSCCC). The crude extracts obtained from SC using 70% ethanol were separated on a macroporous resin column and then eluted with a graded ethanol series. The 70% ethanol fraction was used as the sample for separation of the two triterpenoids by HSCCC. The two-phase solvent system used for HSCCC separation was chloroform-n-butanol-methanol-water (10:0.5:7:4, v/

Structure- and Temperature-Dependent Modulation of the Partitioning of Medium-Chain Fatty Acids and Lipid-Related Volatiles by Dissolved Carbon Dioxide.

The release of lipid-related volatile compounds influences the sensory quality and stability of oil-containing foods; however, the physicochemical role of dissolved carbon dioxide (CO2) in their partitioning remains unclear. This study examined the effects of dissolved CO2 on the gas-liquid partitioning of representative lipid-related compounds, including fatty acids with carbon chain lengths ranging from C4 to C14, aldehydes, ketones, alcohols, and acetate esters, using simplified aqueous model systems. Headspace concentrations under CO2 were compared with those under nitrogen (N2) at 30-50 °

Non-conjugated flexible network for the functional design of silica-based stationary phase for mixed-mode liquid chromatography.

The functionalized non-conjugated organic network modified silica microspheres are proposed as the stationary phase of liquid chromatography, which can effectively avoid some defects of organic framework materials in liquid chromatography. Due to the co-existing of pyridine ring, carbonyl group, amide group and triazine ring in the non-conjugated flexible organic network on the silica surface, the developed mixed-mode stationary phase exhibits multiple interactions between the stationary phase and the analytes during the separation process. A variety of nucleoside bases, organic acids, antibio

Control of the coordination status of the open metal sites in metal-organic frameworks for high performance separation of polar compounds.

Metal-organic frameworks (MOFs) with open metal sites have great potential for enhancing adsorption separation of the molecules with different polarities. However, the elution and separation of polar compounds on such MOFs packed columns using nonpolar solvents is difficult due to too strong interaction between polar compounds and the open metal sites. Here, we report the control of the coordination status of the open metal sites in MOFs by adjusting the content of methanol (MeOH) in the mobile phase for fast and high-resolution separation of polar compounds. To this end, high-performance liqu

Transbilayer Coupling of Lipids in Cells Investigated by Imaging Fluorescence Correlation Spectroscopy.

Plasma membranes host numerous receptors, sensors, and ion channels involved in cellular signaling. Phase separation within the plasma membrane has emerged as a key biophysical regulator of signaling reactions in multiple physiological and pathological contexts. There is much evidence that plasma membrane composition supports the coexistence of liquid-ordered (Lo) and liquid-disordered (Ld) phases or domains at physiological conditions. However, this phase/domain separation is nanoscopic and transient in live cells. It has been recently proposed that transbilayer coupling between the inner and…

Separation of barbiturates and phenylthiohydantoin amino acids using the thermally tuned tandem column concept.

There are many more choices of column type than of eluent type for method development in reversed-phase liquid chromatography. It is common to switch between different column types or between the same type from different suppliers to achieve the desired separations. The key difficulty in modulating band spacing by adjusting the column type is that it is a discontinuous, "hit or miss" proposition. The thermally tuned tandem column (T3C) concept effectively solves this problem by connecting two columns in series and independently controlling the two column temperatures. The columns are chosen to

Elucidating the role of TPGS in felodipine/PVPVA amorphous solid dispersions during water uptake: dissipative particle dynamics simulations using quantum chemistry-based interaction parameters.

Amorphous solid dispersions (ASDs) are an effective formulation approach for improving the solubility of poorly water-soluble drugs. Nevertheless, their drug-release performance depends on the phase separation morphology during the dissolution process. Although the addition of surfactants has been experimentally reported to improve the dissolution behavior of ASDs, experimental methods alone are insufficient to resolve the complex and heterogeneous intermolecular interactions between drugs and excipients in multicomponent systems at the molecular level. In this study, we used dissipative parti…

Preparation, characterization, and separation mechanism of a dehydroabietic-acid-based shape-selective chromatographic stationary phase1.

Chromatographic stationary phases with molecular-shape selectivity are advantageous for the separation and analysis of geometric isomers. Herein, dehydroabietic acid is bonded on the surface of silica microspheres via 3-glycidoxypropyltrimethoxysilane to form a monolayer dehydroabietic-acid stationary phase (Si-DOMM) with a racket-shaped structure. Various characterization techniques indicate that Si-DOMM is successfully prepared, and the separation performance of a Si-DOMM column is evaluated. The stationary phase has a low silanol activity and metal contamination and a high hydrophobicity an

Single-step approach for fabrication of vancomycin-bonded silica monolith as chiral stationary phase.

A vancomycin-bonded silica monolithic column for capillary electrochromatography (CEC) was prepared by a single-step in situ sol-gel approach. This sol-gel process incorporates a synthetic sol-gel precursor which contains a macrocyclic antibiotic, vancomycin, to form a porous silica network inside a fused-silica capillary. To avoid degradation of vancomycin during the column fabrication, a mild step was adopted into the sol-gel process. The performance of the vancomycin chiral stationary phase was investigated by CEC in both the reversed-phase mode and the normal-phase mode. The vancomycin chi

Reversed-phase ion-pair partion chromatography of biogenic catecholamines and their alpha-methyl homologues with tributylphosphate as stationary phase.

Ion-pair partition chromatography is applied to the separation of the biogenic catecholamines and their alpha-methyl homologues. A useful selectivity has been obtained using an adduct-forming organic stationary phase (tributylphosphate). The retention of the compounds can be regulated easily by means of the concentration of the counter-ion (the perchlorate ion) in the mobile phase. The selectivity for separation of amines from amino acids can be influenced by changing the pH of the aqueous phase. The phase system shows a good long-term stability and reproducibility with respect to the capacity
