Dermoscopic pre- and posttreatment evaluation in patients with androgenetic alopecia on platelet-rich plasma-A prospective study.

BACKGROUND: Platelet-rich plasma is rich in growth factors that promote differentiation and growth of dermal papilla cells by various signaling pathways. Dermoscopy is handy, bedside tool that helps to improve diagnostic accuracy in various hair disorders. OBJECTIVES: To evaluate and study the role of dermoscopy in patients with androgenetic alopecia (AGA) pre- and posttreatment with platelet-rich plasma (PRP). METHODS: It is a prospective comparative study. Twenty male patients with AGA, aged 18-45 years with modified Norwood-Hamilton classification grade II-VI were included in this study.

Acetazolamide Inhibits the Level of Tyrosinase and Melanin: An Enzyme Kinetic, In Vitro, In Vivo, and In Silico Studies.

Melanin is the major factor that determines skin color and protects from ultraviolet radiation. In present study we evaluated the anti-melanogenesis effect of acetazolamide (ACZ) using four different approaches: enzyme kinetic, in vitro, in vivo and in silico. ACZ demonstrated significant inhibitory activity (IC50 7.895 ± 0.24 μm) against tyrosinase as compared to the standard drug kojic acid (IC50 16.84 ± 0.64 μm) and kinetic analyses showed that ACZ is a non-competitive inhibitor without cytotoxic effect. In in vitro experiments, A375 human melanoma cells were treated with 20 or 40 μm of ACZ

[Verapamil in conjunction with pressure therapy in the treatment of pathologic scar due burn injury].

BACKGROUND: Keloids and hypertrophic scars are dermal fibro-proliferative disorders unique to humans. Their treatment is a true challenge with multiple options, but not all the time with good results. Unfortunately this problem is not uncommon in patients with history of burn injury. The aim of this article is to evaluate the use of verapamil and pressure garments in patients with hypertrophic or keloid scar caused by burn injury. METHODS: We included patients with a hypertrophic or keloid scar caused by burn injury candidate to treatment with pressure garment. The pathologic scars were evalu

The pineal and regulation of fibrosis: pinealectomy as a model of primary biliary cirrhosis: roles of melatonin and prostaglandins in fibrosis and regulation of T lymphocytes.

Pinealectomy leads to increased formation of fibrous tissue in the abdominal cavity, increased skin pigmentation and elevated cholesterol and alkaline phosphatase levels. It also leads to reduced formation and/or action of prostaglandin (PG) E1 and thromboxane (TX) A2. PGE1 plays an important role in enhancing function of T suppressor lymphocytes which control overactive antibody-producing B lymphocytes. In primary biliary cirrhosis there are increased skin pigmentation, hepatic fibrosis, elevated cholesterol and alkaline phosphatase levels, defective T lymphocytes and hyperactive B lymphocyte

Approach to hypopigmentation disorders in adults.

Acquired hypopigmentation disorders in adults can be classified on the basis of lesion extent, and can generally be divided into disorders with localized, widespread or generalized lesions. Clinical findings, comprising the degree of pigment loss (hypopigmentation and depigmentation) and associated morphological findings (e.g. epidermal changes, infiltration and induration), are used to further distinguish the disorders. Diagnosing the disorders is important because the underlying causes may be treatable and some of the disorders are associated with malignancies. A systemic approach is useful …

Allele variations in the OCA2 gene (pink-eyed-dilution locus) are associated with genetic susceptibility to melanoma.

The occuloalbinism 2 (OCA2) gene, localized at 15q11, encodes a melanosomal transmembrane protein that is involved in the most common form of human occulo-cutaneous albinism, a human genetic disorder characterized by fair pigmentation and susceptibility to skin cancer. We wondered whether allele variations at this locus could influence susceptibility to malignant melanoma (MM). In all, 10 intragenic single-nucleotide polymorphisms (SNPs) were genotyped in 113 patients with melanomas and in 105 Caucasian control subjects with no personal or family history of skin cancer. By comparing allelic di

Paired DNA and RNA sequencing uncovers common and rare variation regulating human retinal gene expression.

Genetic disorders impacting vision affect millions of individuals worldwide, including age-related macular degeneration (common) and inherited retinal disorders (rare). There is an incomplete understanding of the impact of genetic variation on gene expression in the human retina and its role in genetic disorders. Through the generation of whole genome sequencing and bulk RNA-sequencing of neurosensory retina and retinal pigment epithelium from 201 post-mortem eyes, we uncover common and rare genomic variants shaping retinal expression profiles. This includes 1,483,595 significant cis-expressio

Macular holes in inherited retinal dystrophies and vitreoretinopathies.

A full-thickness macular hole is conventionally regarded as a vitreoretinal-interface disorder of later life caused by anomalous perifoveal posterior vitreous detachment, tangential traction, and failure of foveal tissue compliance. This model explains idiopathic macular holes. Secondary macular holes could occur secondary to trauma, retinal detachment and high myopia, but less is known about the nature of macular holes associated with inherited retinal dystrophies and vitreoretinopathies, in which the fovea, retinal pigment epithelium, basement membranes, or vitreous cortex may be genetically

Dermoscopy of genital diseases: a review.

