Mice hypomorphic for Pitx3 show robust entrainment of circadian behavioral and metabolic rhythms to scheduled feeding.

Pitx3ak mice lack a functioning retina and develop fewer than 10% of dopamine neurons in the substantia nigra. Del Río-Martín et al. (2019) reported that entrainment of circadian rhythms to daily light-dark (LD) cycles is absent in these mice, and that rhythms of locomotor activity, energy expenditure, and other metabolic variables are disrupted with food available ad libitum and fail to entrain to a daily feeding. The authors propose that retinal innervation of the suprachiasmatic nucleus is required for development of cyclic metabolic homeostasis, but methodological issues limit interpretati

Hypothalamus but not liver retains daily expression of clock genes during hibernation in terai tree frog (Polypedates teraiensis).

Circadian clock(s) allow an organism to be in synchrony with the surrounding environment and perform daily and seasonal physiological processes, including hibernation, migration, and reproduction. To cope with adverse environmental conditions, organisms have evolved various strategies. Insects undergo diapause, while some higher animals either migrate or hibernate/aestivate during unfavorable environmental conditions. Hibernation is an energy conservation strategy used to cope with adverse environmental conditions. Limited knowledge is available on the physiology of hibernation in non-mammalia

Fasting and refeeding induce differential changes in hypothalamic mRNA abundance of appetite-associated factors in 7 day-old Japanese quail, Coturnix japonica.

There is little information regarding effects of fasting on feeding behavior and hypothalamic physiology in young Japanese quail. The aim was thus to measure food intake and hypothalamic mRNA in response to fasting and refeeding. Five d-old quail ate little during the dark cycle. Food intake was greatest during the first 2 h of the light cycle. Six day-old quail fasted for 6 h ate the most during the first 15 min of refeeding. In 7 d-old quail, 3 h of fasting up-regulated hypothalamic neuropeptide Y (NPY), NPY receptor sub-type 2 (NPYR2), agouti-related peptide (AgRP), orexin receptor 2 (ORXR2

Seasonal Rhythms: The Role of Thyrotropin and Thyroid Hormones.

BACKGROUND: Seasonal changes in various physiological events have been reported in humans, including metabolism, immune function, and mood. However, the molecular and endocrine basis of these seasonal changes remains unclear. SUMMARY: Animals that breed seasonally, such as Japanese quail and the Siberian hamster, have sophisticated seasonal mechanisms, and hence they provide excellent opportunities to understand the underlying processes. Functional genomic analysis in quail uncovered the photoperiodic signal transduction pathway, which regulates avian seasonal reproduction: a long-day stimulu…

Host susceptibility is altered by light intensity after exposure to parasites.

Translating research advances to natural systems using experimental laboratory studies is often difficult because of the variability between … dim light (3 lx) on a 12-12 cycle, and no light (24 hr at 0 lx). We hypothesized that stress due to photoperiod disruption (24 hr of darkness) would result in compromised immune function and lead to higher susceptibility to infection. Prevalence of infected snails differed significantly between the light conditions, and higher susceptibility was observed in t …

Phase-shifting effects of light on the circadian rhythms of 5-methoxytryptophol and melatonin in the chick pineal gland.

In the chick pineal gland, 5-methoxytryptophol and melatonin concentrations fluctuate in a rhythmic manner. These rhythms are circadian in nature persisting in constant darkness and have opposite phases. Acute exposure of chicks to white light (30 lux for 5, 10, 20, and 30 min) at night increased the amount of pineal 5-methoxytryptophol and decreased pineal melatonin content. A 6 hr pulse of light (100 lux) applied early in the subjective night (CT12-CT18) caused a delay in the phase of the circadian rhythms of 5-methoxytryptophol and melatonin by 3.7 and 4.5 h, respectively, compared to untre

Dynamic resetting of the human circadian pacemaker by intermittent bright light.

In humans, experimental studies of circadian resetting typically have been limited to lengthy episodes of exposure to continuous bright light. To evaluate the time course of the human endogenous circadian pacemaker's resetting response to brief episodes of intermittent bright light, we studied 16 subjects assigned to one of two intermittent lighting conditions in which the subjects were presented with intermittent episodes of bright-light exposure at 25- or 90-min intervals. The effective duration of bright-light exposure was 31% or 63% compared with a continuous 5-h bright-light stimulus. Exp

Perinatal development of circadian melatonin production in domestic chicks.

In contrast to the situation in mammals, in which circadian melatonin production by the pineal gland does not begin until some time after birth, the development of pineal gland rhythmicity is an embryonic event in the precocial domestic fowl. A distinct melatonin rhythm was found in 19-d-old chick embryos maintained under light:dark (LD) 16:8. No significant variation in melatonin levels was detected in embryos exposed to LD 8:16. The melatonin rhythm in the pineal gland and plasma of chick embryos incubated for 18 d in LD 12:12 persisted for 2 d in constant darkness indicating that melatonin

Simulations of light effects on the human circadian pacemaker: implications for assessment of intrinsic period.

The sensitivity of the human circadian system to light has been the subject of considerable debate. Using computer simulations of a recent quantitative model for the effects of light on the human circadian system, we investigated these effects of light during different experimental protocols. The results of the simulations indicate that the nonuniform distribution over the circadian cycle of exposure to ordinary room light seen in classical free-run studies, in which subjects select their exposure to light and darkness, can result in an observed period of approximately 25 h, even when the intr

Influence of acute melatonin treatment and light on the circadian melatonin rhythm in the Djungarian hamster.