The male and female external genital regions are anatomical areas in which various types of skin disorders may occur. Although most of these conditions can be diagnosed by means of clinical examination and an accurate medical history, in most cases further investigations with time-consuming and/or invasive procedures are needed in order to reach the correct diagnosis. Dermoscopy, as a modern non-invasive tool, is able to better diagnose pigmented and non-pigmented skin tumours along with various inflammatory and infectious skin and appendage disorders. The aim of this paper was to provide a re

The bisretinoids of retinal pigment epithelium.

The retina exhibits an inherent autofluorescence that is imaged ophthalmoscopically as fundus autofluorescence. In clinical settings, fundus autofluorescence examination aids in the diagnosis and follow-up of many retinal disorders. Fundus autofluorescence originates from the complex mixture of bisretinoid fluorophores that are amassed by retinal pigment epithelial (RPE) cells as lipofuscin. Unlike the lipofuscin found in other cell-types, this material does not form as a result of oxidative stress. Rather, the formation is attributable to non-enzymatic reactions of vitamin A aldehyde in photo

Alcaptonuria and ochronotic arthritis.

The rare hereditary metabolic disorder alcaptonuria is characterized by the inability to metabolize homogentisic acid, an intermediary compound in the catabolism of the aromatic amino acids phenylalanine and tyrosine. The essentially complete deficiency of homogentisic acid oxidase causes a striking accumulation of homogentisic acid and a derived melanin-like pigment in the connective tissues; the latter is termed ochronosis. Urinary homogentisic acid is oxidized rapidly and becomes a brown or black pigment if alkali is added. Older alcaptonurics have intensely pigmented (ochronotic) connectiv

Double layer sign in chorioretinal diseases: Clinical significance and implications from multimodal imaging.

The double-layer sign (DLS) is an emerging optical coherence tomography (OCT) biomarker of growing diagnostic and prognostic relevance in retinal and chorioretinal diseases, including age-related macular degeneration, pachychoroid spectrum disorders. Multimodal imaging, particularly OCT angiography (OCTA), has enhanced our ability to characterize DLS and its role in disease progression and treatment response. DLS is identified as a separation between the retinal pigment epithelium (RPE) and Bruch's membrane, with overlapping phenotypic presentations including flat irregular pigment epithelial

Fibrous dysplasia of bone.

Fibrous dysplasia of bone is a disorder of unknown etiology in which skeletal aberrations constitute the cardinal feature. The condition is often monostotic but may be polyostotic. The disorder may be accompanied by extraskeletal manifestations, such as abnormal cutaneous pigmentation and endocrinopathies, most commonly precocious puberty. Surgical therapy is recommended when bony deformities become substantial or when function is threatened.

Pigmented Onychomatricoma: Clinical and Dermoscopic Findings in a Series of 29 Cases.

INTRODUCTION: Pigmented onychomatricoma is a rare benign fibroepithelial tumor of the nail matrix that may clinically simulate subungual melanoma and other pigmented nail disorders. Because of its rarity and overlapping presentation with malignant nail unit tumors, recognition of its characteristic clinicodermoscopic findings is essential to avoid misdiagnosis and unnecessary aggressive surgical procedures. METHODS: We retrospectively reviewed 29 patients with histopathologically confirmed pigmented onychomatricoma treated between 2015 and 2024. Demographic, clinical, and dermoscopic data wer

Human equilibrative nucleoside transporter-3 (hENT3) spectrum disorder mutations impair nucleoside transport, protein localization, and stability.

Accumulating evidence reveals that sole mutations in hENT3 cause a spectrum of human genetic disorders. Among these include H syndrome, characterized by scleroderma, hyperpigmentation, hypertrichosis, hepatomegaly, cardiac abnormalities and musculoskeletal deformities, pigmented hypertrichotic dermatosis with insulin-dependent diabetes syndrome, characterized by autoantibody-negative diabetes mellitus and skin deformities, familial Rosai-Dorfman disease, characterized by short stature, familial histiocytosis and sinus histiocytosis with massive lymphadenopathy (SHML), characterized by severe t

Deficiency in Lyst function leads to accumulation of secreted proteases and reduced retinal adhesion.

Chediak-Higashi syndrome, caused by mutations in the Lysosome Trafficking Regulator (Lyst) gene, is a recessive hypopigmentation disorder characterized by albinism, neuropathies, neurodegeneration, and defective immune responses, with enlargement of lysosomes and lysosome-related organelles. Although recent studies have suggested that Lyst mutations impair the regulation of sizes of lysosome and lysosome-related organelle, the underlying pathogenic mechanism of Chediak-Higashi syndrome is still unclear. Here we show striking evidence that deficiency in LYST protein function leads to accumulati