The present study tested the hypothesis that acute melatonin treatment affects the biological clock mechanism controlling the circadian melatonin rhythm. Adult Djungarian hamsters were raised in LD 16:8 (lights-on from 0300 to 1900 hr). Pineal and serum melatonin concentrations were assessed at specific clock times during the subsequent LD cycle (day 1) and afterwards in hamsters that remained in constant darkness (day 2), to assess the effects of treatment on the circadian melatonin rhythm. In untreated controls (n = 76), melatonin increased in the pineal and in circulation within the first h

Continuous darkness and continuous light induce structural changes in the rat thymus.

It is known that neuroendocrine responses to environmental stimuli, such as light, can influence immune responses through the pineal gland. It is also known that periods of constant darkness and constant light cause stimulation and inhibition of melatonin secretion from the pineal gland, successively. In this study, we provide experimental evidence that changes in the rhythm of the photoperiod have considerable effects on thymic structure of the rat. Male albino Wistar rats were divided into 3 groups. Group 1 rats were kept in a dark room, group 2 in a room under a bright artificial light and …

Novel anticonvulsant action of chronic melatonin in gerbils.

Melatonin, a hormone from the pineal gland, was tested for its anticonvulsant effects in male gerbils. Daily administration of melatonin (25 micrograms injection-1, s.c.) for ten weeks reduced the number and severity of seizures (total convulsion score: 7.67 +/- 1.83 in controls vs 2.47 +/- 0.90 in melatonin-injected animals, p < 0.05) associated with the injection of the convulsant, pentylenetetrazol (PTZ, 60 mg kg-1, s.c.). However, neither 12 weeks of short photoperiod exposure (LD 10:14) nor biweekly administration of melatonin pellets altered PTZ-induced convulsions. Overall, melatonin-in…

Histochemical localization of cyclic 3',5'-AMP phosphodiesterase in rat pineal gland.

Female Wistar rats maintained in a light:dark 12:12 photoperiod cycle were used to investigate the histochemical localization of the cAMP phosphodiesterase activity in the pineal gland of rats killed in the light period or in the dark period of the diurnal lighting regimen. Enzyme activity was identified along the plasma membrane of pinealocytes. The distribution of phosphodiesterase activity for cyclic guanosine monophosphate was similar to that found for cyclic adenosine monophosphate, suggesting that the same enzyme hydrolyzes both nucleotides. In the theophylline control, the reaction prod…

The Relationship between Autism Spectrum Disorder and Melatonin during Fetal Development.

The aim of this review is to clarify the interrelationship between melatonin and autism spectrum disorder (ASD) during fetal development. ASD refers to a diverse range of neurodevelopmental disorders characterized by social deficits, impaired communication, and stereotyped or repetitive behaviors. Melatonin, which is secreted by the pineal gland, has well-established neuroprotective and circadian entraining effects. During pregnancy, the hormone crosses the placenta into the fetal circulation and transmits photoperiodic information to the fetus allowing the establishment of normal sleep patter…

Purinergic receptors have no major role in control of the circadian rhythm in rate of thymidine incorporation by cultured chick pineal glands.

We have examined the effects of some analogues of adenosine upon the circadian rhythm in rate of thymidine incorporation by cultured chick pineal glands. Incorporation in the early period of the photoperiod on day 2 of culture was slightly inhibited by the adenosine analogue N-ethylcarboxamido-adenosine, but this effect was not countered by the antagonist 8-phenyl-theophylline. Thymidine incorporation was inhibited when glands were continuously exposed to the adenosine transport inhibitor nitrobenzyl-thioinosine, but ongoing incorporation was not inhibited by addition of this agent. Removal of…

Chronopharmacological actions of the pineal gland.

In all mammalian species studied to date, the pineal gland shows a pronounced circadian rhythm with high activity at night and very low activity in daytime, as reflected in the output of its hormone, melatonin. The pineal is one of the few organs which can synthesize melatonin and, under normal circumstances, is virtually the only source of circulating melatonin. Pineal activity is tightly controlled by the light/dark cycle, in such a manner that melatonin can convey information to the organism on the length of the dark phase of the photoperiod. In seasonal breeding species, the 24 hour patter…

Seasonal effects of pinealectomy on the ovary of the catfish Heteropneustes fossilis (Bloch) under normal photothermal conditions.

Pinealectomy of Heteropneustes fossilis held under normal photoperiod and ambient temperature produced season-dependent effects on ovarian activity. When the fish were pinealectomized in the preparatory phase (early light-responsive period) and maintained until the early prespawning phase, there was an accelerated growth of the ovary, as adjudged from increased gonado-somatic index (GSI), ovarian histology, elevated ovarian total protein and ascorbic acid levels, and an increased level of serum alkali-labile protein phosphorus (vitellogenin). Pinealectomy was ineffective in the prespawning and…

Ethanol ingestive behavior as a function of central neurotransmission.

Uncontrollable alcohol ingestive behavior has been linked to deficits of central neurotransmission. The pineal gland plays an important role … for regional differences between alcohol-preferring and alcohol-nonpreferring mice. We believe that photoperiod-induced alcohol ingestive behavior may involve alterations in both pineal and hypothalamic opioid peptides.
